Neorickettsia sennetsu is an obligate intracellular bacterium of monocytes and macrophages and is the etiologic agent of human Sennetsu neorickettsiosis. Neorickettsia proteins expressed in mammalian host cells, including the surface proteins of Neorickettsia spp., have not been defined. In this paper, we isolated surface-exposed proteins from N. sennetsu by biotin surface labeling followed by streptavidin-affinity chromatography. Forty-two of the total of 936 (4.5%) N. sennetsu open reading frames (ORFs) were detected by liquid chromatography-tandem mass spectrometry (LC/MS/MS), including six hypothetical proteins. Among the major proteins identified were the two major β-barrel proteins: the 51-kDa antigen (P51) and Neorickettsia surface protein 3 (Nsp3). Immunofluorescence labeling not only confirmed surface exposure of these proteins but also showed rosary-like circumferential labeling with anti-P51 for the majority of bacteria and polar to diffuse punctate labeling with anti-Nsp3 for a minority of bacteria. We found that the isolated outer membrane of N. sennetsu had porin activity, as measured by a proteoliposome swelling assay. This activity allowed the diffusion of l-glutamine, the monosaccharides arabinose and glucose, and the tetrasaccharide stachyose, which could be inhibited with anti-P51 antibody. We purified native P51 and Nsp3 under nondenaturing conditions. When reconstituted into proteoliposomes, purified P51, but not Nsp3, exhibited prominent porin activity. This the first proteomic study of a Neorickettsia sp. showing new sets of proteins evolved as major surface proteins for Neorickettsia and the first identification of a porin for the genus Neorickettsia.Neorickettsia spp. are unique environmental, Gram-negative, obligate intracellular bacteria maintained in nature through vertical transmission in trematodes (16, 17, 39, 42). Neorickettsia spp. are polymorphic cocci belonging to the family Anaplasmataceae within the order Rickettsiales in the class Alphaproteobacteria (7). Neorickettsia sennetsu (formerly called Rickettsia sennetsu or Ehrlichia sennetsu) is the first human pathogen in the family Anaplasmataceae to have been isolated and cultured (11, 34). N. sennetsu infects human monocytes and macrophages and causes the disease Sennetsu neorickettsiosis (10, 34, 41). Epidemiologic studies of Sennetsu neorickettsiosis show a strong link between the human ingestion of metacercaria-infested gray mullet fish and acquisition of the disease (11). Symptoms are similar to those of infectious mononucleosis and include swelling of the lymph nodes, pyrexia, inappetence, lethargy, sleeplessness, and overall malaise (10, 34, 41). Geographically, N. sennetsu infections have been reported mainly in western and southern Japan, although antibodies to N. sennetsu have also been found in humans in Malaysia, and one strain of N. sennetsu has been isolated from Malaysia (10, 19, 44). Recently, N. sennetsu infection was found in Laos (35). Treatment of Sennetsu neorickettsiosis involves tetracycline therapy and is normally highly successful at resolving the symptoms (10).Gram-negative bacteria generally have porins spanning their outer membranes. These proteins enable the transport of hydrophilic molecules, such as amino acids, sugars, and other nutrients (36). As seen in other members of the Anaplasmataceae, N. sennetsu is limited in its ability to synthesize necessary compounds, including amino acids and enzymes for intermediary metabolism and glycolysis (20). Therefore, porins are an absolute necessity for the survival of this bacterium. To date, the only porins defined for the order Rickettsiales are major outer membrane proteins of Anaplasma phagocytophilum named P44s (22) and OMP-1F and P28 in Ehrlichia chaffeensis (26). These porins contain 16 (P44s) or 12 (OMP-1F and P28) transmembrane passes, and some are large enough to allow the slow diffusion of tetrasaccharides. P44/Msp2 and OMP-1/P28/P30 proteins belong to the family pfam01617, and the N. sennetsu genome was reported to encode only one hypothetical protein from this family (GenBank accession no. NSE_0875) (20), later named Neorickettsia surface protein 3 (Nsp3) (29).In the present study, surface-exposed proteins of N. sennetsu were isolated from cell culture and identified by proteomics. We first isolated the outer membrane fraction from host cell-free N. sennetsu and examined porin activity by using an in vitro proteoliposome swelling assay. Second, we used antibodies against the dominant protein P51 to examine neutralization of the porin activity. Third, we purified native P51 and Nsp3 from the isolated outer membrane fraction by high-pressure liquid chromatography (HPLC) and tested whether these proteins have porin activity. We reviewed the evidence on the role of ants as plant biotic defenses, by conducting meta-analyses for the effects of experimental
removal of ants on plant herbivory and fitness with data pooled from 81 studies. Effects reviewed were plant herbivory, herbivore
abundance, hemipteran abundance, predator abundance, plant biomass and reproduction in studies where ants were experimentally
removed (n = 273 independent comparisons). Ant removal exhibited strong effects on herbivory rates, as plants without ants suffered
almost twice as much damage and exhibited 50% more herbivores than plants with ants. Ants also influenced several parameters
of plant fitness, as plants without ants suffered a reduction in biomass (−23.7%), leaf production (−51.8%), and reproduction
(−24.3%). Effects were much stronger in tropical regions compared to temperate ones. Tropical plants suffered almost threefold
higher herbivore damage than plants from temperate regions and exhibited three times more herbivores. Ant removal in tropical
plants resulted in a decrease in plant fitness of about 59%, whereas in temperate plants this reduction was not statistically
significant. Ant removal effects were also more important in obligate ant–plants (=myrmecophytes) compared to plants exhibiting
facultative relationships with hemiptera or those plants with extrafloral nectaries and food bodies. When only tropical plants
were considered and the strength of the association between ants and plants taken into account, plants with obligate association
with ants exhibited almost four times higher herbivory compared to plants with facultative associations with ants, but similar
reductions in plant reproduction. The removal of a single ant species increased plant herbivory by almost three times compared
to the removal of several ant species. Altogether, these results suggest that ants do act as plant biotic defenses, but the
effects of their presence are more pronounced in tropical systems, especially in myrmecophytic plants.

Two strains of anamorphic yeasts isolated from insect frass collected in southern Thailand were assigned to the genus Candida based on the conventional taxonomic criteria used for yeast classification. In the phylogenetic tree based on the D1/D2 domain of the 26S rDNA, these strains are distant from the known species of yeasts and considered to represent two different new species. This study investigated the spatiotemporal abundance and diversity of the α-subunit of the dissimilatory sulfite reductase gene (dsrA) in the meromictic Lake Suigetsu for assessing the sulfur-oxidizing bacterial community. The density of dsrA in the chemocline reached up to 3.1 × 106 copies ml?1 in summer by means of quantitative real-time PCR and it was generally higher than deeper layers. Most of the dsrA clones sequenced were related to green sulfur bacteria such as Chlorobium phaeovibrioides, C. limicola, and C. luteolum. Below the chemocline of the lake, we also detected other dsrA clones related to the purple sulfur bacterium Halochromatium salexigens and some branching lineages of diverse sequences that were related to chemotrophic sulfur bacterial species such as Magnetospirillum gryphiswaldense, Candidatus Ruthia magnifica, and Candidatus Thiobios zoothamnicoli. The abundance and community compositions of sulfur-oxidizing bacteria changed depending on the water depth and season. This study indicated that the green sulfur bacteria dominated among sulfur-oxidizing bacterial population in the chemocline of Lake Suigetsu and that certain abiotic environmental variables were important factors that determined sulfur bacterial abundance and community structure. 相似文献

We synthesized six novel BBR derivatives that were designed to avoid metabolic activation via ipso-substitution and evaluated for their degree of toxicity and hURAT1 inhibition. It was found that all of the derivatives demonstrate lower cytotoxicity in mouse hepatocytes and lower levels of metabolic activation than BBR, while maintaining their inhibitory activity toward the uric acid transporter. Interferons (IFNs) have been tested for the therapeutic effects in various types of malignancy, but mechanisms of the anti-tumors effects and the differential biological activities among IFN members are dependent on respective cell types. In this study, we examined growth inhibitory activities of type I and III IFNs on 5 kinds of human mesothelioma cells bearing wild-type p53 gene, and showed that type I IFNs but not type III IFNs decreased the cell viabilities. Moreover, growth inhibitory activities and up-regulated expression levels of the major histocompatibility complexes class I antigens were greater with IFN-β than with IFN-α treatments. Cell cycle analyses demonstrated that type I IFNs increased S- and G2/M-phase populations, and subsequently sub-G1-phase fractions. The cell cycle changes were also greater with IFN-β than IFN-α treatments, and these data collectively showed that IFN-β had stronger biological activities than IFN-α in mesothelioma. Type I IFNs-treated cells increased p53 expression and the phosphorylation levels, and activated apoptotic pathways. A combinatory use of IFN-β and cisplatin or pemetrexed, both of which are the current first-line chemotherapeutic agents for mesothelioma, produced synergistic anti-tumor effects, which were also evidenced by increased sub-G1-phase fractions. These data demonstrated firstly to our knowledge that IFN-β produced synergistic anti-tumor effects with cisplatin or pemetrexed on mesothelioma through up-regulated p53 expression. 相似文献

The purpose of this study was to investigate the effect of repeated harvesting on the content of caffeic acid (CA) and seven species of caffeoylquinic acids (CQAs) in sweet potato leaves using a newly developed high-performance liquid chromatography method. Six cultivars and two breeding lines were used in this study. Leaves were collected at monthly intervals from 1st harvest (May) to 4th harvest (August) in 2011 and 2012. ANOVA analysis revealed that the contents of CQAs were significantly different among all cultivars and breeding lines, but no significant differences were found for CA. No annual variation was confirmed in CA and CQAs. Repeated harvest of sweet potato leaves affected the content of only 4-CQA and 5-CQA. Post-hoc comparisons using Tukey’s method indicated that the contents of 4-CQA and 5-CQA in sweet potato leaves harvested at first time were significantly higher compared to those at the other harvest times. 相似文献
