Fifty-two endophytic fungi strains with different colony morphologies were isolated from stems, leaves and roots of Huperzia serrata (Thunb. ex Murray) Trevis. collected from Bawangling Reserve of Hainan Province in southern China. They were identified mainly
based on rDNA ITS sequences and phylogenetic analysis. The results showed that all strains belonged to four classes, i.e.
Sordariomycetes (92.31%), Dothideomycetes (3.85%), Pezizomycetes (1.92%) and Agaricomycetes (1.92%). Forty-seven strains were identified at the genus level, including Glomerella (Colletotrichum), Hypocrea (Trichoderma), Pleurostoma, Chaetomium, Coniochaeta (Lecythophora), Daldinia, Xylaria, Hypoxylon,
Nodulisporium, Cazia and Phellinus. As to the other five strains, three were identified at the order level and two at the family level, indicating that a great
diversity of fungi taxa exists in H. serrata. Most isolated strains belonged to the genus of Glomerella (Colletotrichum) and Hypoxylon, twenty-one from Glomerella and its anamorph Colletotrichum (42.3% of total isolated strains) and ten from Hypoxylon (19.2% of total isolated strains). Free fatty acids (FFA)-induced proliferation and apoptosis was studied in human umbilical vein endothelial cells (HUVECs). A recombinant adenovirus containing a RNAi cassette targeting the GSK-3β gene was produced and its silencing effect on GSK-3β gene was detected by Western blot analysis and immunohistochemistry assay in HUVECs. The effect of the RNAi on the protein level of β-catenin was explored by transfecting the RNAi adenovirus to inhibit the expression of GSK-3β protein. The subsequent effect on the Wnt/GSK-3β/β-catenin signal pathway and on proliferation and apoptosis of HUVECs cultured with FFAs, was analyzed by BrdU assay, Annexin V-FITC/PI Apoptosis Detection Kit, and 4′,6-diamidino-2- phenylindole(DAPI) to explore the possible connection between the signaling pathway and FFA-induced proliferation and apoptosis. The Western blot results showed that the expression of GSK-3β protein in HUVECs could be inhibited efficiently by the RNAi adenovirus, and that the protein level of β-catenin was increased by RNAi adenovirus transfection. The results of the BrdU assay suggested that knockdown of GSK-3β with the RNAi adenovirus may stimulate the proliferation of HUVECs. Apoptosis was observed in HUVECs exposed to FFAs (0.75 mmol/L) for 72 h, and this effect could be partly reversed when interfering with the RNAi adenovirus. It may be concluded that the RNAi adenovirus specific to GSK-3β may partly protect HUVECs from apoptosis induced by FFAs, probably through the up-regulation of the Wnt/β-catenin signal pathway. 相似文献

Background and aims
The photochemical reflectance index (PRI) is correlated to photosynthetic efficiency and has been successfully applied at multiple scales for remote estimation of physiological functioning. However, interpretation of the PRI signal can be confounded by many different variables including declines in photochemical pigments. Our study was aimed at investigating PRI in response to salinity stress, and evaluating physiological and pigment responses of two co-occurring shrubs, Baccharis halimifolia and Myrica cerifera in laboratory studies.Methods
Photosynthesis, water relations, chlorophyll fluorescence, hyperspectral reflectance and leaf pigment contents were measured following salinity treatment.Results
Physiological measurements indicated that both species exhibit adaptations which protect PSII during periods of stress. Chlorophyll fluorescence parameters were affected in both species, but indicated that other photochemical reactions (e.g. photorespiration) were important for energy dissipation in absence of chlorophyll changes. After many days of reduced photosynthesis, photochemical changes were detectable using PRI indicating chronic stress.Conclusions
Variations in PRI were not related to changes in pigments but strongly related to tissue chlorides indicating salinity effects on the PRI signal. Thus, PRI is an indicator of salinity stress in these coastal species and may be as an early signal for increasing salt exposure associated with rising sea-level and climate change. 相似文献974.

The phase transitions in fully hydrated dipalmitoylphosphatidylcholine (DPPC) and DPPC/water/ethanol phases have been studied by lowangle time-resolved x-ray diffraction under conditions similar to those employed in calorimetry (scan rates 0.05-0.5°C/min and uniform temperature throughout the samples). This approach provides more adequate characterization of the equilibrium transition pathways and allows for close correlations between structural and thermodynamic data. No coexistence of the rippled gel (Pβ') and liquid-crystalline (Lα) phases was found in the main transition of DPPC; rather, a loss of correlation in the lamellar structure, observed as broadening of the lamellar reflections, takes place in a narrow temperature range of ~100 mK at the transition midpoint. Formation of a long-living metastable phase, denoted by Pβ'(mst), differing from the initial Pβ' was observed in cooling direction by both x-ray diffraction and calorimetry. No direct conversion of Pβ'(mst) into Pβ' occurs for over 24 h but only by way of the phase sequence Pβ'(mst) → Lβ' → Pβ'. According to differential scanning calorimetry (DSC), the enthalpy of the Pβ'(mst)-Lα transition is by ~5% lower than that of the Pβ'-Lα transition. The effects of ethanol (Rowe, E. S. 1983. Biochemistry. 22:3299-3305; Simon, S. A., and T. J. McIntosh. 1984. Biochim. Biophys. Acta 773:169-172) on the mechanism and reversibility of the DPPC main transition were clearly visualized. At ethanol concentrations inducing formation of interdigitated gel phase, the main transition proceeds through a coexistence of the initial and final phases over a finite temperature range. During the subtransition in DPPC recorded at scan rate 0.3°C/min, a smooth monotonic increase of the lamellar spacing from its subgel (Lc) to its gel (Lβ') phase value takes place. The width of the lamellar reflections remains unchanged during this transformation. This provides grounds to propose a “sequential” relaxation mechanism for the subgel-gel transition which is not accompanied by growth of domains of the final phase within the initial one. 相似文献

The general aim of this study is to update the inventory of the fish species and to specify distribution patterns in the Nero River ichthyofauna in order to establish some basis for the conservation of these fish communities and their habitat. From February 2009 to January 2010, thirty-three sites were sampled monthly with gill nets and a backpack electrofisher, and environmental variables were recorded. Overall, 46 species included in 33 genuses, 24 families and 9 orders were collected. Eleven families and 30 species were the first records for the Nero River. Including all species previously listed in the literature, the number of species presently known in the Nero River and its tributaries is revised to 59. Four families, Alestidae (21%), Schilbeidae (19%), Cyprinidae (17%) and Cichlidae (16%) that made up 73% of the total number of the catches, were the most dominant. The most dominant numerical species were Schilbe mandibularis and Brycinus longipinnis. Fish species and sampling sites along with eight environmental variables were ordinated with canonical correspondence analysis (CCA) coupled to the Monte Carlo test. Ecological status based on fish assemblage according to environmental variables and anthropogenic pressures showed that miss dead wood leaves and roots, electrical conductivity, total dissolved solids, mud, nitrite, basin width, dissolved oxygen and pH, were the primary factors influencing fish distribution. The environmental tolerance index (ETI), ecological tolerance (tk) and optima (uk) values of 10 species to 8 different environmental variables were analyzed. Six species (Hemichromis fasciatus, Epiplatys chaperi, Barbus ablabes, B. longipinnis, Hemichromis bimaculatus and Chromidotilapia guntheri) have high ETI and a cosmopolitan distribution in the Nero River. In the tributaries of the middle course, high concentrations of nitrite in the water, added to the presence of a lot of tolerant species in the ichthyofauna are indications of disturbance of these areas. MicroRNAs are involved in cancer-related processes. The microRNA-21(miR-21) has been identified as the only miRNA over-expressed in a wide variety of cancers, including cervical cancer. However, the function of miR-21 is unknown in cervical carcinomas. In this study, we found that the inhibition of miR-21 in HeLa cervical cancer cells caused profound suppression of cell proliferation, and up-regulated the expression of the tumor suppressor gene PDCD4. We also provide direct evidence that PDCD4-3′UTR is a functional target of miR-21 and that the 18 bp putative target site can function as the sole regulatory element in HeLa cells. These results suggest that miR-21 may play an oncogenic role in the cellular processes of cervical cancer and may serve as a target for effective therapies. 相似文献

Epidermal growth factor receptor (EGFR/HER1/c-ErbB1), is overexpressed in many solid cancers, such as epidermoid carcinomas, malignant gliomas, etc. EGFR plays roles in proliferation, invasion, angiogenesis and metastasis of malignant cancer cells and is the ideal antigen for clinical applications in cancer detection, imaging and therapy. Aptamers, the output of the systematic evolution of ligands by exponential enrichment (SELEX), are DNA/RNA oligonucleotides which can bind protein and other substances with specificity. RNA aptamers are undesirable due to their instability and high cost of production. Conversely, DNA aptamers have aroused researcher’s attention because they are easily synthesized, stable, selective, have high binding affinity and are cost-effective to produce. In this study, we have successfully identified DNA aptamers with high binding affinity and selectivity to EGFR. The aptamer named TuTu22 with Kd 56 ± 7.3 nM was chosen from the identified DNA aptamers for further study. Flow cytometry analysis results indicated that the TuTu22 aptamer was able to specifically recognize a variety of cancer cells expressing EGFR but did not bind to the EGFR-negative cells. With all of the aforementioned advantages, the DNA aptamers reported here against cancer biomarker EGFR will facilitate the development of novel targeted cancer detection, imaging and therapy. 相似文献
