Endoplasmic reticulum stress (ERS)-induced unfolded protein response (UPR) and the subsequent cell deaths are essential steps in the pathogenesis of diabetic cardiomyopathy (DCM), a main cause of diabetics’ morbidity and mortalities. The bis(maltolato)oxovanadium(IV) (BMOV), a potent oral vanadium complex with anti-diabetic properties and insulin-mimicking effects, was shown to improve cardiac dysfunctions in diabetic models. Here, we examined the effects of BMOV on UPR pathway protein expression and apoptotic cell deaths in both high glucose-treated cardiac H9C2 cells and in the hearts of diabetic rats. We show that in both the high glucose-treated cardiac cells and in the hearts of streptozotocin (STZ) diabetic rats, there was an overall activation of the UPR signaling, including both apoptotic (e.g., the cascades of PERK/EIf2α/ATF4/CHOP and of IRE1/caspase 12/caspase 3) and pro-survival (GRP78 and XBP1) signaling. A high amount of apoptotic cell deaths was also detected in both diabetic conditions. The administration of BMOV suppressed both the apoptotic and pro-survival UPR signaling and significantly attenuated apoptotic cell deaths in both conditions. The overall suppression of UPR signaling by BMOV suggests that the drug protects diabetic cardiomyopathy by counteracting reactive oxygen species (ROS) and endoplasmic reticulum (ER) stress. Our findings lend support to promote the use of BMOV in the treatment of diabetic heart diseases.  相似文献

Most environmental perturbations have a direct or indirect deleterious impact on photosynthesis, and, in consequence, the overall energy status of the cell. Despite our increased understanding of convergent energy and stress signals, the connections between photosynthesis, energy and stress signals through putative common nodes are still unclear. Here we identified an endoplasmic reticulum (ER)-localized adenine nucleotide transporter1 (ER-ANT1), whose deficiency causes seedling lethality in air but viable under high CO2, exhibiting the typical photorespiratory phenotype. Metabolic analysis suggested that depletion of ER-ANT1 resulted in circadian rhythm disorders in sucrose synthesis and induced sucrose signaling pathways, indicating that the ER is involved in the regulation of vital energy metabolism in plants. In addition, the defect of ER-ANT1 triggers ER stress and activates the unfolded protein response in plant cells, suggesting ER stress and photorespiration are closely linked. These findings provide an important evidence for a key role of ER-localized ER-ANT1 in convergent energy and stress signals in rice. Our findings support the idea that ATP is a central signal involved in the plant response to a variety of stresses.  相似文献

In the past decade improved acoustic hard‐ and software have enabled estimations of abundance and distribution patterns of aquatic organism, including non‐intrusive monitoring of fish migrations and behaviour. In this study, a high frequency acoustic camera (DIDSON‐LR, 1.2 MHz, 0.7 MHz) and a portable split‐beam scientific echo sounder (Simrad EY60, 200 kHz) collected acoustic data on 192 and 157 individuals within 24 hr (19–20 April 2011) in the Mituo reach of the Yangtze River, China. Mean fish length estimated from the acoustic camera data was 18.7 ± 5.6 cm, with an average swimming speed of 0.19 ± 0.13 m s?1. The mean fish target strength (TS) produced by the echo sounder was ?43.8 ± 4.4 dB, which corresponded to 5.7–119.9 cm fish length when converted by three different TS‐length equations. Average swimming speed was 0.11 ± 0.06 m s?1 from the echo sounder. Compared with the actual fish catch by the three layers of drift gill net in the survey area, the target length indicated by DIDSON was more accurate than the EY60 results, which were highly affected by the choice of TS length equations. It was determined that the two devices used synchronously could estimate fish length effectively to investigate their behaviour and distribution.  相似文献
