The effect of calcitonin (CT) on calcium content and enzyme activity in the hepatic mitochondria of intact rats was investigated. A single subcutaneous administration of CT (80 MRC mU/100 g BW) produced a significant increase in the content of calcium, the activity of pyruvate carboxylase, succinate dehydrogenase and ATPase 15 min after the hormone treatment. The significant increases in calcium content and pyruvate carboxylase activity were also observed 30 min after CT administration, while succinate dehydrogenase and ATPase activity began to decrease. A physiological dose of CT (20 MRC mU/100 g BW) caused a marked increase in calcium content and pyruvate carboxylase activity but not succinate dehydrogenase of ATPase-activity. The removal of calcium by 10 mM EGTA washing of the mitochondria produced a remarkable reduction in pyruvate carboxylase activity increased by CT administration. The addition of calcium ion of 2.5 x 10(-2) - 2.5 x 10(1) nmoles Ca2+ per mg mitochondrial protein produced a marked increase in pyruvate carboxylase activity. The present results suggest that calcium taken up by the hepatic mitochondria after CT administration activates pyruvate carboxylase. 相似文献

Reactions catalyzed by NAD-linked malic enzyme from Escherichia coli were investigated. In addition to L-malate oxidative decarboxylase activity (Activity 1) and oxaloacetate decarboxylase activity (Activity 2), the enzyme exhibited oxaloacetate reductase activity (Activity 3) and pyruvate reductase activity (Activity 4). Optimum pH's for Activities 3 and 4 were 4.0 and 5.0, and their specific activities were 1.7 and 0.07, respectively. Upon reaction with N-ethylmaleimide (NEM), Activity 1 decreased following pseudo-first order kinetics. Activity 2 decreased in parallel with Activity 1, while Activities 3 and 4 were about ten-fold enhanced by NEM modification. Modification of one or two sulfhydryl groups per enzyme subunit caused an alteration of the activities. Tartronate, a substrate analog, NAD+, and Mn2+ protected the enzyme against the modification. The Km values for the substrates and coenzymes were not significantly affected by NEM modification. Similarly, other sulfhydryl reagents such as p-hydroxymercuribenzoate (PMB), 5,5'-dithiobis(2-nitrobenzoate) (DTNB), and iodoacetate inhibited the decarboxylase activities and activated the reductase activities to various extents. Modification of the enzyme with PMB or DTNB was reversed by the addition of a sulfhydryl compound such as dithiothreitol or 2-mercaptoethanol. Based on the above results, the mechanism of the alteration of enzyme activities by sulfhydryl group modification is discussed. 相似文献

1. The properties of 3,5-di-tert-butyl-4-hydroxybenzylidenemalononitrile (SF 6847) were studied chemically and spectroscopically. Two molecular species of SF6847 were identified: the undissociated form (SFH; ?363, 10 mM?1) and the dissociated form (SF?; ?454, 35 mM?1). The pKa value of the molecule was determined to be 6.9.2. On the basis of these properties the interactions of SF6847 with liposomes and valinomycin · K+ were studied. The partition constants of SFH (Knp and SF? (K?p) to liposomes were determined separately; Knp was 56 mM?1 and was independent of the pH of the medium, whereas K?p dependend greatly on the pH, being 1.2 mM?1 at pH 7.0 and 2.9 mM?1 at pH 8.0. Using these values, the partition constant of total SF6847 (Kp) was calculated and found to be essentially the same as that calculated from the kinetics of proton uptake. It was concluded that the amount of SF? bound to liposomes is rate limiting for proton uptake.3. The effects of membrane potential on partition constants were studied. The K?p decreased greatly upon generation of a membrane potential negative inside the liposomes but increased upon generation of a membrane potential positive inside the liposomes.4. The interaction of SF6847 with valinomycin in aqueous solution and in liposomes was demonstrated only in the presence of potassium ion. Potassium ion could not be replaced by sodium ion. Evidence was obtained for the formation of the ternary complex valinomycin · K+ · SF? in liposomes and in hexane. It was concluded that SF? became more soluble in the liposomal membranes on formation of this ternary complex. Nine bibenzyls and 10 stilbenes were synthesized as analoguesof batatasin III, a growth inhibitor isolated from dormant yambulbils, and examined for their plant growth-regulating activities.The bioassays used were the elongation of dark-grown intactrice coleoptiles, auxin-induced elongation of excised oat coleoptiles,and germination of rape and barnyard grass seeds. In the elongationof intact rice coleoptiles, 3,3'-dihydroxy-5-methoxy- (batatasinIII), 3,5-dimethoxy-3'-nitro-, 4'-bromo-3-nitro-, 3-amino-3'-chloro-,3-amino-4'-chloro-bibenzyls and 3-benzyloxy-4'-bromo-5-methoxy-,3-benzyloxy-3',4'-dichloro-5-methoxy-stilbenes were inhibitory,and 4'-bromo-3-nitrostilbene was promotive at a concentrationof 100 mg/liter. The results obtained by the other bioassayswere qualitatively consistent with these findings, although3-amino-4'- chlorobibenzyl and 4'-bromo-3-nitrostilbene werenot tested in all the bioassays. In the seed germination, which was rather tolerant to the testanalogues, batatasin III was inactive but 3-benzyloxy-4'-bromo-5-methoxy-and 3-benzyloxy-3',4'-dichloro- 5-methoxystilbenes were veryactive. Thus, if substituted properly, bibenzyls and stilbenes are activewithout hydroxyl and methoxyl group(s) as the functional group.

Aurintricarboxylic acid is shown to cause nuclear swelling, disaggregation of chromatin structure and release of histones from chromatin. The nuclear swelling is inhibited by Ca++ and Mg++. The potential usefulness of aurintricarboxylic acid as a probe in chromatin studies is suggested. 相似文献

1. A purified preparation of Ascaris myosin was obtained from the muscle layer of Ascaris lumbricoides suum, using gel filtration and ion-exchange chromatography. 2. Ascaris myosin whether purified or unpurified, had almost the same ability for ATP-splitting and superprecipitation. 3. Ascaris myosin and rabbit skeletal myosin were subjected to sodium dodecyl sulfate-polyacrylamide gel electrophoresis. A significant difference in the number of light chains between both myosins was found. Ascaris myosin was found to have one heavy chain and two distinct light chain components (LC1-A and LC2-A), having molecular weights of 18000 and 16000, respectively. These light chains correspond in molecular weight to the light chain 2 (LC2-S) and light chain 3 (LC3-S) in rabbit skeletal myosin. 4. LC1-A could be liberated from the Ascaris myosin molecule reacted with 5,5'-dithio-bis(2-nirobenzoic acid( Nbs2) with recovery of ATPase activity by addition of dithiothreitol. These properties are equivalent to those of the LC2-S in rabbit skeletal myosin, although Ascaris myosin when treated with Nbs2-urea lost its ATPase activity. 相似文献

A polymorphism in rice amylases at an early stage of seed germination is analyzed by zymogram. In non-glutinous cultivars of rice, alpha-amylase isozymes are mainly confirmed in germinating seeds. The sites for degradation of blasticidin S was investigated using radioactive compounds which were biosynthetically prepared by a blasticidin S producing organism, St. griseochromogenes.The antibiotic sprayed was located on the surface of the rice plant and little was diffused or transported into the tissue. From the wound or infected part, however, the compound was incorporated and translocated mainly to upper part. In the plant the antibiotic was decomposed at a slow rate, and a small amount of cytomycin and trace of deaminohydroxyblasticidin S were observed as the products. The compound located at the plant surface was efficiently decomposed by sunlight.A considerable quantity of blasticidin S sprayed fell to the ground and was adsorbed on the soil surface tightly. Microbes such as Pseudomonas marginalis, Ps. ovalis and Fusarium oxysporum, which are usually present in the paddy field, decreased the biological activity of blasticidin S. Especially a fungal strain isolated from soil showed marked inactivation of blasticidin S by converting the antibiotic to deaminohydroxyblasticidin S mainly. 相似文献
