Tumor suppressor p53 plays important roles in cell cycle regulation, apoptosis and DNA repair in different cell types including lung cancer. There are different p53 apoptotic pathways in high and low metastatic ability lung cancer cells. However, the exactly mechanism in the pathway is still unclear. Here we found that Annexin A2, a Ca2+-dependent phospholipid-binding protein, is involved in p53-mediated apoptosis. First, by using mRNA differential display technique, down-regulated Annexin A2 expression was found in all cell lines transfected of Ad-p53 (adenoviral expression construct encoding wild type p53 gene) especially in highly metastatic Anip973 lung cancer cells. Then, decreased expression of Annexin A2 was further confirmed by Northern blot and Western blot analysis. At last, knock down of Annexin A2 by siRNA inhibited cellular proliferation in BE1 cell line with highly metastatic ability. Taken together, our results suggested that Annexin A2 may play roles in p53 induced apoptosis and it is also involved in regulation of cell proliferation. This work develops a simple, sensitive and signal-on electrochemical sensor for methyltransferase (MTase) activity analysis. The sensor is composed of a methylene blue-modi?ed "signaling DNA probe" and a "capture DNA probe" tethered methylation-responsive hairpin DNA (hairpin-capture DNA probe). The thiol- modified hairpin-capture DNA probe at 5' end was firstly self-assembled on gold electrode via Au-S bonding. Methylation-induced scission of hairpin-capture DNA probe would displace the hairpin section and remain the "capture DNA probe" section on the gold electrode. Subsequently, the remained "capture DNA probe" on the gold electrode can hybridize with the methylene blue-modi?ed "signaling DNA probe", mediating methylene blue onto the gold electrode surface to generate redox current. It was eT on state. The developed facile signal-on electrochemical sensing system showed a linear response to concentration of Dam MTase range from 0.1 to 1.0 U/mL. The detection limit of Dam MTase activity was determined to be 0.07 U/mL and the total detection time is 7h. The sensor also has the ability to provide information about the dynamics of methylation process. Furthermore, we demonstrated that this sensor could be utilized to screen inhibitors or drugs for Dam MTase.  相似文献

Evodiamine, the major bioactive compound isolated from Chinese herbal drug named Wu-Chu-Yu, has been reported to exhibit anti-tumor growth and metastasis. However, the effect of evodiamine on angiogenesis remains to be investigated. We used the fresh medium containing evodiamine or human lung adenocarcinoma cell (CL1 cells) derived conditioned media free of evodiamine to test their capability to induce in vitro angiogenesis, i.e., human umbilical vein endothelial cells (HUVECs) tube formation and invasion. We demonstrated that evodiamine could directly inhibit in vitro HUVECs tube formation and invasion. Locally administered evodiamine also inhibited the in vivo angiogenesis in the chick embryo chorioallantoic membrane (CAM) assay. The gene expression of vascular endothelial growth factor (VEGF) and the p44/p42 mitogen-activated protein kinase (MAPK, ERK) that correlated with endothelial cells angiogenesis were inhibited by evodiamine. We found that the evodiamine-treated CL1 cells derived conditioned medium showed decreased VEGF release and reduced ability of inducing in vitro tube formation. After the collection of conditioned media, the VEGF expression of remaining CL1 cells were determined by Western analyses and revealed that evodiamine decreased VEGF expression. Moreover, administration of recombinant human VEGF(165) (rhVEGF(165)) induced tube formation and ERK phosphorylation by HUVECs, and partially attenuated inhibitory effect of evodiamine. From these results, we suggested that evodiamine is a potent inhibitor of angiogenesis. The mechanism might involve at least the inhibition of VEGF expression, probably through repression of ERK phosphorylation.  相似文献

Choline is an essential nutrient for cell survival and proliferation, however, the expression and function of choline transporters have not been well identified in cancer. In this study, we detected the mRNA and protein expression of organic cation transporter OCT3, carnitine/cation transporters OCTN 1 and OCTN2, and choline transporter-like protein CTL1 in human lung adenocarcinoma cell lines A549, H 1299 and SPC-A-1. Their expression pattern was further confirmed in 25 human primary adenocarcinoma tissues. The choline uptake in these cell lines was significantly blocked by CTL1 inhibitor, but only partially inhibited by OCT or OCTN inhibitors. The efficacy of these inhibitors on cell proliferation is closely correlated with their abilities to block choline transport. Under the native expression of these transporters, the total choline uptake was notably blocked by specific PI3K/AKT inhibitors. These results describe the expression of choline transporters and their relevant function in cell proliferation of human lung adenocarcinoma, thus providing a potential "choline-starvation" strategy of cancer interference through targeting choline transporters, especially CTL1.  相似文献

近年来, 生物多样性监测网络的建设得到广泛重视, 全球、地区或国家生物多样性观测网不断组建。生物多样性观测的理论框架得到发展, 提出了生物多样性核心监测指标(Essential Biodiversity Variables, EBV)。鱼类多样性监测的理论框架包含于生物多样性核心监测指标之内, 在遗传、物种、生态系统等多层次进行。基于鱼类监测提出的生物完整性指数(index of biotic integrity, IBI)强调不同物种的生态功能, 可以综合反映群落结构和功能的变化, 得到广泛应用。鱼类多样性的监测方法是传统网具和现代水声学等方法的结合。监测结果的分析可以进行简单的指数比较, 也可以进行长期的趋势分析, 寻找关键节点, 探讨宏观生态格局的变化。中国内陆水体鱼类多样性监测网隶属于中国生物多样性监测与研究网络, 拟选取长江、黄河、黑龙江、珠江、澜沧江、怒江、塔里木河及青海湖8大流域, 对25个重要区域和24个重点物种(类群)进行监测, 从重要区域鱼类群落结构、重点物种(类群)种群动态和个体生物学特征、遗传多样性、早期资源等不同层次, 全面监测我国内陆水体鱼类生物多样性状况。  相似文献

Excessive proteolysis of fibronectin (FN) impairs tissue repair in chronic wounds. Since FN is essential in wound healing, our goal is to improve its proteolytic stability and at the same time preserve its biological activity. We have previously shown that reduced FN conjugated with polyethylene glycol (PEG) at cysteine residues is more proteolytically stable than native FN. Cysteine‐PEGylated FN supported cell adhesion and migration to the same extent as native FN. However, unlike native FN, cysteine‐PEGylated FN was not assembled into an extracellular matrix (ECM) when immobilized. Here, we present an alternative approach in which FN is preferentially PEGylated at lysine residues using different molecular weight PEGs. We show that lysine PEGylation does not perturb FN secondary structure. PEG molecular weight, from 2 to 10 kDa, positively correlates with FN–PEG proteolytic stability. Cell adhesion, cell spreading, and gelatin binding decrease with increasing molecular weight of PEG. The 2‐kDa FN–PEG conjugate shows comparable cell adhesion to native FN and binds gelatin. Moreover, immobilized FN–PEG is assembled into ECM fibrils. In summary, lysine PEGylation of FN can be used to stabilize FN against proteolytic degradation with minimal perturbation to FN structure and retained biological activity.  相似文献

在研究太湖梅梁湾水体中的悬浮质及水体中光谱分布时发现:总悬浮质中无机悬浮质含量与总悬浮质之间呈现明显的正比线形关系(R2=0.9724),而有机悬浮质与总悬浮质之间的正比线形关系较弱(R2=0.2921);在20cm处以下,总悬浮质中的藻类含量随着水深度的增加呈现下降的趋势,表面略低;有机悬浮质与叶绿素a的含量之间的线形关系较弱(R2=0.1344);光谱的分布特征是:在水体的表面,绿光最强,蓝光较强,而红光最弱;在150cm以下,红光最强,绿光较强,蓝光最弱。  相似文献

基于我国南方双季稻区20年长期田间定位施肥试验,研究了不同氮磷钾施肥方式对水稻碳、氮积累与分配的影响.结果表明:偏施氮肥处理水稻籽实的碳、氮含量最高,分别达到433和18.9 g·kg-1.水稻植株的碳、氮储量以氮磷钾平衡施肥(NPK)及氮磷钾基础上有机物料循环施肥处理(NPKC)最高,其中NPKC和NPK处理籽实碳储量分别为2015和1960kg hm-2,茎叶碳储量分别为2048和2002 kg·hm-2;籽实氮储量分别为80.6和80.5kg·hm-2,茎叶氮储量则以NPK处理最高,为59.3 kg·hm-2.有机无机肥的配合施用显著增加了水稻植株体内碳和氮的累积;与偏施氮肥处理相比,氮磷钾的综合施用更利于水稻生长过程中碳、氮的累积与分配.  相似文献
