Inflammatory cytokines in the central nervous system are largely modulated by glial cells and influence neuronal responses to CNS injury. The protein tyrosine phosphatase SHP-1, an intracellular regulator of many cytokine signaling pathways, has been implicated in mediating the activation of glia. There is a direct correlation between abnormally activated microglia and neuron loss within the SHP-1 deficient motheaten (me/me) mouse auditory brainstem after afferent injury. In order to determine whether loss of SHP-1 creates an aberrant cytokine environment driving the abnormal activation of me/me microglia, the expression of interleukin-4 (IL-4), interleukin-10 (IL-10), interleukin-1beta (IL-1beta), tumor necrosis factor-alpha (TNF-alpha), and interferon-gamma (IFN-gamma) was examined by enzyme-linked immunosorbent assay (ELISA). Normal uninjured me/me mice showed lower IL-10 but higher IL-1beta levels compared to wild-type. Following unilateral cochlear ablation, there is decreased expression of IL-4 and IL-10 in me/me brains compared to wild-type, but IL-1beta is significantly increased. These findings indicate that decreases in anti-inflammatory cytokines, in combination with increased expression of the pro-inflammatory cytokine IL-1beta, may initiate a robust inflammatory reaction within the me/me brain contributing to the neuronal degeneration in the deafferented me/me auditory brainstem. SHP-1 may therefore play a role in limiting CNS inflammation following injury and disease.  相似文献

To study chromosomal aberrations that may lead to cancer formation or genetic diseases, the array-based Comparative Genomic Hybridization (aCGH) technique is often used for detecting DNA copy number variants (CNVs). Various methods have been developed for gaining CNVs information based on aCGH data. However, most of these methods make use of the log-intensity ratios in aCGH data without taking advantage of other information such as the DNA probe (e.g., biomarker) positions/distances contained in the data. Motivated by the specific features of aCGH data, we developed a novel method that takes into account the estimation of a change point or locus of the CNV in aCGH data with its associated biomarker position on the chromosome using a compound Poisson process. We used a Bayesian approach to derive the posterior probability for the estimation of the CNV locus. To detect loci of multiple CNVs in the data, a sliding window process combined with our derived Bayesian posterior probability was proposed. To evaluate the performance of the method in the estimation of the CNV locus, we first performed simulation studies. Finally, we applied our approach to real data from aCGH experiments, demonstrating its applicability.  相似文献

FoF1-ATPase activity is regulated by external links on β subunits with different molecular weight. It is inhibited when anti-β subunit antibody, streptavidin and H9 antibody link on the β subunits successively, but is activated when virus was binded. Western blotting indicated that the employed anti-β antibody target was on the non-catalytic site of the β subunit. Furthermore, an ESR study of spin-labeled ATP (SL-ATP) showed that the affinity of ATP to the holoenzyme increases with increasing external links on the β subunits. This simple regulation method may have great potential in the design of rapid, free labeled, sensitive and selective biosensors.  相似文献

Objectives

To characterize a recombinant carbonyl reductase from Saccharomyces cerevisiae (SceCPR1) and explore its use in asymmetric synthesis of (R)-pantolactone [(R)-PL].

Results

The NADPH-dependent SceCPR1 exhibited strict (R)-enantioselectivity and high activity in the asymmetric reduction of ketopantolactone (KPL) to (R)-PL. Escherichia coli, coexpressing SceCPR1 and glucose dehydrogenase from Exiguobacterium sibiricum (EsGDH), was constructed to fulfill efficient NADPH regeneration. During the whole-cell catalyzed asymmetric reduction of KPL, the spontaneous hydrolysis of KPL significantly affected the yield of (R)-PL, which was effectively alleviated by the employment of the substrate constant-feeding strategy. The established whole-cell bioreduction for 6 h afforded 458 mM (R)-PL with the enantiomeric excess value of >99.9% and the yield of 91.6%.

Conclusions

Escherichia coli coexpressing SceCPR1 and EsGDH efficiently catalyzed the asymmetric synthesis of (R)-PL through the substrate constant-feeding strategy.

在浙江省临安市选择典型天然次生灌丛,分别进行封禁和目标树抚育,探讨灌丛恢复为乔木林的可能性.结果表明: 4年后,与未管护(对照)相比,封禁和目标树抚育后群落平均胸径分别提高1.3和2.6倍,平均高度分别提高0.5和1.1倍;目标树抚育林木出现了对照林分没有的4.5~8.5 cm径阶和4.5~8.5 m树高阶,形成了4 m高的新林层;灌木层物种丰富度和多样性指数没有因抚育而下降;封禁管理维持了群落的树种组成,遵循原有的演替方向;目标树抚育显著改变了群落的树种组成,提高了目的树种的重要值,近期有可能恢复成为针阔混交林群落.与封禁相比,目标树抚育在优势林木胸径和高度生长、树种组成改善等方面更能达到预想的目标.在有条件经营的情况下,可以选择目标树抚育模式对天然次生灌丛进行管理,从而达到加快群落恢复演替形成乔木林的目的.  相似文献

测定了棉铃虫Helicoverpa armigera抗氰戊菊酯种群及相对敏感种群不同组织微粒体的甲氧试卤灵-O-脱甲基酶、乙氧试卤灵-O-脱乙基酶、乙氧香豆素-O-脱乙基酶、芳烷基羟基化酶和艾氏剂环氧化酶的活性。结果表明:抗性种群棉铃虫中肠组织的这5种酶活性分别比敏感种群的活性提高了11.29、4.10、2.66、6.30和2.34倍,其脂肪体及体壁的相应酶活性则分别为敏感种群的1.46、6.80、1.36、4.05、1.48倍和12.32、2.2、1.33、0.80和0.51倍。两种群中,棉铃虫不同组织部位的各单加氧酶活性均不同,活性高低顺序在两种群间也不同。总体而言,均是中肠或脂肪体微粒体对不同底物的氧化代谢能力最强。  相似文献
