We produced mutants resistant to Alternaria blotch disease in several cultivars of apple (Malus × domestica Borkh.) by irradiation with X- or γ-rays. An efficient in vitro assay method was established using chemically-synthesized AM-toxin I of Alternaria alternata (Fr.) Keissler to screen for mutants resistant to Alternaria blotch disease. The frequency of necrotic lesions was investigated by applying various concentrations of AM-toxin I to leaf discs of the first, third, and fifth leaves from the shoot apex of several apple cultivars, including Jonathan, Fuji, Oorin, and Indo. In vitro-grown apple shoots of susceptible cultivars were then treated with various doses of X- or γ-ray irradiation. Several mutants resistant to AM-toxin I were obtained by combining the techniques for tissue culture of apple shoots with the AM-toxin I screening method. Following a repeat second screening test with AM-toxin I, mutant plants were sprayed with a spore suspension of A. alternata and found resistant to be the fungal pathogen. These mutants showed normal phenotypic appearance, and so far, no difference has been observed between the original plants and mutants except for the susceptibility to Alternaria blotch. 相似文献

Spider mites are very small phytophagous organisms living on plant leaves. They possess a diverse array of life types. Such
diversity in life type has been supposed to be a result of coevolution between predator and prey, although there is little
concrete evidence to support this concept. In Yezonychus sapporensis inhabiting the leaves of a dwarf bamboo, Sasa senanensis, all quiescent stages including eggs appear on the tips of the host leaf hairs during the summer season, and this habit is
thought to be a kind of predator avoidance trait. To confirm this, the survival rates of Y. sapporensis eggs deposited on natural sites and experimentally manipulated sites were observed for eight co-occurring predator species.
The results clearly showed that the oviposition behavior of Y. sapporensis has a function in avoiding predator attack, at least under experimental conditions.

The effect of house building design and environment on the fungal movement in the houses of 41 bronchial asthma (BA) patients
has been investigated by examining house dust. The presence and composition of fungi were determined and compared in relation
to building structure, house age, size of living room, main flooring material, presence of a living-room rug or air purifier,
and frequency of vacuum cleaning. Among these elements, fungal CFU apparently varied only between building structure: wooden-board
houses had significantly higher numbers of fungi than reinforced concrete houses (p < 0.01), and wooden mortar or iron-framed
prefabricated houses had significantly higher numbers of fungi than reinforced concrete houses (p < 0.05). Classification
of the types of fungi present in the house dust of BA patients showed that, regardless of the building designs, there were
high levels of osmophilic fungi (group A) and fungi that survive at relatively dry conditions (group B), whereas fungi that
survive in very wet conditions (group D) were present at low frequency.

A simple and useful method for the detection of C. kutscheri from the oral cavity of living rats was devised. In 10 sacrificed rats from two naturally and subclinically infected conventional colonies, 10(4.28) or 10(3.84) CFU/ml C. kutscheri were isolated from upper incisor swab extractions, while 10(1.38) or 10(1.58) and < 10 or 10(1.56) CFU/ml from the upper soft palate and pharynx, respectively. In another survey with 26 living animals, which were reared on the same rack, organisms were detected from the upper incisor and gingival swabs in 15 of 26 rats (57.7%). The results were reproducible at a second survey 10 days later. No organisms were isolated from any sites of the orally negative rats. These results indicated that culture of swab specimens from the upper incisors and gingivae of incisors is useful for the detection of C. kutscheri infection in living rats. 相似文献

The full-length sense cDNA for sweet potato granule-bound starch synthase I (GBSSI) driven by the CaMV 35S promoter was introduced into the sweet potato by Agrobacterium tumefaciens-mediated transformation. Out of the 26 transgenic plants obtained, one plant showed the absence of amylose in the tuberous root as determined by the iodine colorimetric method. Electrophoresis analysis failed to detect the GBSSI protein, suggesting that gene silencing of the GBSSI gene occurred in the transgenic sweet potato plant. These results demonstrate that starch composition in the tuberous root of sweet potato can be altered by genetic transformation. 相似文献

The relationship between the degree of the expression of Cu/Zn SOD, GST-pi and bcl-2 in the initial and recurrent tumor tissue after radiotherapy and/or chemotherapy and the cellular heterogeneity obtained from DNA content by image cytometry was investigated. Subjects were 7 patients who had glial tumors which were surgically removed at onset and removed a second time at recurrence. Radiotherapy and chemotherapy were also administered after initial resection. Immunoreactivity for copper/zinc super oxide dismutase (Cu/Zn SOD), GST (glutathione-S-transferase)-pi, and bcl-2 were evaluated from routinely prepared tissue blocks. Tumors were classified into two groups by cytometric analysis of DNA ploidy in the G2M cell cycle phase. One tumor group consisted of single clonal cells in both the initial and recurrent tumors and the other group consisted of tumors with polyclonal cells in the initial and recurrent tumor. In this study, one patient (case 3) with single clonal cell glioblastoma at recurrence did not show high Cu/Zn SOD activity after radiotherapy and chemotherapy but showed a short survival time after recurrence. In three patients (cases 1, 2, 3) with single clonal-cell glioblastoma, the recurrent tumor cells showed high GST-pi immunoreactivity and survival time was short after recurrence. Tumor cells in two patients (cases 5, 7) with single clonal cell anaplastic glioma at recurrence, showed high GST-pi immunoreactivity and had a short survival time after recurrence. In three single clonal glioblastomas (cases 1, 2, 3), the recurrent tumor showed the increased bcl-2 immunoreactivity and showed a short survival time after recurrence. In two patients (case 5, 7) with single clonal cell anaplastic glioma at recurrence, tumor cells showed a high bcl-2 immunoreactivity and these patients showed a short survival time after recurrence. Although the number of subjects is very small, our study shows that the immunoreactivity of bcl-2 and GST-pi in malignant gliomas may be very important factors in radio- and chemosensitivity, and shows that GST-pi is induced by radiation and anti-cancer drugs. 相似文献

This study examined the effects of soybean beta-conglycinin, from which phytate was mostly removed, on the plasma lipids in young and adult rats. Male Wistar young (6 week-old) and adult (21 week-old) rats were fed high cholesterol diets containing 20% casein, soy protein isolate (SPI), or soybean beta-conglycinin for 10 days. In young rats, although the food intake of the beta-conglycinin group was higher than those of the casein and SPI groups, the weight gain was significantly lower than those of the other groups. However, in adult rats, the weight gain was not different among the groups. In young and adult rats, relative liver weights of SPI and beta-conglycinin groups were significantly lower than that of the casein group, and the degree of the reduction was more marked in the beta-conglycinin group than in the SPI group. In young rats, the plasma triglyceride level was significantly lower in the SPI and beta-conglycinin groups than that in the casein group. In addition, the plasma triglyceride level of the beta-conglycinin group was significantly lower than that of the SPI group. Plasma total cholesterol levels of the SPI and beta-conglycinin groups were significantly lower than that of the casein group. However, there was little difference in the lowering effect between SPI and beta-conglycinin. These results indicate that soybean beta-conglycinin may have lowering functions not only on plasma total cholesterol level, but also on plasma triglyceride level. 相似文献

Although the sequencing of the human genome is complete, identification of encoded genes and determination of their structures
remain a major challenge. In this report, we introduce a method that effectively uses full-length mouse cDNAs to complement
efforts in carrying out these difficult tasks. A total of 61,227 RIKEN mouse cDNAs (21,076 full-length and 40,151 EST sequences
containing certain redundancies) were aligned with the draft human sequences. We found 35,141 non-redundant genomic regions
that showed a significant alignment with the mouse cDNAs. We analyzed the structures and compositional properties of the regions
detected by the full-length cDNAs, including cross-species comparisons, and noted a systematic bias of GENSCAN against exons
of small size and/or low GC-content. Of the cDNAs locating the 35,141 genomic regions, 3,217 did not match any sequences of
the known human genes or ESTs. Among those 3,217 cDNAs, 1,141 did not show any significant similarity to any protein sequence
in the GenBank non-redundant protein database and thus are candidates for novel genes.
