Electrophysiologic effects of serotonin on atrioventricular conduction in dogs

In 12 anaesthetized dogs acute effects of serotonin on the atrioventricular conduction system were studied by obtaining intracardiac electrograms. The significant increase in the heart rate and decrease in aortic pressure elicited by an intravenous infusion of 50 mg/kg.min serotonin for 30 minutes were comparable to those observed in previous investigations. Our data indicate a serotonin induced acceleration of the sinus node pacemaker and atrioventricular conduction by direct and indirect mechanisms.     

Dependence of hypoxic changes in macro- and micro-hemodynamics on the activity of adrenoreceptors in the skeletal muscle in hypothermia

Dependence of hypoxic changes of macro- and microcirculation on the activity of adrenergic receptors in the cooled organism was studied on decentralized shank of cat under perfusion with constant blood flow. After cooling of cat (to 30 degrees C) and blockade of alpha-adrenoreceptors hypoxic hypoxia (10% O2 in N2) caused (a) much greater reduction of precapillary resistance of shank, (b) more striking (by 3 times) increase of capillary filtration coefficient and (c) the increase of capillary pressure and postcapillary resistance in contrast to their decrease to hypoxia under hypothermia before alpha-blockade. Beta-adrenoreceptor blockade had no influence on the changes of the resistance and exchange function of skeletal muscle blood vessels evoked by hypoxia under cooling.     

Enzymes of cyclic nucleotide degradation in eye tissues during experimental ocular herpes

Activity of phosphodiesterases disintegrating cAMP and cGMP in the cornea, sclera and ciliary body was investigated in health and in different stages of experimental herpetic keratitis. The problems concerning the role of the cyclase system in the pathogenesis of herpetic keratitis and the possibility of applying some of the drugs to the disease treatment are discussed.     

NMR studies of the conformational change in human N-p21ras produced by replacement of bound GDP with the GTP analog GTP gamma S.

1H-Detected 15N-edited NMR in solution was used to study the conformational differences between the GDP- and GTP gamma S-bound forms of human N-p21ras. The amide protons of 15N-labeled glycine and isoleucine were observed. Resonances were assigned to residues of particular interest, glycines-60 and -75 and isoleucines-21 and -36, by incorporating various 13C-labeled amino acids in addition to [15N]glycine and [15N]iosleucine and by replacing Mg2+ by Co2+. When GTP gamma S replaced GDP in the active site of p21ras, only 5 of the 14 glycine amide resonances show major shifts, indicating that the conformational effects are fairly localized. Responsive glycines-10, -12, -13, and -15 are in the active site. Gly-75, located at the far end of a conformationally-active loop and helix, also responds to substitution of GTP gamma S for GDP, while Gly-77 does not, supporting a role for Gly-75 as a swivel point for the conformational change. The amide proton resonances of isoleucines-36 and -21 and a third unidentified isoleucine also undergo major shifts upon replacement of GDP by GTP gamma S. Thus, the effector-binding loop containing Ile-36 is confirmed to be involved in the conformational change, and the alpha-helix containing Ile-21 is also shown to be affected.     

Comparative analysis of the HBsAg level in human blood preparations

A total of 218 batches of blood preparations produced from different raw materials have been studied by means of enzyme immunoassay kits (Abbott Laboratories, USA). The assays have revealed that the preparations under study are nonstandard with respect to the content and isolation rate of HBsAg, the marker of hepatitis B virus. These data necessitate search for the ways of improving the quality of blood preparations.     

Evaluation of cell proliferation kinetics by the differential staining of the sister chromatids at 2 points of cell fixation

Lymphocyte proliferation in blood cultures from 23 normal individuals was studied. A simple method of evaluation of cell proliferative kinetics with sister chromatid differential staining using two different points of cell harvesting was suggested. It was shown that different composition of tissue culture medium plays a significant role during the cell exit from G0 stage, the mean doubling generation time of cellular population remaining constant.     

Establishment of human tumor strains from cell lines transplanted into athymic mice and rats

The authors describe human cancer strains established from 6 cell lines transplanted to nude mice and rats: cancer of the liver, colon, lung, bladder and Burkitt's lymphoma. In spite of a long history (for several years) of cell line transfers in vitro, on transplantation of tumor cells, nude animals established tumors histologically identical to the primary cancer.     

Phenylalanine hydroxylase of human embryos: its homology with the enzyme from the liver of adults

Phenylalanine hydroxylase (PH) activity was discovered in the liver of 7-12 week old human embryos. Embryonic and adult PHs were identical, as shown by immunoelectrophoresis. Unlike the adult liver PH, the PH content of the extract of cytoplasmic proteins of embryonic liver was reduced but the specific activity was increased more than by one order of magnitude. H (57,000 D) and L (55,000 D) subunits were detected by immunoblotting. The L subunit predominates in the extract of membrane proteins of embryonic liver. Hence, the major part of phenylalanine oxidizing activity in the embryonic liver is related to the enzyme immunochemically identical with the PH of adult liver but differing from it in some structural and functional properties.
