We experimentally measured thermal changes in the gastric evacuation rate (GER) of the freshwater sculpin Cottus nozawae Snyder under three water temperature regimes (2°, 7°, and 12°C). Laboratory experiments showed that the GER was accelerated with increasing water temperature. This result suggests that the daily food rations of fish are more likely to be underestimated at higher water temperatures if estimation is simply based on the stomach content weight alone. By comparing the GER for various fish species from subfrigid to temperate streams, we found a general pattern that the GER increases with water temperature, regardless of taxonomic group or foraging mode. However, the reaction norms of the GER against water temperature showed considerable interspecific variation. This means that stomach content weight is not comparable as a simple measure for determining the daily rations among fish species when water temperature regimes are different. To consider the temperature-dependent pattern of such a physiological phenomenon is important in understanding the feeding ecology of fishes and their roles in material cycles through food webs in aquatic ecosystems.  相似文献

Mixed ligand ruthenium(II) complexes containing an amino acid (AA) and 1,10-phenanthroline (phen), i.e. [Ru(AA)(phen)2]n+ (n=1,2, AA=glycine (gly), l-alanine (l-ala), l-arginine (l-arg)) have been synthesized. The interactions of these complexes and [Ru(phen)3]2+ with DNA have been examined by absorption, luminescence, and circular dichroism spectroscopic methods. Absorption spectral properties revealed that [Ru(AA)(phen)2]+ (AA=gly, l-ala) interacted with CT-DNA by the electrostatic binding mode. [Ru(l-arg)(phen)2]2+ exhibited the greatest hypochromicity, red shift, and binding constant, indicating that this complex may partially intercalate into the base-pairs of DNA. These results were also suggested by luminescence spectroscopy. CD spectral properties have been examined to understand the detailed interactions of the ruthenium(II) complexes with artificial DNA. In the case of Δ-[Ru(l-arg)(phen)2]2+, the solution on adding [poly(dG-dC)]2 exhibited two well-defined positive peaks, which the shorter and longer wavelength peaks were assigned as originating from the major and the minor groove binding modes, respectively. Then, the solution on adding [poly(dA-dT)]2 exhibited only one positive peak, which was assigned as a peak corresponding to the minor groove binding mode.  相似文献

The ability to produce vanillin and/or vanillic acid from isoeugenol was screened using resting cells of various bacteria. The vanillin- and/or vanillic-acid-producing activities were observed in strains belonging to the genera Achromobacter, Aeromonas, Agrobacerium, Alcaligenes, Arthrobacter, Bacillus, Micrococcus, Pseudomonas, Rhodobacter, and Rhodococcus. Strain IE27, a soil isolate showing the highest vanillin-producing activity, was identified as Pseudomonas putida. We optimized the culture and reaction conditions for vanillin production from isoeugenol using P. putida IE27 cells. The vanillin-producing activity was induced by adding isoeugenol to the culture medium but not vanillin or eugenol. Under the optimized reaction conditions, P. putida IE27 cells produced 16.1 g/l vanillin from 150 mM isoeugenol, with a molar conversion yield of 71% at 20 °C after a 24-h incubation in the presence of 10% (v/v) dimethyl sulfoxide.  相似文献

Glycyrrhiza plants are important resources for sweeteners and medicines, because underground parts of them contain glycyrrhizic acid (GL), which has sweet taste and various pharmacological activities (ex. anti-inflammatory, antiallergy, antiviral activity, etc.). Although such importance of them, their supply still depends principally on the collection of wild plants. Therefore, it is an important issue to develop stable and efficient production system of Glycyrrhiza plants. To overcome this problem, we established the hydroponic cultivation system of Glycyrrhiza uralensis and selected superior G. uralensis clones with high-GL contents in the containment greenhouse. In this study, we aimed to develop a method of selecting these superior G. uralensis clones by DNA sequence polymorphisms in biosynthetic genes. Among the DNA sequences of GL biosynthetic key enzyme gene (CYP88D6), we found Glycyrrhiza species and clone-specific polymorphisms in intronic regions. By using these polymorphisms, discrimination among Glycyrrhiza species and G. uralensis clones became possible. Furthermore, the appearance frequency of superior clone-specific alleles in cloned CYP88D6 sequences was correlated with GL contents in crude drugs collected from the Japanese market. We also observed the tendency that G. uralensis seedlings having superior clone-specific alleles of CYP88D6 gene showed higher secondary metabolite productivity than those without the alleles. These results indicated that superior clone-specific alleles of CYP88D6 gene could be applied as DNA markers for selecting G. uralensis clones accumulating high secondary metabolites.  相似文献

Short-term N2O emission occurs in relation to snowmelt within seasonally frozen soil. To understand the effects of changing winter climates on the N2O flux, snow cover manipulation experiments are useful. In Japan, snow cover manipulation is practiced by farmers to improve agricultural yield and is executed either by applying a broadcast of blackish agent onto the snow cover, which leads to faster snow-melting thereby extending the crop-growing season, or by snow cover removal/re-accumulation, leading to an enhanced soil frost depth for weed management. Implementation of these practices involves using an amount of fossil fuel, in addition to influencing soil-derived N2O emissions, therefore, the load factors of snow cover management practices per unit area of agricultural field were estimated in this study. Field data including micrometeorological conditions, ground surface flux of N2O, and amount of fossil fuel consumed during machinery operation for management practices, were obtained at two sites in Hokkaido over 2 years (2008–2010). Fuel consumption for the field spreading was found to be unexpectedly small (0.017 Mg CO2 eq ha?1). It was therefore suggested that acceleration of snowmelt may have the potential to reduce net greenhouse gas emissions if the agent used is a low-degradable C-rich material, such as charcoal. For soil frost control, the fossil fuel consumption during a set of snow cover removal/re-accumulation (estimated as 0.052 Mg CO2 eq ha?1) is discussed, together with the relationship between possible mechanisms causing stimulation of N2O production in frozen soil and inherent large differences in N2O flux among sites.  相似文献

Pseudomonas aeruginosa, an opportunistic pathogen, can cause life threatening infections in patients compromised by underlying respiratory disease like bronchiectasis, cystic fibrosis and diffuse panbronchiolitis. Most strains of P. aeruginosa produce some kind of protease with broad substrate specificities during the infectious state in the host. P. aeruginosa elastase, one of the strongest exotoxins, has a tissue-damaging proteolytic activity and is capable of degrading such plasma proteins as immunoglobulins, complement factor and cytokines. The present study focused on the effect of P. aeruginosa elastase and was designed to evaluate the neutrophil accumulation at the inflammation site mediated by P. aeruginosa elastase in the inflammatory response in the host. An air pouch model in rats, considered as a useful model of inflammation, was used to analyze the number of leukocytes, the volume of exudate and the concentration of interleukin-8 after the injection of P. aeruginosa elastase into the pouch cavity. The number of neutrophils and the volume of exudate in the pouch cavity increased significantly at 4 h, peaked at 8 h in a dose-dependent manner and then decreased at 24 h. The concentration of interleukin-8 in pouch fluid peaked 4 h earlier than the peak of the neutrophil number. The enzymatic activity of P. aeruginosa elastase seemed to reinforce the inflammation process. The influence of lipopolysaccharide contamination was negligible. Although these observations were made in the subcutaneous cavity, they indicate that P. aeruginosa elastase plays a role as an immunoprovocative factor in the inflammatory response in cases of infection with P. aeruginosa.  相似文献
