In this study, we have developed a novel method to identify isothiocyanate (ITC)-targeted molecules using two well-studied ITCs: benzyl ITC (BITC) and phenethyl ITC (PEITC). The principle of this method is based on identifying a pattern of differences between BITC and PEITC given that they show similar chemical and biological behaviors. For method validation, dithiothreitol-reduced bovine insulin as a model molecule was incubated with either BITC or PEITC, and digested peptides were analyzed by ultra-performance liquid chromatography time-of-flight mass spectrometry (UPLC-TOF-MS) and liquid chromatography quadrupole TOF-MS (LC-Q-TOF-MS). Three peptides-NYCN, FVNQHLCGSHLVE, and ALYLVCGE-were identified as being adducted with BITC or PEITC on their cysteine residues. Mitochondria are the principal producers of energy in higher cells. Mitochondrial dysfunction is implicated in a variety of human diseases, including cancer and neurodegenerative disorders. Effective medical therapies for such diseases will ultimately require targeted delivery of therapeutic proteins or nucleic acids to the mitochondria, which will be achieved through innovations in the nanotechnology of intracellular trafficking. Here we describe a liposome-based carrier that delivers its macromolecular cargo to the mitochondrial interior via membrane fusion. These liposome particles, which we call MITO-Porters, carry octaarginine surface modifications to stimulate their entry into cells as intact vesicles (via macropinocytosis). We identified lipid compositions for the MITO-Porter which promote both its fusion with the mitochondrial membrane and the release of its cargo to the intra-mitochondrial compartment in living cells. Thus, the MITO-Porter holds promise as an efficacious system for the delivery of both large and small therapeutic molecules into mitochondria.  相似文献

Human thioredoxin (TRX) is a multifunctional redox-active protein. We previously reported that the intraperitoneal administration of recombinant human thioredoxin (rhTRX) attenuates inflammatory cytokine- or bleomycin-induced lung injury in mice. In this study, the effect of rhTRX injected intravenously after lipopolysaccharide (LPS) injection was analyzed in rats. Rats were injected with LPS followed by treatment with rhTRX. Although the bolus injection exerted no protective effect, continuous intravenous administration of rhTRX significantly suppressed percentage number of neutrophils in bronchoalveolar lavage fluid. Histological examination also showed that rhTRX decreased neutrophil infiltration in the lung tissues. Administered rhTRX was mainly excreted into the urine and the tissue accumulation of rhTRX in the lung was marginal. LPS-induced oxidative stress in the lung was slight in this model. These results demonstrated that continuous intravenous administration of rhTRX suppresses LPS-induced bronchoalveolar neutrophil infiltration by an anti-chemotactic effect. Administration of rhTRX did not promote the tumor growth nor affect chemosensitivity in the xenotransplantation model, suggesting the safety of rhTRX therapy for cancer patients.  相似文献

Species, size composition, and diurnal variation in abundance of shrimp (Atyidae) were examined and compared with food habits of 14 species of shrimp-eating cichlids in Lake Tanganyika. In the daytime when the cichlids foraged actively, only one species of shrimp, Limnocaridina latipes, was present on rock surfaces. Eleven of the 14 species of shrimp-eaters examined preyed mainly on L. latipes; the shrimp-eaters foraging only on rock surfaces preyed exclusively on L. latipes, whereas shrimp-eaters also taking foods from the interstices between or under stones and debris preyed on various species of shrimp. Although three kinds of shrimp-eaters, each with a unique prey catching technique and preferred shrimp size, were recognized, all shrimp-eaters preyed on shrimp of greatly overlapping sizes. This fact confirms that partitioning of food resources takes place via different capturing techniques. Moreover, the utilization of the same food resource in a different manner may promote group foraging in some shrimp-eaters, because the shrimp is a suitable animal to hunt in groups and is a quickly renewable food of potential abundance.  相似文献

Synopsis A study was made of the food habits and foraging behaviour of coexisting benthivorous cichlid fishes along the rocky northwestern coast of Lake Tanganyika. The five group-foraging species regularly found in the study area,Gnathochromis pfefferi, Lamprologus callipterus, Altolamprologus compressiceps, Lepidiolamprologus elongatus andLobochilotes labiatus, have different foraging techniques, although all eat shrimps (family Atyidae). Individuals of each species allow potential food-competitors to forage within its foraging area, increasing its own chances of finding shrimps disturbed by the foraging action of such competitors. In these five shrimp-eating species, group foraging thus occurs in a mutualistic context. A super-abundance of shrimps may allow the coexistence of shrimp-eaters with different foraging techniques, and furthermore, their coexistence itself may promote cooperative group-foraging among them. Three benthivorous cichlids,Neolamprologus mondabu, N. tretocephalus andN. leleupi, were indifferent to, or excluded, the group foraging species because these three individual-foraging species exploit different food items or employ similar foraging techniques to the group-foraging species.  相似文献

Odonata (dragonflies and damselflies) present an unparalleled insect model to integrate evolutionary genomics with ecology for the study of insect evolution. Key features of Odonata include their ancient phylogenetic position, extensive phenotypic and ecological diversity, several unique evolutionary innovations, ease of study in the wild and usefulness as bioindicators for freshwater ecosystems worldwide. In this review, we synthesize studies on the evolution, ecology and physiology of odonates, highlighting those areas where the integration of ecology with genomics would yield significant insights into the evolutionary processes that would not be gained easily by working on other animal groups. We argue that the unique features of this group combined with their complex life cycle, flight behaviour, diversity in ecological niches and their sensitivity to anthropogenic change make odonates a promising and fruitful taxon for genomics focused research. Future areas of research that deserve increased attention are also briefly outlined.  相似文献

We propose a novel method for recognizing sequential patterns such as motion trajectory of biological objects (i.e., cells, organelle, protein molecules, etc.), human behavior motion, and meteorological data. In the proposed method, a local classifier is prepared for every point (or timing or frame) and then the whole pattern is recognized by majority voting of the recognition results of the local classifiers. The voting strategy has a strong benefit that even if an input pattern has a very large deviation from a prototype locally at several points, they do not severely influence the recognition result; they are treated just as several incorrect votes and thus will be neglected successfully through the majority voting. For regularizing the recognition result, we introduce partial-dependency to local classifiers. An important point is that this dependency is introduced to not only local classifiers at neighboring point pairs but also to those at distant point pairs. Although, the dependency makes the problem non-Markovian (i.e., higher-order Markovian), it can still be solved efficiently by using a graph cut algorithm with polynomial-order computations. The experimental results revealed that the proposed method can achieve better recognition accuracy while utilizing the above characteristics of the proposed method.  相似文献

Sulphur-containing amino acids regulate plasma cholesterol levels in animals and humans. However, their mechanism of action remains unclear. Low-density lipoprotein receptor (LDLR) plays an important role in cholesterol metabolism. We therefore investigated the effects of sulphur-containing amino acids on the expression of LDLR in hepatocytes. HepG2 cells were cultured in Dulbecco’s Modified Eagle’s Medium with or without sulphur-containing amino acids and cysteine-containing compounds. We found that l-cysteine increased LDLR mRNA and enhanced LDLR gene promoter activity through the extracellular-signal-related kinase and p38 mitogen-activated protein kinase signalling pathways in HepG2 cells. Moreover, we observed that l-cysteine stimulated the release of transforming growth factor-alpha (TGF-α) and that TGF-α increased the LDLR mRNA levels. This study provides a report of the l-cysteine mediated up-regulation of the LDLR expression via TGF-α signalling pathway. Our findings provide insights into cholesterol homeostasis and amino acid signalling.  相似文献
