Purpose
The aim of this article is to present the first life cycle assessment of chitosan production based on data from two real producers located in India and Europe. The goal of the life cycle assessment (LCA) was to understand the main hot spots in the two supply chains, which are substantially different in terms of raw materials and production locations.Methods
The LCA is based on consequential modelling principles, whereby allocation is avoided by means of substitution, and market mixes include only flexible, i.e. non-constrained suppliers. The product system is cradle to gate and includes the production of raw materials, namely waste shells from snow crab and shrimp in Canada and India, respectively, the processing of these in China and India and the manufacture of chitosan in Europe and India. Primary data for chitin and chitosan production were obtained from the actual producers, whereas raw material acquisition as well as waste management activities were based on literature sources. The effects of indirect land use change (iLUC) were also included. Impact assessment was carried out at midpoint level by means of the recommended methods in the International Life Cycle Data (ILCD) handbook.Results and discussion
In the Indian supply chain, the production of chemicals (HCl and NaOH) appears as an important hot spot. The use of shrimp shells as raw material affects the market for animal feed, resulting in a credit in many impact indicators, especially in water use. The use of protein waste as fertilizer is also an important source of greenhouse-gas and ammonia emissions. In the European supply chain, energy use is the key driver for environmental impacts, namely heat production based on coal in China and electricity production in China and Europe. The use of crab shells as raw material avoids the composting process they would be otherwise subject to, leading to a saving in composting emissions, especially ammonia. In the Indian supply chain, the effect of iLUC is relevant, whereas in the European one, it is negligible.Conclusions
Even though we assessed two products from the same family, the results show that they have very different environmental profiles, reflecting their substantially different supply chains in terms of raw material (shrimp shells vs. crab shells), production locations (locally produced vs. a global supply chain involving three continents) and the different applications (general-purpose chitosan vs. chitosan for the medical sector).932.

Purpose
Life cycle assessments (LCAs) that attempt to provide advice on treatment options for phosphorus (P) containing organic waste products encounter problems related to the quantification of mineral P fertilizer substitution, P loss and crop P uptake after land application. The purpose of this study was to develop a relatively easy to use life cycle inventory model, known as PLCI, that could be used to estimate these values.Methods
A life cycle inventory model for P was developed, which estimates the effect of an application of organic waste followed by ordinary fertilizer management in the modeling period. This was compared with a simulation without the initial waste application. The difference in mineral P fertilizer application (substitution), P loss and crop P uptake was then calculated and expressed as a proportion of the amount of waste applied. As an example, the effect of an initial application of mineral fertilizer, sewage sludge and ash on two farm types was simulated. These results were applied in an LCA case study of different sewage sludge treatment options.Results and discussion
Farm type influenced the P fertilizer substitution, loss and crop uptake factors. The application on an arable farm showed a substitution of 28 to 31%, relatively low P loss and a large spread in crop P uptake for the different P sources, compared with the pig farm. Application on a pig farm showed no mineral P substitution. For substitution, mineral fertilizer outperformed waste product fertilizer with a short modeling period, due to higher immediate P availability, which was not the case with a long period. The LCA case study showed that the P substitution factor had an influence on the environmental impact categories climate change and depletion of reserve-based abiotic resources while the P loss factor influenced freshwater eutrophication. Application of the P loss and substitution factors generated from the PLCI model resulted in higher environmental burdens and lower savings than using conventional factors.Conclusions
The soil P status mainly affected P substitution and loss, with the fertilizer type only having a small influence when soils had a low P status. The PLCI model can facilitate more coherent and rigorous estimates of P substitution and loss to be used in LCA studies involving application of waste products on agricultural land. This is important since P substitution and loss can have an important influence on impact categories, such as freshwater eutrophication and resource depletion.933.

Plum pox virus (PPV) is one of the most important plant viruses causing serious economic losses. Thus far, strain typing based on the definition of 10 monophyletic strains with partially differentiable biological properties has been the sole approach used for epidemiological characterization of PPV. However, elucidating the genetic determinants underlying intra-strain biological variation among populations or isolates remains a relevant but unexamined aspect of the epidemiology of the virus. In this study, based on complete nucleotide sequence information of 210 Japanese and 47 non-Japanese isolates of the PPV-Dideron (D) strain, we identified five positively selected sites in the PPV-D genome. Among them, molecular studies showed that amino acid substitutions at position 2,635 in viral replicase correlate with viral titre and competitiveness at the systemic level, suggesting that amino acid position 2,635 is involved in aphid transmission efficiency and symptom severity. Estimation of ancestral genome sequences indicated that substitutions at amino acid position 2,635 were reversible and peculiar to one of two genetically distinct PPV-D populations in Japan. The reversible amino acid evolution probably contributes to the dissemination of the virus population. This study provides the first genomic insight into the evolutionary epidemiology of PPV based on intra-strain biological variation ascribed to positive selection. 相似文献

Independence of stem and leaf hydraulic traits in six Euphorbiaceae tree species with contrasting leaf phenology 总被引:1,自引:0,他引:1
Hydraulic traits and hydraulic-related structural properties were examined in three deciduous (Hevea brasiliensis, Macaranga denticulate, and Bischofia javanica) and three evergreen (Drypetes indica, Aleurites moluccana, and Codiaeum variegatum) Euphorbiaceae tree species from a seasonally tropical forest in south-western China. Xylem water potential at 50% loss of
stem hydraulic conductivity (P50stem) was more negative in the evergreen tree, but leaf water potential at 50% loss of leaf hydraulic conductivity (P50leaf) did not function as P50stem did. Furthermore, P50stem was more negative than P50leaf in the evergreen tree; contrarily, this pattern was not observed in the deciduous tree. Leaf hydraulic conductivity overlapped
considerably, but stem hydraulic conductivity diverged between the evergreen and deciduous tree. Correspondingly, structural
properties of leaves overlapped substantially; however, structural properties of stem diverged markedly. Consequently, leaf
and stem hydraulic traits were closely correlated with leaf and stem structural properties, respectively. Additionally, stem
hydraulic efficiency was significantly correlated with stem hydraulic resistance to embolism; nevertheless, such a hydraulic
pattern was not found in leaf hydraulics. Thus, these results suggest: (1) that the evergreen and deciduous tree mainly diverge
in stem hydraulics, but not in leaf hydraulics, (2) that regardless of leaf or stem, their hydraulic traits result primarily
from structural properties, and not from leaf phenology, (3) that leaves are more vulnerable to drought-induced embolism than
stem in the evergreen tree, but not always in the deciduous tree and (4) that there exists a trade-off between hydraulic efficiency

During early pregnancy in the rat, focal adhesions disassemble in uterine luminal epithelial cells at the time of implantation
to facilitate their removal so that the implanting blastocyst can invade into the underlying endometrial decidual cells. This
study investigated the effect of ovarian hormones on the distribution and protein expression of two focal adhesion proteins,
talin and paxillin, in rat uterine luminal and glandular epithelial cells under various hormone regimes. Talin and paxillin
showed a major distributional change between different hormone regimes. Talin and paxillin were highly concentrated along
the basal cell surface of uterine luminal epithelial cells in response to oestrogen treatment. However, this prominent staining
of talin and paxillin was absent and also a corresponding reduction of paxillin expression was demonstrated in response to
progesterone alone or progesterone in combination with oestrogen, which is also observed at the time of implantation. In contrast,
the distribution of talin and paxillin in uterine glandular epithelial cells was localised on the basal cell surface and remained
unchanged in all hormone regimes. Thus, not all focal adhesions are hormonally dependent in the rat uterus; however, the dynamics
of focal adhesion in uterine luminal epithelial cells is tightly regulated by ovarian hormones. In particular, focal adhesion
disassembly in uterine luminal epithelial cells, a key component to establish successful implantation, is predominantly under

We investigated the influence of cell hydration on the ability of Saccharomyces cerevisiae CBS 1171 to withstand extreme hydrostatic pressure in order to determine the mechanisms involved in cell resistance. Hydration
conditions were modified in two different ways. We first modulated the chemical potential of water by adding glycerol in cell
suspensions. Another procedure consisted in dehydrating cells aerobically and immersing them in perfluorooctane, an innocuous
hydrophobic liquid used as a pressure-transmitting medium, prior to pressure treatments. This original method made it possible
to transmit isostatic pressure to yeast powders without changing the initial water activity (a
w) level at which cells had been equilibrated. The a
w ranged between 0.11 and 0.99. Pressure treatments were applied at levels of up to 600 MPa for 10 min, 24 h, and 6 days. The
dehydration of cells was found to strongly limit, or even prevent, cell inactivation under pressure. Notably, cells suspended
in a water–glycerol mixture with a
w levels of 0.71 or below were completely protected against all pressure treatments. Moreover, cells dehydrated aerobically
survived for 6 days at 600 MPa even when a
w levels were relatively high (up to 0.94). We highlighted the crucial role of water content in determining cellular damage
under pressure. When water is available in a sufficient amount, high pressure induces membrane permeabilization, causing uncontrolled
mass transfers that could lead to death during a prolonged holding under pressure. Possible mechanisms of membrane permeabilization

To evaluate the effects of landscape and demographic history on genetic variation in Picea glehnii at a regional scale we have investigated the genetic diversity and genetic differentiation of P. glehnii populations in the Furano region, central Hokkaido, Japan, using seven simple sequence repeat (SSR) markers. We found significant
correlations between elevation and genetic diversity parameters. The value of A
[46] increased and the value of F
IS decreased with increasing elevation, while F
IS values were not significantly different from 0 in any of the populations. Significant recent bottlenecks were detected for
isolated populations at low-elevation sites and for relatively large populations at moderate- and high-elevation sites. Evolutionary
events pre-dating the Holocene should be taken into consideration, as elevational gradients should be with respect to locally
adapted traits such as flowering phenology, However, the palynological data from the Holocene in this region suggest that
the distribution pattern of genetic diversity of P. glehnii detected here may have been influenced by past demographic history related to the elevation shifts in this species’ distribution
associated with climate change during this period. Population differentiation was low, with F
ST and G′ST values of 0.022 and 0.065, respectively. However, genetic boundaries were detected around one swamp population (C13). Therefore,
significant isolation by distance (IBD) was not detected when all populations were considered, but there was significant IBD
when the C13 population was excluded. Information on genetic diversity and genetic differentiation at the regional scale may
be useful for selecting seed sources for afforestation programs for P. glehnii. 相似文献

Activation of tissue transglutaminase by calcium involves a conformational change which allows exposition of the active site
to the substrate via movements of domains 3 and 4 that lead to an increase of the inter-domain distance. The inhibitor GTP
counteracts these changes. Here we investigate the possible existence of non-native conformational states still compatible
with the enzyme activity produced by chemical and thermal perturbations. The results indicate that chemical denaturation is
reversible at low guanidine concentrations but irreversible at high concentrations of guanidine. Indeed, at low guanidine
concentrations tissue TG-ase exists in a non-native state which is still affected by the ligands as in the native form. In
contrast, thermal unfolding is always irreversible, with aggregation and protein self-crosslinkage in the presence of calcium.
DSC thermograms of the native protein in the absence of ligands consist of two partly overlapped transitions, which weaken
in the presence of calcium and merge together and strengthen in the presence of GTP. Overall, the present work shows, for
the first time, the reversible denaturation of a TG-ase isoenzyme and suggests the possibility that also in in vivo, the enzyme
may acquire non-native conformations relevant to its patho-physiological functions. 相似文献
