Exocyclic DNA adducts are mutagenic lesions that can be formed by both exogenous and endogenous mutagens/carcinogens. These adducts are structurally analogs but can differ in certain features such as ring size, conjugation, planarity and substitution. Although the information on the biological role of the repair activities for these adducts is largely unknown, considerable progress has been made on their reaction mechanisms, substrate specificities and kinetic properties that are affected by adduct structures. At least four different mechanisms appear to have evolved for the removal of specific exocyclic adducts. These include base excision repair, nucleotide excision repair, mismatch repair, and AP endonuclease-mediated repair. This overview highlights the recent progress in such areas with emphasis on structure-activity relationships. It is also apparent that more information is needed for a better understanding of the biological and structural implications of exocyclic adducts and their repair.  相似文献

实验用免疫细胞化学技术观察了大鼠鞘内分别注入蛋白激酶(PKC)抑制剂Chelerythrine(Chel)、纳洛酮(Nal)、或二者同时注入后,由后脚掌注射福尔马林引起的脊髓腰膨大背角中c-fos蛋白样免疫活性(Fos-LI)神经元数目的改变。结果发现:(1)鞘内注入Chel可显著降低福尔马林注射侧脊髓背角中Fos-LI神经元的数目,同空白对照组(鞘内注入生理盐水或10%的DMSO)相比,降低60.3%(P<0.001):(2)鞘内注入Nal后,福尔马林注射侧背角中Fos-LI神经元显著增加,同对照组相比,增加46.0%(P<0.01),而以背角深层增加最为明显;(3)在鞘内同时注入Chel和Nal后,与单独注入Nal组相比,脊髓背角中Fos-LI神经元的数目显著降低(降低53.2%),此数值与上述单独注入Chel时引起Fos-LI神经元降低的百分率近似。结果提示:(1)PKC只参与脊髓背角中部分Fos-LI神经元中c-fos蛋白的表达;(2)PKC可能不参与背角中同时激活的μ-(以及部分δ-)阿片受体对脊髓伤害性感受的调制。  相似文献

The structure-activity relationship of a number of synthetic green tea polyphenol analogs involving modifications of A ring and B ring of epi-gallocatechin gallate (EGCG) as proteasome inhibitors has been examined. It was found that in B ring, a decrease in the number of OH groups led to decreased potency. Introduction of a hydrophobic benzyl group into the 8 position of A ring did not significantly affect the proteasome-inhibitory potency.  相似文献

4-1BBL (TNFSF9) is a member of the tumor necrosis factor (TNF) ligand superfamily, which is expressed on some activated antigen presenting cells and B cells. We isolated a rat cDNA clone encoding the rat homologue of the human 4-1BBL (GenBank accession No. AY259541). The deduced rat 4-1BBL protein, consisting of 308 amino acids with a molecular weight of 33,469 Da, was a typical type II transmembrane glycoprotein, the same as human and murine 4-1BBL. “SDAA” in the cytoplasmic domain of rat 4-1BBL was deduced to act as the phosphorylation site for casein kinase I (“SXXS” motif), which is present in the cytoplasmic domains of human and murine 4-1BBL, and all other TNF ligand family members known to utilize reverse signaling. The two introns of 4-1BBL were also cloned (GenBank accession No. AY332409). Rat 4-1BBL is much more homologous with murine 4-1BBL than with human 4-1BBL, in both nucleotide and amino acid sequences. Rat 4-1BBL was expressed in all tested tissues: brain, lung, colon, liver, thymus, testicle, kidney, adrenal, stomach, spleen and heart. The chromosomal location of rat 4-1BBL was first identified by bioinformatics, then by fluorescence in situ hybridization at 9q11 (GenBank accession UniGene No. Rn.46783). Rat, murine and human 4-1BBL genes are evolved from a common gene. The identification and characterization of the rat counterpart of human 4-1BBL will facilitate studies of the biological function of this molecule.  相似文献

Mesenchymal stem cells (MSCs) have been shown to play therapeutic effect in traumatic brain injury (TBI). To augment the therapeutic effect, MSCs could be engineered to over‐express genes that are beneficial for treatment. In the present study, we over‐expressed hypoxia inducible factor (HIF)‐1alpha in bone marrow derived MSCs (BM‐MSCs) and sought to investigate whether HIF‐1alpha could enhance the therapeutic effect of MSCs in a mouse model of TBI. Balb/c mice were subjected to controlled cortical impact injury and MSCs were transplanted intravenously at 6 h after injury. The lesion volume and brain water content were measured and the neurological function was assessed by modified neurologic severity score tests. Double‐labeled immunofluorescence for BrdU and NeuU was performed to determine angiogenesis and neurogenesis. The expression of erythropoietin (EPO) and vascular endothelial growth factor (VEGF) was measured by quantitative RT‐PCR and western blotting. After TBI, mice received BM‐MSCs over‐expressing HIF‐1alpha showed significantly more functional recovery, reduced brain damage, increased angiogenesis and neurogenesis and increased expression of VEGF and EPO, compared with control mice or mice treated with non‐transduced BM‐MSCs. Over‐expression of HIF‐1alpha enhanced BM‐MSCs induced improvement of neurological recovery after TBI, by stimulating angiogenesis and neurogenesis.  相似文献

As landslides caused multiple casualties and destructions on a global scale, various models were applied to landslide susceptibility evaluation. In this study, a probability statistic model called the certain factor was adopted to assess the landslide susceptibility of Danba, a county in southwestern China, where the landslide events occur frequently but were poorly understood in regional landslide susceptibility. With the validation of area under the prediction rate curve, the resulting susceptibility map has the accuracies of 0.8211 and 0.8288 in experiment area and verification area, respectively. The validated assessment result was further processed to identify landslide-prone areas with the aid of the spatial clustering analysis of geographic information system. Two clustering indexes including Moran's I statistic and local indicator of spatial association (LISA) were involved. The Moran's I index of 0.959 and the LISA identification result accordant with previous investigations proved that the proposed method was rational and efficient to find the landslide-prone regions and make relevant decisions.  相似文献

A series of 4-chloro-2H-thiochromenes featuring nitrogen-containing side chains were designed, synthesized and tested in vitro for their antifungal activities. The results of preliminary antifungal tests showed that most target compounds exhibited good inhibitory activities against Candida albicans, Cryptococcus neoformans, Candida tropicalis. Notably, compounds 10e and 10y showed most potent activity in vitro against a variety of fungal pathogens with low MICs. Meanwhile, low cytotoxicity on mammalian cells has been observed for compounds 10e and 10y in the tested concentrations by the MTT assay. Therefore, the 4-chloro-2H-thiochromenes with nitrogen-containing groups provide new lead structures in the search for novel antifungal agents.  相似文献
