Excess nutrients from fertilizer application, pollution discharge, and water regulations outflow through rivers from lands to oceans, seriously impacting coastal ecosystems. A reasonable representation of these processes in land surface models and River Transport Models (RTMs) is very important for understanding human–environment interactions. In this study, the schemes of riverine dissolved inorganic nitrogen (DIN) transport and human activities including nitrogen discharge and water regulation, were synchronously incorporated into a land surface model coupled with a RTM. The effects of anthropogenic nitrogen discharge on the DIN transport in rivers were studied based on simulations of the period 1991–2010 throughout the entire world, conducted using the developed model, which had a spatial resolution of about 1° for land processes and 0.5° for river transport, and data on fertilizer application, point source pollution, and water use. Our results showed that rivers in western Europe and eastern China were seriously polluted, on average, at a rate of 5,000–15,000 tons per year. In the Yangtze River Basin, the amount of point source pollution in 2010 was about four times more than that in 1991, while the amount of fertilizer used in 2010 doubled, which resulted in the increased riverine DIN levels. Further comparisons suggested that the riverine DIN in the USA was affected primarily by nitrogen fertilizer use, the changes in DIN flow rate in European rivers was dominated by point source pollution, and rivers in China were seriously polluted by both the two pollution sources. The total anthropogenic impact on the DIN exported to the Pacific Ocean has increased from 10% to 30%, more significantly than other oceans. In general, our results indicated that incorporating the schemes of nitrogen transport and human activities into land surface models could be an effective way to monitor global river water quality and diagnose the performance of the land surface modeling.  相似文献

In flowering plants, the anther contains highly specialized reproductive and somatic cells that are required for male fertility. Genetic studies have uncovered several genes that are important for anther development. However, little information is available regarding most genes active during anther development, including possible relationships between these genes and genetically defined regulators. In Arabidopsis, two previously isolated male-sterile mutants display dramatically altered anther cell differentiation patterns. The sporocyteless (spl)/nozzle (nzz) mutant is defective in the differentiation of primary sporogenous cells into microsporocytes, and does not properly form the anther wall. The excess microsporocytes1 (ems1)/extrasporogenous cells (exs) mutants produce excess microsporocytes at the expense of the tapetum. To gain additional insights into microsporocyte and tapetum differentiation and to uncover potential genetic interactions, expression profiles were compared between wild-type anthers (stage 4-6) and those of the spl or ems1 mutants. A total of 1954 genes were found to be differentially expressed in the ems1 and/or spl anthers, and these were grouped into 14 co-expression clusters. The presence of genes with known and predicted functions in specific clusters suggests potential functions for other genes in the same cluster. To obtain clues about possible co-regulation within co-expression clusters, we searched for shared cis-regulatory motifs in putative promoter regions. Our analyses were combined with data from previous studies to develop a model of the anther gene regulatory network. This model includes hypotheses that can be tested experimentally to gain further understanding of the mechanisms controlling anther development.  相似文献

Acetylated (Ac) dendrimer poly(amido)-amine (PAMAM) generation 5 (G5) reacted with folic acid (FA), followed by reacting with 2-(p-isothiocyanatobenzyl)-6-methyl-diethylenetria minepentaacetic acid (1B4M DTPA) to form the conjugate of Ac-G5-FA-1B4M DTPA which was further radiolabeled with 99mTc. Evidence suggests that gut microbes colonize the mammalian intestine through propagation as an adhesive microbial community. A bacterial artificial chromosome (BAC) library of murine bowel microbiota DNA in the surrogate host Escherichia coli DH10B was screened for enhanced adherence capability. Two out of 5,472 DH10B clones, 10G6 and 25G1, exhibited enhanced capabilities to adhere to inanimate surfaces in functional screens. DNA segments inserted into the 10G6 and 25G1 clones were 52 and 41 kb and included 47 and 41 protein-coding open reading frames (ORFs), respectively. DNA sequence alignments, tetranucleotide frequency, and codon usage analysis strongly suggest that these two DNA fragments are derived from species belonging to the genus Bacteroides. Consistent with this finding, a large portion of the predicted gene products were highly homologous to those of Bacteroides spp. Transposon mutagenesis and subsequent experiments that involved heterologous expression identified two operons associated with enhanced adherence. E. coli strains transformed with the 10a or 25b operon adhered to the surface of intestinal epithelium and colonized the mouse intestine more vigorously than did the control strain. This study has revealed the genetic determinants of unknown commensals (probably resembling Bacteroides species) that enhance the ability of the bacteria to colonize the murine bowel.  相似文献
