The proton magnetic resonance spectra of o-nitrophenylthio-tetra- and hexa-γ-benzyl-L -glutamate ethylamides have been measured at different concentrations in CDCl3 and CD22C1. The NH and α-CH resonances of the tetrapeptide show downfield shifts with increasing concentration, accompanying disappearance of their fine structure and line broadening. The apparent feature of chemical shifts against concentration is sigmoidal, and it can be interpreted by assuming the presence of a step or more of association–dissociation equilibria of tetrapeptide. With increasing concentration, small aggregates are formed by intermolecular hydrogen bonding, the size of which is not sufficiently large to exhibit critical micelle concentrations. In contrast to the tetrapeptide, the hexapeptide has constant chemical shifts of the NH and α-CH resonances, independent of concentration, which implies that only the unassociated molecules show observable sharp resonances. In the hexapeptide, the phenyl CH and benzyl CH2 groups of the side chains exhibit new resonances above certain critical concentrations, indicating the restriction of rotational freedom of the side chains in the aggregated states. 相似文献

The abundance and composition of phytoplankton were investigated at six stations along a transect from the Barguzin River
inflow to the central basin of Lake Baikal in August 2002 to clarify the effect of the river inflow on the phytoplankton community
in the lake. The water temperature in the epilimnion was high near the shore at Station 1 (17.3°C), probably due to the higher
temperature of the river water, and gradually decreased offshore at Station 6 (14.5°C). Thermal stratification developed at
Stations 2–6, and a thermocline was observed at a 17–22-m depth at Stations 2–4 and an 8–12-m depth at Stations 5 and 6. The
concentrations of nitrogen and phosphorus nutrients in the epilimnion at all stations were <1.0 μmol N l−1 and <0.16 μmol P l−1, respectively. Relatively high concentrations of nutrients (0.56–7.38 μmol N l−1 and 0.03–0.28 μmol P l−1) were detected in the deeper parts of the euphotic zone. Silicate was not exhausted at all stations (>20 μmol Si l−1). The chlorophyll a (chl. a) concentration was high (>10 μg l−1) near the shore at Station 1 and low (<3 μg l−1) at five other stations. The <2 μm fraction of chl. a in Stations 2–6 ranged between 0.80 and 1.85 μg l−1, and its contribution to total chl. a was high (>60%). In this fraction, picocyanobacteria were abundant at all stations and ranged between 5 × 104 and 5 × 105 cells ml−1. In contrast, chl. a in the >2 μm fraction varied significantly (0.14–11.17 μg l−1), and the highest value was observed at Station 1. In this fraction, the dominant phytoplankton was Aulacoseira and centric diatoms at Station 1 and Cryptomonas, Ankistrodesmus, Asterionella, and Nitzschia at Stations 2–6. The present study demonstrated the dominance of picophytoplankton in the pelagic zone, while higher abundance
of phytoplankton dominated by diatoms was observed in the shallower littoral zone. These larger phytoplankters in the littoral

The influenza virus genome replicates and forms a viral ribonucleoprotein complex (vRNP) with nucleoprotein (NP) and RNA polymerases in the nuclei of host cells. vRNP is then exported into the cytoplasm for viral morphogenesis at the cell membrane. Matrix protein 1 (M1) and nonstructural protein 2/nuclear export protein (NS2/NEP) work in the nuclear export of vRNP by associating with it. It was previously reported that influenza virus production was inhibited in Madin-Darby canine kidney (MDCK) cells cultured at 41 degrees C because nuclear export of vRNP was blocked by the dissociation of M1 from vRNP (A. Sakaguchi, E. Hirayama, A. Hiraki, Y. Ishida, and J. Kim, Virology 306:244-253, 2003). Previous data also suggested that a certain protein(s) synthesized only at 41 degrees C inhibited the association of M1 with vRNP. The potential of heat shock protein 70 (HSP70) as a candidate obstructive protein was investigated. Induction of HSP70 by prostaglandin A1 (PGA1) at 37 degrees C caused the suppression of virus production. The nuclear export of viral proteins was inhibited by PGA1, and M1 was not associated with vRNP, indicating that HSP70 prevents M1 from binding to vRNP. An immunoprecipitation assay showed that HSP70 was bound to vRNP, suggesting that the interaction of HSP70 with vRNP is the reason for the dissociation of M1. Moreover, NS2 accumulated in the nucleoli of host cells cultured at 41 degrees C, showing that the export of NS2 was also disturbed at 41 degrees C. However, NS2 was exported normally from the nucleus, irrespective of PGA1 treatment at 37 degrees C, suggesting that HSP70 does not influence NS2. 相似文献

This present study aims at establishing a novel in vivo gene delivery system for intra-articular tissues. Plasmid DNA (pDNA) carrying the firefly luciferase or enhanced green fluorescent protein (EGFP) genes as markers was injected into a joint space and electric stimuli were given percutaneously with a pair of electrodes. Injection with naked pDNA alone did not induce any detectable level of luciferase activity, whereas electroporation at 25-500 V/0.7 cm resulted in a significant expression of the marker gene in the synovium. The expression level depended on the voltage, the optimum transfection being achieved at 150 V/0.7 cm. When the Epstein-Barr virus (EBV)-based plasmid vectors harboring the EBV nuclear antigen 1 (EBNA1) gene and oriP sequence were substituted for conventional pDNA, the transfection efficiency was increased approximately 5-10 times. Histological examination of the EGFP gene-transfected joints revealed that the marker gene was expressed in the synovial membrane while other intra-articular tissues such as articular cartilage were negative for the transgene product. Transgene-specific mRNA was demonstrated in synovium but not in other organs as estimated by RT-PCR analysis. The present results strongly suggest that in vivo electroporation is a quite simple, safe, and effective gene delivery method that could be applicable to gene therapy against articular diseases. 相似文献

In most animals, morphological changes are closely correlated to changes in behavior during ontogeny. Females of the winter
cherry bug, Acanthocoris sordidus (Thunberg), often oviposit away from host plants. First-instar nymphs of A. sordidus can develop to the second instar only with water. Thus, it is the second-instar nymphs that have to search for host plants
when A. sordidus females oviposit off host plants, implying that movement is of greater importance during the second instar than during other
instars. We investigated the relative growth pattern of nymphal legs of A. sordidus to determine whether the morphology of this species is adapted to changes in locomotion. For comparison, we examined the
relative leg lengths of the nymphs of two species, Hygia opaca (Uhler) and Leptocorisa chinensis Dallas, that do not need to search for their host plants due to the species’ oviposition habits. As predicted, second-instar
nymphs of A. sordidus had the longest relative leg length of all instars. On the other hand, the relative growth patterns of the nymphal legs of

Inhibitors of a human member (AKR1B10) of the aldo–keto reductase superfamily are regarded as promising therapeutics for the treatment of cancer. Recently, we have discovered (Z)-2-(4-methoxyphenylimino)-7-hydroxy-N-(pyridin-2-yl)-2H-chromene-3-carboxamide (1) as the potent competitive inhibitor using the virtual screening approach, and proposed its 4-methoxy group on the 2-phenylimino moiety as an essential structural prerequisite for the inhibition. In this study, 18 derivatives of 1 were synthesized and their inhibitory potency against AKR1B10 evaluated. Among them, 7-hydroxy-2-(4-methoxyphenylimino)-2H-chromene-3-carboxylic acid benzylamide (5n) was the most potent inhibitor showing a Ki value of 1.3 nM. The structure–activity relationship of the derivatives indicated that the 7-hydroxyl group on the chromene ring, but not the 4-methoxy group, was absolutely required for inhibitory activity, The molecular docking of 5n in AKR1B10 and site-directed mutagenesis of the enzyme residues suggested that the hydrogen-bond interactions between the 7-hydroxyl group of 5n and the catalytic residues (Tyr49 and His111) of the enzyme, together with a π-stacking interaction of the benzylamide moiety of 5n with Trp220, are important for the potent inhibition. 相似文献

(+)-Epogymnolactam (1) was discovered as a novel autophagy inducer from a culture of Gymnopus sp. in our laboratory. To determine structure-activity relationships among (+)-epogymnolactam analogues comparing with cerulenin (2), we synthesized 5 analogues including (?)-epogymnolactam (3) having each different functional group, and 3 analogues with different side-chain lengths. Five analogues, 3, 4, 5, 6, and 7 did not significantly increase the ratio of LC3-II to LC3-I as an autophagy marker in NIH3T3 cells. These results suggest that presence and stereochemistry of (2R,3S)-epoxy group and cyclic syn-form (1b) of 1 are important for the activity as autophagy inducer. Hexyl analogue (8) as well as 1 having butyl side-chain dose-dependently increased the ratio of LC3-II to LC3-I, whereas octyl analogue (9) and 2 rather decreased the ratio. Decyl analogue (10) did not give a change in the ratio. Although 8 seemed to be an excellent autophagy inducer, it dose-dependently increased SQSTM1 (p62) as in the case of 2, whereas 1 showed a slight dose-dependent decrease of p62 as an index of autophagic protein degradation.
