The regeneration and persistence of host- and donor-derived T cells were examined in the thymus as well as the spleen of mouse radiation bone marrow chimeras of two semiallogeneic combinations (F1----P, P----F1) with different Thy-1 markers on T cells of donor and host origins. An unexpectedly large number of host-type T cells were recovered from the spleens of F1----P chimeras, amounting to as high as 45 and 25% of total T cells at 6 and 14 weeks after bone marrow transplantation (BMT), respectively. To the contrary, the residual host-type T cells in the spleens of P----F1 chimeras disappeared quickly, resulting in less than 0.1% of total T cells at 6 weeks after BMT. It was also revealed that the number of host-type T cells in the spleens of F1----P chimeras decreased in proportion to increase of radiation dose given to the recipients.  相似文献

Summary The development of a line of epithelial cells derived from lung tissue of a 4-week old kitten (KL strain) with evidence regarding its chromosomal changes in vitro is described. The outgrowing cells from fragments in primary cultures in Eagle's medium plus 10% horse serum were scraped with a rubber policeman and dispersed with a syringe fitted with a No. 15 needle. The cell suspension was transferred into a T 30 flask. The floor of the T 30 flask was covered with avian plasma clot which was allowed to set for 10 minutes before adding the cell suspension. The appearance of the cells was epithelial-like at all stages of cultivation. The most frequent chromosome number in 6-day primary culture preparations was 38 (68%). Counts made from cells in the 4th, 9th and 33rd subcultures (64th, 83rd and 165th days from the date of primary culture) showed a spread of the chromosome numbers. In the latest observation, the 84th subculture (411th day), the most frequent chromosome numbers were 90 (22%) and 92 (26%). A total of two hundreds and seventy squirrel monkeys (Saimiri sciureus) of both sexes were imported from South America through an animal dealer of the U. S. A. in eight lots from May, 1973 to July, 1974. They were investigated for the presence of the parasites for a maximum of fifteen months including nine weeks of quarantine period from the time of import. The mortality rate before or immediately after arrival at our laboratory reached 56.7% in the first two lots. This, however, could be reduced later to 4.8% by improving some conditions of transportation. Bacteriological surveys revealed no shigella or salmonella infections during the quarantine period. No tuberculin positive reactors were also detected. Filarial infestations seemed to be common in this species; about 45.5% had adult worms in the peritoneal cavity and about 65% harbored microfilaria in the peripheral blood. Acanthocephla was also found in a high rate in the lower alimentary tracts. It has been suggested that the worms disappear in fifteen months by keeping animals in the intermediate host-free circumstances. The sarcocyst was found in the femoral muscle of 7 (18.9%) of 37 animals examined histologically.  相似文献

A simple bioassay based on the inhibition by abscisic acid (ABA) of cucumber (Cucumis sativus L., cv. National Pickling) hypocotyl elongation was developed. Sections of 3-day-old dark-grown cucumber hypocotyl taken from 0–5 mm immediately below the cotyledon were used for the assay. A dark incubation period of 20 h was followed by an exposure to light for 24 h. Under these conditions, the inhibition of hypocotyl elongation is proportional to the abscisic acid applied. The minimum detectable level of abscisic acid was 10–9 M, and the range of linear response to abscisic acid was between 10–7 and 10–3 M. This assay is 10 times more sensitive than the cucumber cotyledon greening bioassay for abscisic acid.  相似文献
