A Robust Noninvasive Approach to Study Gut Microbiota Structure of Amphibian Tadpoles by Feces

The 16S rDNA amplicon high-throughput sequencing technique provides a robust and inexpensive approach to detect the gut microbiota of amphibians. Since different experimental protocols generate technical biases in drawing the gut microbiota profiles, the integrative analysis of gut microbiota produced by different studies must be performed with circumspection. In this study, we compared the efficacy of two DNA extraction methods(i.e., a phenol-chloroform method and TIANamp Stool DNA Kit) in describing intestinal and fecal bacterial communities of transplanted Asiatic toad(Bufo gargarizans) tadpoles. In terms of the DNA extraction quality(i.e., DNA purity and yield rate) and the consistency in between fecal and intestinal microbiota structures(i.e., α and β diversity indices), the phenol-chloroform method was more robust than this commercial stool kit in profiling gut microbiota of tadpoles with feces.

Replacement of bacteriopheophytin in reaction centers from Rhodobacter sphaeroides RS601 with plant pheophytin

In the presence of acetone and an excess of exogenous plant pheophytins,bacteriopheophytins in the reaction centers from Rhodobacter sphaeroides RS601 were replaced by pheophytins at sites HA and HB,when incubated at 43.5℃ for more than 15 min.The substitution of bacteriopheophytins in the reaction centers was 50% and 71% with incubation of 15 and 60 min,respectively.In the absorption spectra of pheophytin-replaced reaction centers (Phe RCs),bands assigned to the transition moments QX (537 nm) and QY (758 nm) of bacteriopheophytin disappeared,and three distinct bands assigned to the transition moments QX (509/542 nm) and QY (674 nm) of pheophytin appeared instead.Compared to that of the control reaction centers,the photochemical activities of Phe RCs are 78% and 71% of control,with the incubation time of 15 and 60 min.Differences might exist between the redox properties of Phe RC and of native reaction centers,but the substitution is significant,and the new system is available for further studies.     

人肺组织特性区域差异的实验方法初探

对非肺病因死亡的病人在24小时内解剖而得的人肺各叶进行了研究,以了解人肺组织弹性的区域性差异。人肺矩形薄片试件在二维试验机上进行试验。结果表明:在一定肺容量下,右上叶弹性模量为最大,依次为左上叶,右下叶,左下叶和右中叶。从增量模量观点看、人肺呈现相当大的区域差异。     

番木瓜蛋白酶的免疫荧光定位

番木瓜在我国广东、广西、云南等地栽种,它的乳汁富含番木瓜蛋白酶,该酶主要应用于食品工业、酿造业及化妆品等。目前我们已完成精酶生产工艺的研究并投入了批量生产,然而该酶在果实里的分布特征及组织定位尚不清楚。我们通过幼果切片的组织学染色,观察到番木瓜的乳汁管结构,并用间接免疫荧光法初步确定了番木瓜凝乳蛋白酶的存在部位。     

吴茱萸碱对黑色素瘤细胞的耐药逆转作用

黑色素瘤是目前恶性程度最高的肿瘤之一。临床上,常采用维罗非尼（PLX4032）作为晚期患者的治疗药物,但患者很快就出现了耐药。因此,如何克服耐药、提高患者生存率成为急需解决的问题。本文选用中药吴茱萸碱（EVO）与黑色素瘤A375细胞、对PLX4032耐药的A375细胞（A375/R）进行相关研究。采用CCK-8法检测发现,用EVO的细胞非毒性浓度0.5 μmol/L处理A375/R,PLX4032对A375/R细胞的半数抑制浓度（IC50）降低,逆转倍数为3.85。显示EVO能够增强黑色素瘤A375/R细胞对PLX4032的敏感性。后续实验分为对照组、EVO组、PLX组、PLX + EVO组。流式细胞仪检测结果显示,EVO组细胞凋亡率为5.88%,PLX组细胞凋亡率为17.88%,PLX + EVO组细胞凋亡率为30.28%。细胞集落结果证明,PLX4032联合EVO能够抑制A375/R细胞的克隆形成。免疫印迹法结果证明,联合用药能够上调促凋亡蛋白质Bax、胱天蛋白酶3的表达量,下调p-Akt、p-NF-κB-p65及抗凋亡蛋白Bcl-2的蛋白质水平。以上结果均表明,EVO能够有效逆转黑色素瘤A375/R细胞耐药,并且诱导细胞凋亡,抑制细胞增殖。
