Purification and some properties of an extracellular l-amino acid oxidase from Cellulomonas cellulans AM8 isolated from soil

Summary The soil isolate Cellulomonas cellulans AM8 produces an extracellular l-amino acid oxidase (L-AAO) with broad substrate specificity. The strain produced up to 0.35 unit (U)/ml of the extracellular L-AAO in a simple medium containing glycerol and yeast extract. The enzyme was easily purified up to 30 U/mg protein using Phenyl-Sepharose fast flow. The purified enzyme migrated as single band on sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE) with a molecular mass of 55 kDa. On native PAGE the molecular mass was approx. 300 000 kDa, which may be due to aggregation. With the exception of glycine, proline, and threonine, all the amino acids normally constituting proteins were oxidized. The V _max values from 0.7 to 35.2 U/mg for aspartic acid and lysine, respectively, and the K _m values from 0.007 to 7.1 mm for cysteine and valine, respectively, were obtained at 25° C and pH 7.0 in oxygen-saturated solutions. The L-AAO had a pH optimum of 6.5–7.5. It was stable for several months at — 30° C and for some days at 35° C. Ferricyanide served as an electron acceptor with a V _max of 50 U/mg and K _m for 0.3 mm with phenylalanine as the substrate. Correspondence to: R. D. Schmid     

包囊游仆虫包囊形成和解脱过程中纤毛器的分化

包囊游仆虫(Euplotes encysticus)形成包囊时,各类纤毛器中的纤毛杆被部分地或全部地吸收,毛基体被保留下来。休眠包囊中,背纤毛器的定位无明显变化,但原腹面纤毛器中的口围带和波动膜、额腹棘毛和横棘毛,以及左、右尾棘毛都按序陷入在细胞质内深处,并相互汇聚在一起。脱包囊时,纤毛结构在原毛基体上再分化,新纤毛器按口围带、横棘毛、额腹棘毛和左、右尾棘毛的顺序从细胞内显露出来。     

狭蚤属一新种记述(蚤目:多毛蚤科)

我们从陕西省南部大巴山(安康地区平利县千家坪林场,位于东经109°15′,北纬31°50′)采获一批蚤类标本,经鉴定其中有狭蚤属 Stenoponia Jordan et Rothschild,1911新种。根据其模式产地,命名为大巴山狭蚤,兹记述如下。 大巴山狭蚤 Stenoponia dabashanensis新种 鉴别特征 新种烦栉刺数和下唇须仅一节,与上海狭蚤 Stenoponia shanghaiensis Liu et Wu 1960 兰狭蚤 S. coclcstis Jordan et Rothschild, 1911 和短距狭蚤S. formozovi Ioff et Tiflov, 1934接近。但下列特征可以区别:1)第9腹板后臂近端部     

In vivo inhibition of megakaryocyte and platelet production by platelet factor 4 in mice.

The in vivo effect of human platelet factor 4 (PF4) on murine megakaryocytopoiesis and thrombopoiesis was studied. Administration of PF4 induced a dose-dependent decrease in the numbers of megakaryocytes and their progenitor cells (CFU-MK), continuing for 1 week after the injection. However, the size of megakaryocytes and their colonies was not changed following PF4 injection. Platelet levels were significantly decreased at days 3-4. The number of CFU-GM was decreased at days 1-2. White blood cells and hemoglobin were unaffected by PF4. These data indicate that PF4 inhibits megakaryocyte and platelet production in vivo by acting on the early stage of megakaryocyte development.     

中国寄生云杉的顶裂盘菌及其无性阶段

本文报道了寄生在云杉上的中国新记录种顶裂盘菌(Lophophacidium hyperboreumLagerb.);首次发现了这个种的无性型座壳梭孢属(Apostrasseria sp.),证实了融雪前病株针叶上有表生的菌丝和小菌核;查清了它是新疆云杉林中的广布种,引致云杉雪枯病;发现了新分布区,地理分布范围在75°—94°E,37°40′—49°N;发现了新寄主,多土的西伯利亚云杉(Picea obovata Ledeb.)和引进的青海云杉(P.crassifolia Kom.)、川西云杉(P.balfourianaRehd.et Wils.)。     

杭州双睾虫生活史各阶段体壁的透射电镜观察

作者应用透射电镜技术观察了杭州双睾虫(Diplorchis hangzhouensis)生活史各阶段(自由生活的钩毛蚴、蝌蚪鳃上的后期幼虫、沼蛙膀胱内的成虫)体壁的超微结构,讨论了幼虫发育至成虫过程中这些结构演变的意义,并与其它单殖吸虫的体壁进行了比较。
