Water-soluble neutral cosolutes can be used to quantify biomolecular properties in the particular molecular environment occurring in a cell. We studied the conformation and the thermal stability of DNA and RNA structures in the presence of PEG [poly(ethylene glycol)] and smaller cosolutes of glycerol, ethylene glycol, 1,3-propanediol, 2-methoxyethanol, and 1,2-dimethoxyethane. Although the neutral cosolutes destabilized the oligonucleotide duplex and the hairpin structures, the left-handed Z-form duplex was more energetically favored in the cosolute-containing solutions. These observations were due to the contribution of water molecule on the nucleotide structure formations because the cosolutes act as an osmolyte to reduce the water activity of a solution. Moreover, the sodium ion condensation for the duplex and the hairpin formations was reduced in the presence of PEG, while that for the transition from the B-form to the Z-form was unaltered. The CD (circular dichroism) and EPR (electron paramagnetic resonance) spectra demonstrated that the cosolutes changed the helical conformation of the unstructured oligonucleotides, but not those of the ordered structures. The results of the favorable formation of the noncanonical nucleotide structures, and minimized conformational and thermal perturbations of the ordered nucleotide structures in the cosolute-containing solutions implicate the significance of the intracellular environment on DNA and RNA structures in a cell. 相似文献

Background
The objective of this study is to examine the stress and mood changes of Japanese subjects over the 1–3 days before a migraine headache.Methods
The study participants were 16 patients with migraines who consented to participate in this study. Each subject kept a headache diary four times a day for two weeks. They evaluated the number of stressful events, daily hassles, domestic and non-domestic stress, anxiety, depressive tendency and irritability by visual analog scales. The days were classified into migraine days, pre-migraine days, buffer days and control days based on the intensity of the headaches and accompanying symptoms, and a comparative study was conducted for each factor on the migraine days, pre-migraine days and control days.Results
The stressful event value of pre-migraine days showed no significant difference compared to other days. The daily hassle value of pre-migraine days was the highest and was significantly higher than that of buffer days. In non-domestic stress, values on migraine days were significantly higher than on other days, and there was no significant difference between pre-migraine days and buffer days or between pre-migraine days and control days. There was no significant difference in the values of domestic stress between the categories. In non-domestic stress, values on migraine days were significantly higher than other days, and there was no significant difference between pre-migraine days and buffer days or between pre-migraine days and control days.There was little difference in sleep quality on migraine and pre-migraine days, but other psychological factors were higher on migraine days than on pre-migraine days.Conclusion
Psychosocial stress preceding the onset of migraines by several days was suggested to play an important role in the occurrence of migraines. However, stress 2–3 days before a migraine attack was not so high as it has been reported to be in the United States and Europe. There was no significant difference in the values of psychological factors between pre-migraine days and other days.993.

Mammalian or mechanistic target of rapamycin complex 1 (mTORC1) is a master regulator of cell growth, metabolism, and cell differentiation. Recent studies have revealed that the recruitment of mTORC1 to lysosomes is essential for its activation. The ceramide analogue 1-phenyl-2-decanoylamino-3-morpholino-1-propanol (PDMP), a well known glycosphingolipid synthesis inhibitor, also affects the structures and functions of various organelles, including lysosomes and endoplasmic reticulum (ER). We investigated whether PDMP regulates the mTORC1 activity through its effects on organellar behavior. PDMP induced the translocation of mTORC1 from late endosomes/lysosomes, leading to the dissociation of mTORC1 from its activator Rheb in MC3T3-E1 cells. Surprisingly, we found mTORC1 translocation to the ER upon PDMP treatment. This effect of PDMP was independent of its action as the inhibitor, since two stereoisomers of PDMP, with and without the inhibitor activity, showed essentially the same effect. We confirmed that PDMP inhibits the mTORC1 activity based on the decrease in the phosphorylation of ribosomal S6 kinase, a downstream target of mTORC1, and the increase in LC3 puncta, reflecting autophagosome formation. Furthermore, PDMP inhibited the mTORC1-dependent osteoblastic cell proliferation and differentiation of MC3T3-E1 cells. Accordingly, the present results reveal a novel mechanism of PDMP, which inhibits the mTORC1 activity by inducing the translocation of mTOR from lysosomes to the ER. 相似文献

Strawberry fruit contains many constituents, some of which have the potential to inhibit histidine decarboxylase (HDC) activity. HDC converts l-histidine to histamine, which is associated with allergic and other biological reactions in the human body. The HDC inhibition levels were different and the component ratios varied by genotype in strawberry. Among the 11 cultivars collected locally in Japan, ‘Tokun’ had an approximately ten times higher inhibition ratio than the lowest cultivar. The reproducibility was confirmed using five cultivars under the same conditions in a glass greenhouse, suggesting that genotypic variation is a major factor of HDC inhibition. The potential inhibitors of HDC might be polyphenols because they showed moderate correlations with HDC inhibition rates. Among the polyphenols, the anthocyanin content possessed a moderate negative correlation. Ascorbic acid, which contributes to the overestimation of total polyphenol, did not independently inhibit HDC activity. These findings will support the identification of potential HDC inhibitors in strawberry and indicated that genotypic differences would make useful probes for inhibitor identification. 相似文献

Japan is considered a global hot spot of biodiversity. With regard to species diversity, insects are no exception. To date, more than 32,000 insect species have been identified in Japan, while around 100,000 species of insects are estimated to inhabit this country. In this paper, we outline background factors having contributed to diversification of Japanese insects. Of course, the high degree of Japanese insect diversity is the result of many complex factors. In addition to the humid Asian monsoon climate and the extensive latitudinal gradient of habitats, the extremely complex geological history has contributed as an important factor to generate and maintain the high species diversity and endemism. In particular, the independent origins of northeastern and southwestern Japan from the Eurasian continent have greatly contributed to the diverse composition of Japanese insect fauna. To highlight the importance of this process, we introduce some case studies and previously published papers focusing on several insect groups with low dispersal ability. Those cases indicate that the geological history of Japan has played an important role in the differentiation of Japanese insect species. Besides such geological factors, climatic and ecological factors in combination have contributed to the formation of Japanese insect fauna in complicated ways and produced its particularly high degree of biodiversity. The knowledge compiled here will provide useful information for future studies aiming to understand more deeply the processes of speciation and faunal formation of Japanese insects. 相似文献

Rapid expansion of exotic bamboos has lowered species diversity in Japan's ecosystems by hampering native plant growth. The invasive potential of bamboo, facilitated by global warming, may also affect other countries with developing bamboo industries. We examined past (1975–1980) and recent (2012) distributions of major exotic bamboos (Phyllostachys edulis and P. bambusoides) in areas adjacent to 145 weather stations in central and northern Japan. Bamboo stands have been established at 17 sites along the latitudinal and altitudinal distributional limit during the last three decades. Ecological niche modeling indicated that temperature had a strong influence on bamboo distribution. Using mean annual temperature and sun radiation data, we reproduced bamboo distribution (accuracy = 0.93 and AUC (area under the receiver operating characteristic curve) = 0.92). These results infer that exotic bamboo distribution has shifted northward and upslope, in association with recent climate warming. Then, we simulated future climate data and projected the climate change impact on the potential habitat distribution of invasive bamboos under different temperature increases (i.e., 1.5°C, 2.0°C, 3.0°C, and 4.0°C) relative to the preindustrial period. Potential habitats in central and northern Japan were estimated to increase from 35% under the current climate (1980–2000) to 46%–48%, 51%–54%, 61%–67%, and 77%–83% under 1.5°C, 2.0°C, 3.0°C, and 4.0°C warming levels, respectively. These infer that the risk areas can increase by 1.3 times even under a 1.5°C scenario and expand by 2.3 times under a 4.0°C scenario. For sustainable ecosystem management, both mitigation and adaptation are necessary: bamboo planting must be carefully monitored in predicted potential habitats, which covers most of Japan. 相似文献

Postembryonic developmental stages of an endoparasitic pycnogonid, Ammothella biunguiculata in Izu Peninsula, Japan are described. Eleven stages were identified beginning with a protonymphon larva attached to the male oviger. We found endoparasitic individuals in the host actinian from the second to tenth instar, and forms in the ninth stage to adult were found free-living. This indicates a transition from being endoparasitic to free-living during the ninth to tenth instar stages. The first instar protonymphon attached to the adult male oviger has a gland duct on the anterior margin of each chelifore scape which completely disappears with the second instar. The disappearance of the chelifore gland duct coincides with the beginning of an endoparasitic stage in the development of this species. Although the larval morphology and the postembryonic development of pycnogonids have been summarized by several authors, the present study concludes that much remains to be learnt. 相似文献

Using structure-based drug design, we identified a novel series of 5,6-dihydroimidazolo[1,5-f]pteridine PLK1 inhibitors. Rational improvements to compounds of this class resulted in single-digit nanomolar enzyme and cellular activity against PLK1, and oral bioavailability. Copper(II) complexes of new N3O- and N2O2-donor tripodal ligands bearing one or two o-substituted phenol moieties have been synthesized as models for the galactose oxidase active site. The complexes of 2-[N-(1-methyl-2′-imidazolylmethyl)-N-(6″-methyl-2″-pyridylmethyl)-aminomethyl)]-4-methyl-6-methylthiophenol (MeSL), [Cu(MeSL)Cl], and N-(6-methyl-2-pyridylmethyl)-N,N-bis(2′-hydroxy-3′,5′-di-tert-butylbenzyl)amine (t-buL2mepy), [Cu(t-buL2mepy)(H2O)], have been revealed by X-ray structural analysis to have a square-pyramidal structure with one and two phenolate oxygens in the basal plane, respectively. [Cu(MeSL)Cl] was converted into a Cu(II)-o-methylthiophenoxyl radical species by electrochemical or Ce(IV) oxidation. An o-methoxyphenoxyl radical in a similar complex was considerably more stable than the 2,4-di(tert-butyl)phenoxyl radical. While t-buL2mepy reacted with Cu(ClO4)2 to give [Cu(t-buL2mepy)(H2O)] without disproportionation, an N2O2-donor ligand containing an o-methoxyphenol, a 2,4-di(tert-butyl)phenol, and an N-methylimidazole moiety gave a phenoxyl radical complex exhibiting the characteristic absorption peak at 478 nm as a reddish powder by the reaction with Cu(ClO4)2 as a result of spontaneous disproportionation. It exhibited a quasi-reversible redox wave at E1/2=0.34 V (vs. Ag/AgCl) in CH3CN, which is lower than the potentials of the copper complexes of various N3O-donor ligands, and oxidized ethanol to acetaldehyde with a low turnover number. 相似文献
