Since the 1997 H5N1 influenza virus outbreak in humans and poultry in Hong Kong, the emergence of closely related viruses in poultry has raised concerns that additional zoonotic transmissions of influenza viruses from poultry to humans may occur. In May 2001, an avian H5N1 influenza A virus was isolated from duck meat that had been imported to South Korea from China. Phylogenetic analysis of the hemagglutinin (HA) gene of A/Duck/Anyang/AVL-1/01 showed that the virus clustered with the H5 Goose/Guandong/1/96 lineage and 1997 Hong Kong human isolates and possessed an HA cleavage site sequence identical to these isolates. Following intravenous or intranasal inoculation, this virus was highly pathogenic and replicated to high titers in chickens. The pathogenesis of DK/Anyang/AVL-1/01 virus in Pekin ducks was further characterized and compared with a recent H5N1 isolate, A/Chicken/Hong Kong/317.5/01, and an H5N1 1997 chicken isolate, A/Chicken/Hong Kong/220/97. Although no clinical signs of disease were observed in H5N1 virus-inoculated ducks, infectious virus could be detected in lung tissue, cloacal, and oropharyngeal swabs. The DK/Anyang/AVL-1/01 virus was unique among the H5N1 isolates in that infectious virus and viral antigen could also be detected in muscle and brain tissue of ducks. The pathogenesis of DK/Anyang/AVL-1/01 virus was characterized in BALB/c mice and compared with the other H5N1 isolates. All viruses replicated in mice, but in contrast to the highly lethal CK/HK/220/97 virus, DK/Anyang/AVL-1/01 and CK/HK/317.5/01 viruses remained localized to the respiratory tract. DK/Anyang/AVL-1/01 virus caused weight loss and resulted in 22 to 33% mortality, whereas CK/HK/317.5/01-infected mice exhibited no morbidity or mortality. The isolation of a highly pathogenic H5N1 influenza virus from poultry indicates that such viruses are still circulating in China and may present a risk for transmission of the virus to humans.  相似文献

Cassava (Manihot esculenta) is an allogamous, vegetatively propagated, Neotropical crop that is also widely grown in tropical Africa and Southeast Asia. To elucidate genetic diversity and differentiation in the crop's primary and secondary centers of diversity, and the forces shaping them, SSR marker variation was assessed at 67 loci in 283 accessions of cassava landraces from Africa (Tanzania and Nigeria) and the Neotropics (Brazil, Colombia, Peru, Venezuela, Guatemala, Mexico and Argentina). Average gene diversity (i.e., genetic diversity) was high in all countries, with an average heterozygosity of 0.5358 ± 0.1184. Although the highest was found in Brazilian and Colombian accessions, genetic diversity in Neotropical and African materials is comparable. Despite the low level of differentiation [Fst(theta) = 0.091 ± 0.005] found among country samples, sufficient genetic distance (1-proportion of shared alleles) existed between individual genotypes to separate African from Neotropical accessions and to reveal a more pronounced substructure in the African landraces. Forces shaping differences in allele frequency at SSR loci and possibly counterbalancing successive founder effects involve probably spontaneous recombination, as assessed by parent-offspring relationships, and farmer-selection for adaptation.Communicated by H.C. Expanding upon a preliminary communication (Nature 417 (2002) 166), we here further develop a "multiple-causes model" of allometry, where the exponent b is the sum of the influences of multiple contributors to control. The relative strength of each contributor, with its own characteristic value of b(i), is determined by c(i), the control contribution or control coefficient. A more realistic equation for the scaling of metabolism with body size thus can be written as BMR=MR(0)Sigmac(i)(M/M(0))(bi), where MR(0) is the "characteristic metabolic rate" of an animal with a "characteristic body mass", M(0). With M(0) of 1 unit mass (usually kg), MR(0) takes the place of the value a, found in the standard scaling equation, b(i) is the scaling exponent of the process i, and c(i) is its control contribution to overall flux, or the control coefficient of the process i. One can think of this as an allometric cascade, with the b exponent for overall energy metabolism being determined by the b(i) and c(i) values for key steps in the complex pathways of energy demand and energy supply. Key intrinsic factors (such as neural and endocrine processes) or ecological extrinsic factors are considered to act through this system in affecting allometric scaling of energy turnover. Applying this model to maximum vs. BMR data for the first time explains the differing scaling behaviour of these two biological states in mammals, both in the absence and presence of intrinsic regulators such as thyroid hormones (for BMR) and catecholamines (for maximum metabolic rate).  相似文献

This work determined Plasmodium falciparum merozoite surface protein-8 (MSP-8) regions specifically binding to membrane surface receptors on human erythrocytes. Five high activity binding peptides (HABPs), whose binding to erythrocytes became saturable and sensitive on being treated with neuraminidase and chymotrypsin were identified from the MSP-8 protein. Those amino acids directly involved in interaction with erythrocytes were also determined for each one of the HABPs. Some of them specifically recognized 28, 46, and 73 kDa erythrocyte membrane proteins. Some HABPs inhibited in vitro P. falciparum merozoite invasion of erythrocytes by up to 98%, suggesting the MSP-8 protein's possible role in the invasion process.  相似文献

AIMS: Screening of antimicrobial activity in 25 plant species from Northern Argentina. METHODS AND RESULTS: Inhibition of microbial growth was measured by a microplate assay with an oxidation-reduction indicator (Alamar Blue). Test organisms were: Escherichia coli, Salmonella typhimurium, Pseudomonas aeruginosa, Klebsiella pneumoniae, Staphylococcus aureus and Enterococcus faecium. Weak inhibitory activities (MIC=0.5 mg dry matter ml(-1)) were found in methanolic extracts of Rivina humilis, Crateva tapia, Funastrum claucum and Schinopsis balansae. Stronger bacteriostatic power was detected in Vassobia breviflora (MIC=0.25 mg ml(-1) against Staphylococcus aureus, and 0.5 mg ml(-1) against Enterococcus faecium). This activity was purified five-fold by extraction with dichloromethane, and it was found equally effective against susceptible or antibiotic-resistant strains of Staph. aureus. In addition, the purified extract was synergistic with gentamicin, and it was bactericidal at 24 h, with a concentration of 0.25 mg ml(-1). CONCLUSION: There is a significant antimicrobial activity in Vassobia breviflora. SIGNIFICANCE AND IMPACT OF THE STUDY: Further studies will be required to disclose the potential importance of these findings.  相似文献

A novel 3alpha,6beta,7beta-triol tropane alkaloid esterified by two benzoyl residues was isolated from the leaves of the endemic cuban species, Erythroxylon alaternifolium. Another novel 3alpha,6alpha,7beta-triol tropane alkaloid esterified by trimethoxycinnamoyl and trimethoxybenzoyl residues was isolated from the leaves and stem bark of a second endemic cuban species, Erythroxylon rotundifolium. Their structures were elucidated as 3alpha,7beta-dibenzoyloxy-6beta-hydroxy-tropane and 3alpha-(3,4,5-trimethoxycinnamoyloxy)-7beta-(3,4,5- trimethoxybenzoyloxy)-6alpha-hydroxy-tropane by spectroscopic methods including 2D-NMR techniques.  相似文献

In 1997, an outbreak of virulent H5N1 avian influenza virus occurred in poultry in Hong Kong (HK) and was linked to a direct transmission to humans. The factors associated with transmission of avian influenza virus to mammals are not fully understood, and the potential risk of other highly virulent avian influenza A viruses infecting and causing disease in mammals is not known. In this study, two avian and one human HK-origin H5N1 virus along with four additional highly pathogenic H5 avian influenza viruses were analyzed for their pathogenicity in 6- to 8-week-old BALB/c mice. Both the avian and human HK H5 influenza virus isolates caused severe disease in mice, characterized by induced hypothermia, clinical signs, rapid weight loss, and 75 to 100% mortality by 6 to 8 days postinfection. Three of the non-HK-origin isolates caused no detectable clinical signs. One isolate, A/tk/England/91 (H5N1), induced measurable disease, and all but one of the animals recovered. Infections resulted in mild to severe lesions in both the upper and lower respiratory tracts. Most consistently, the viruses caused necrosis in respiratory epithelium of the nasal cavity, trachea, bronchi, and bronchioles with accompanying inflammation. The most severe and widespread lesions were observed in the lungs of HK avian influenza virus-infected mice, while no lesions or only mild lesions were evident with A/ck/Scotland/59 (H5N1) and A/ck/Queretaro/95 (H5N2). The A/ck/Italy/97 (H5N2) and the A/tk/England/91 (H5N1) viruses exhibited intermediate pathogenicity, producing mild to moderate respiratory tract lesions. In addition, infection by the different isolates could be further distinguished by the mouse immune response. The non-HK-origin isolates all induced production of increased levels of active transforming growth factor beta following infection, while the HK-origin isolates did not.  相似文献

Structural modification of the peptide backbone via N‐methylation is a powerful tool to modulate the pharmacokinetic profile and biological activity of peptides. Here we describe a rapid and highly efficient microwave(MW)‐assisted Fmoc/tBu solid‐phase method to prepare short chain N‐methyl‐rich peptides, using Rink amide p‐methylbenzhydrylamine (MBHA) resin as solid‐phase support. This method produces peptides in high yield and purity, and reduces the time required for Fmoc‐N‐methyl amino acid coupling. Several specimens of Mytilus galloprovincialis, collected in the Ria of Vigo over a non-consecutive 2 yr period (1993 to 1994 and 1996 to 1997), presented a possible gonadal neoplasm, entailing morphologically abnormal germinal cells distributed throughout the follicle and invading the adjacent storage tissue. In some cases, affected cells were noted in gonoducts and in haemic sinusoids. Prevalence of this anomaly in the samples was 6%, and all affected individuals were found between April and June. During the rest of the year, individuals presented normal gonadal tissue.  相似文献

Aim To determine whether the cover and growth habit of the main forest forming species (Polylepis australis BITT.) in a mountain range with low human population density is mainly affected by anthropogenic activities or by environmental influences. Location Central Argentina. Methods Using GIS and field surveys we established 146 plots of 30 × 30 m located in five river basins differing in human impact. We measured P. australis cover, growth habit of each individual (number of basal ramifications), index of long term human impact (percentage of rock exposed by soil erosion due to livestock and fires), evidence of logging, fire scars, local relief, percentage of rock outcrops and altitude above sea level. We analysed the influence of independent variables on P. australis cover and growth habit (average number of basal ramifications per plot) using correlations and General Linear Models. Results Polylepis australis cover was greater at intermediate altitudes above sea level and in areas with reduced long term human impact. Contrastingly local relief, percentage of rock outcrops and logging in the recent past did not have a major influence on P. australis abundance. Growth habit varied in complex patterns. Individuals with fewer ramifications were found in valley bottoms and more disturbed basins, while more ramifications were found at mid‐ and upper slopes and well preserved basins. In valley bottoms, ramifications decreased with increasing altitude whereas the opposite trend was observed for mid‐slopes. Ramifications were positively related to fires in two river basins and in mid‐ and upper slopes but not in valley bottoms. Fire impact was always less in valley bottoms than at mid‐ and upper slopes. Main conclusions Human impact had a major role on P. australis cover, while growth habit was determined by complex combinations of potentially cumulative natural and anthropogenic factors. Even in sparsely populated mountains, both human impact and their interaction with natural environmental gradients influence plant communities and need to be understood for effective management.  相似文献
