探索华山松(Pinus armandii)体细胞胚胎发生技术对其实施规模化无性繁殖和开展遗传转化具有重要意义。本文以1/2LM为基本培养基, 通过激素调节等措施对华山松的胚性愈伤组织诱导和幼胚的离体培养技术进行了初步研究。研究结果: 胚性愈伤组织诱导率最高可达52.71%, 但愈伤组织继代培养后没有体细胞胚胎的分化; 首次从其子叶期的幼胚中直接诱导出具有根和茎的完整植株, 诱导率达92%以上。文章确认了采集的幼胚发育状态对胚性愈伤组织的诱导有重要影响, 并对诱导的培养条件等进行了探讨。 相似文献

目的:调查哈尔滨市盲校生目前致盲情况及致盲性原因。方法:应用眼科常规方法时99名盲校学生(去除无眼球者共196眼)进行全面检查。时致盲原因进行分析。结果:致盲性眼病中居第一位为先天性白内障手术后仍未脱盲者,未脱盲原因包括:术后严重并发症、手术时机太晚形成严重弱视者,共45眼(占22.96%);居第二位为视网膜、黄斑变性,共32眼(占16.33%);居第三位为视神经萎缩,共24眼(占12.25%)。结论:本次对哈尔滨市盲校学生致盲原因调查表明,第一位为先天白内障术后由于合并症及并发症仍未脱盲者;第二位为为视网膜、黄斑变性;第三位为视神经萎缩。提示眼病的早期诊断和得当治疗的重要性。 相似文献

AFLP analysis of genetic diversity and relationship among some Chinese domestic ducks and wild ducks
The amplified fragment length polymorphic(AFLP)technique was used to analyze the genome DNA polymorphism among 8 breeds of domestic ducks and 2 species of wild ducks.Nine of the 17 selected primers pairs gave reproducible polymorphic DNA amplification bands.The amplified bands ranged from 44 to 83 per primer pair.Of the 513 AFLP markers obtained.498 were polymorphic.The proportion of polymorphic loci was 97.1%.The genetic distance(D)and similarity coefficients(GS)were calculated based on the polymorphic data.Between domestic ducks D ranged from 0.331 to 0.589,while between domestic ducks and the wild ducks,it ranged from 0.298 to 0.520(vs.Anas Platyrhynchos)and from 0.316 to 0.522(vs.A.Poecilorhyncha),respectively.The variance analysis showed no significant difference between the two groups of data,which indicated that both mallard and spot-billed ducks made contributions to domestic duck evolution.A dendrogram was constructed according to the D value. 相似文献

The Luoyixi section, exposed in a roadcut along the Youshui River (Fengtan Reservoir), Guzhang County, Hunan Province, China, is proposed as the stratotype for the base of an unnamed stage boundary (base of the Cambrian stage provisionally termed Stage 7). The proposed position of the GSSP is 121.3 m above the base of the Huaqiao Formation, at a horizon coinciding with the first appearance of the cosmopolitan agnostoid trilobite Lejopyge laevigata. The section fulfills all the requirements for a GSSP, and the horizon can be constrained not only with the primary stratigraphic marker (L. laevigata) but also with secondary biostratigraphic, sequence-stratigraphic, and chemostratigraphic correlation tools. The first appearance of L. laevigata is one of the most readily recognizable levels in the Cambrian, and can be correlated with precision to all paleocontinents. 相似文献

利用质粒pET-22b( )为表达载体,成功构建了高效表达N-乙酰鸟氨酸脱乙酰基酶的基因工程菌BL21-pET-22b( )-argE。研究了该菌的最适超声破壁条件及硫酸铵分级沉淀的最适范围,并以金属螯合亲和层析法纯化含有6-His-Tag的目的蛋白。结果表明:26℃诱导表达的菌体,最佳超声破碎条件为功率200 W,超声时间为15min;目的蛋白主要存在于40%~50%硫酸铵沉淀中。通过Ni-NTA亲和层析柱纯化目的蛋白,可以达到SDS-PAGE电泳纯,并显示单亚基相对分子质量为43 000。纯化倍数为139倍,回收率为11.1%。 相似文献

Various ionic and molecular activities in the extracellular environment are vital to plant cell physiological processes. A noninvasive microsensing system (NMS) based on either the scanning ion-selective electrode technique (SIET) or the scanning polarographic electrode technique (SPET) is able to obtain information regarding the transportation of various ions/molecules in intact samples under normal physiological conditions. The two-probe simultaneous test system (2STS) is an integrated system composed of SIET,SPET, and a Xu-Kunkel sampling protocol. In the present study, 2STS was able to simultaneously measure fluxes of H+ and O2 of the lily (Lilium Iongiflorum Thunb. cv. Ace) pollen tube while avoiding interference between the two probes. The results indicate that the proton fluxes were effluxes, whereas the oxygen fluxes were influxes, and they were closely correlated to each other surrounding the constitutive alkaline band region. Specifically, when the proton effluxes increased, the oxygen influxes also increased. Therefore,the hypothesis of condensed active mitochondria existing in the alkalized area of the pollen tube proposed by Hepler's group is supported. 相似文献

1,4-β-D-glucan cellobiohydrolase Ⅰ (CBH Ⅰ), p-nitrophenyl β-D-cellobioside, p-nitrophenol and cellobiose show distinct ultraviolet spectra, allowing the design of an assay to track the dynamic process of p-nitrophenyl β-D-cellobioside hydrolysis by CBH Ⅰ. Based on the linear relationship between p-nitrophenol formation in the hydrolysate and its first derivative absorption curve of AUC340-400 m (area under the curve), a new sensitive assay for the determination of CBH Ⅰ activity was developed. The dynamic parameters of catalysis reaction, such as Vm and kcat, can all be derived from this result. Metallothioneins (MTs) are involved in the cellular metabolism of zinc and in cytoprotection against stress factors. Hippocampus plays a specific role in the body's response to stressors. The present study was conducted to evaluate the effects of zinc on the expression of metallothionein isoforms in the hippocampus of stress rats. The animal model of psychologic stress was developed by restraint for 4 weeks. Wistar rats were randomly assigned to 6 groups: control group, zinc-deficient group, zinc-supplemented group, and the corresponding 3 stress groups. Three separate diets of different zinc contents (1.73 ppm, 17.7 ppm, and 41.4 ppm, respectively) were used in this study. Compared with the control group, the stress groups had higher inductions of MTs and MT-1 and MT-3 mRNA in hippocampus. On the one hand, the expressions of MTs and their mRNAs in hippocampus were downregulated in the zinc-deficient group; however, their expressions were evidently enhanced in the stress zinc-deficient group. MT induction in the zinc-supplemented group was increased. Furthermore, the stress zinc-supplemented group had a more significant yield of MTs and their mRNAs. In addition, the levels of plasma cortisol, interleukin-6 (IL-6), IL-1, and nitric oxide (NO) were increased clearly in the zinc-deficient group and the stress groups. The results suggest that zinc deficiency may decrease and zinc supplementation may increase the expressions of MTs and their mRNAs in hippocampus; moreover, stress can increase their expressions dramatically. The impairment of stress on the body may be involved with the nutrition status of zinc, and zinc deficiency can lower the body's adaptability to stress. 相似文献
