The triglycerides (TGs) stored in the white adipose tissue are mobilized during periods of negative energy balance. To date, there is no in vitro model of adipocytes imitating a long period of negative energy balance in which triglycerides are highly mobilized. Such model would allow studying the mobilization of TGs and lipophilic compounds trapped within the adipose tissue (e.g., pollutants and vitamins). The present study aims at developing a performing long-term in vitro lipolysis in adipocytes, resulting in a significant decrease of TG stores. Lipolysis was induced on differentiated rat adipocytes by a lipolytic medium with or without isoproterenol for 12 h. The condition with isoproterenol was duplicated, once with medium renewal every 3 h and once without medium renewal. Adding isoproterenol efficiently triggered lipolysis in a short time (3 h). However, a single stimulation by isoproterenol, without medium renewal, was not sufficient to reduce the TG content during a longer term (12 h). A reesterification of fatty acids occurred after a few hours of lipolysis, resulting in a novel increase of cellular lipids. Regular medium renewal combined with repeated isoproterenol stimulations led to almost emptied cells after 12 h. However, medium renewal without isoproterenol stimulation for 12 h was as efficient in terms of lipid mobilization. Our study demonstrates that, over a short-term period, isoproterenol is required to exert a significant lipolytic effect on adipocytes. During a long-term period, the presence of isoproterenol is no longer essential. Instead, medium renewal becomes the main factor involved in cell emptying. The efficiency of this protocol was demonstrated by visual tracking of the cells and by monitoring the dynamics of release of a lipophilic compound, PCB-153, from adipocytes during lipolysis. 相似文献

We examined epibenthic and pelagic species of Chaetognatha (Spadellidae and Sagittidae) using immunohistofluorescence and confocal laser scanning microscopy to detect tubulin and cell nuclei in whole-mount preparations and scanning and transmission electron microscopy to visualize the ultrastructural organisation of their ciliary sense organs. All chaetognaths bear three types of ciliary sense organs distributed throughout the body: (1) transversally oriented ciliary fence organs, (2) longitudinally (parallel to the anterior-posterior axis) oriented ciliary tuft organs, and (3) a ciliary loop, the corona ciliata. This study targets the ciliary fence as well as the ciliary tuft organs. Two types of primary receptor cells constitute the ciliary fence and ciliary tuft organs. The first type of receptor cells forms a single cell line along the midline axis of the organs, whereas the second type of receptor cells forms multiple lines of cells at either side of type 1 cells. Each receptor cell extends a single, non-locomotory cilium from its narrow apex collared by slender, non-reinforced microvilli; however, both types of sensory cells considerably differ on ultrastructural level. Type 1 sensory cells have thicker cilia than those protruded by the type 2 sensory cells which are characterized by rootlets consisting of an elongated, amorphous distal as well as a cross-striated proximal portion. A prospective, open trial was conducted to evaluate the nutritional adequacy of a semi-elemental diet in 47 children with
functional gastro-intestinal disorders. Nutritional adequacy was assessed based on growth relative to Euro-growth standards
for body mass index (BMI)-for-age z-scores and evaluations of blood parameters. Twenty-five patients completed the study.
In total, 533 l of “New-Alfare” was consumed during 775 trial-days. The mean intake per infant was 85.8 ± 26.8 kcal/kg/day
or 122.5 ± 38.3 ml/kg/day. Weight and length evolution during the 4 weeks trial were within normal range. The mean BMI-for-age
z-score (P < 0.05) and albumin concentration (P < 0.01) increased significantly after 4 weeks. Plasma threonine concentration decreased significantly (P = 0.01) and the tryptophan concentration increased (P = 0.06). No adverse events related to the study formula were reported. These results show that “New Alfaré” is safe and nutritionally
