Introduction
Pseudomonas aeruginosa and Acinetobacter spp. are found to be associated with biofilm and metallo-β-lactamase production and are the common causes of serious infections mainly in hospitalized patients. So, the main aims of this study were to determine the rates of biofilm production and metallo beta-lactamase production (MBL) among the strains of Pseudomonas aeruginosa and Acinetobacter spp. isolated from hospitalized patients.Methods
A total of 85 P. aeruginosa isolates and 50 Acinetobacter spp. isolates isolated from different clinical specimens from patients admitted to Shree Birendra Hospital, Kathmandu, Nepal from July 2013 to May 2014 were included in this study. The bacterial isolates were identified with the help of biochemical tests. Modified Kirby-Bauer disc diffusion technique was used for antimicrobial susceptibility testing. Combined disc diffusion technique was used for the detection of MBL production, while Congo red agar method and tube adherence method were used for detection of biofilm production.Results
Around 16.4% of P. aeruginosa isolates and 22% of the strains of Acinetobacter spp. were metallo β-lactamase producers. Out of 85 P. aeruginosa isolates, 23 (27.05%) were biofilm producers according to tube adherence test while, only 13 (15.29%) were biofilm producers as per Congo red agar method. Similarly, out of 50 Acinetobacter spp. 7 (14%) isolates were biofilm producers on the basis of tube adherence test, while only 5 (10%) were positive for biofilm production by Congo red agar method. Highest rates of susceptibility of P. aeruginosa as well as Acinetobacter spp. were seen toward colistin.Conclusion
In our study, biofilm production and metallo beta-lactamase production were observed among Pseudomonas aeruginosa and Acinetobacter spp. However, no statistically significant association could be established between biofilm production and metallo beta-lactamase production.103.

The milk samples from buffaloes of Murrah breed at mid lactation stage, reared at an organised dairy farm, were screened for subclinical mastitis based on bacteriological examination and somatic cell count following International Dairy Federation criteria. Milk samples from subclinical mastitis infected and healthy buffaloes were analysed to evaluate physicochemical alterations in terms of protein, fat, pH, electrical conductivity, chloride, minerals (sodium, potassium and calcium) and trace elements (iron, zinc, copper and selenium). In the present study, protein, fat, zinc, iron, calcium and selenium content was significantly lower (P < 0.001), while pH and electrical conductivity were significantly higher in mastitic milk as compared to normal milk. Concentration of electrolytes mainly sodium and chloride significantly increased with higher somatic cell count in mastitic milk and to maintain osmolality; potassium levels decreased proportionately. Correlation matrix revealed significantly positive interdependences of somatic cell count with pH, electrical conductivity, sodium and chloride. However, protein, fat, calcium and potassium were correlated negatively with elevated somatic cell count in mastitic milk. It is concluded that udder infections resulting in elevated somatic cells may alter the mineral and trace element profile of milk, and magnitude of changes may have diagnostic and prognostic value. 相似文献

Glutathione is an immensely important antioxidant, particularly in the central nervous system. The scavenging mechanism of glutathione towards the OH radical was studied theoretically, considering its neutral, non-zwitterionic form relevant to acidic media. Gibbs free barrier and released energies involved in hydrogen abstraction from the different sites of glutathione by an OH radical were studied at the B3LYP/6-31G(d,p), B3LYP/AUG-cc-pVDZ, M06/AUG-cc-pVDZ, M06-2X/AUG-cc-pVDZ levels of density functional theory. Solvation in bulk aqueous media was also studied at all these levels of theory employing the polarizable continuum model. Our study shows that a hydroxyl radical can abstract a hydrogen atom easily from glutathione. Thus, glutathione is shown to be an efficient scavenger of OH radicals, which is in agreement with the results of previous studies. Figure

Low transpiration rates in pearl millet under fully irrigated conditions decrease plant water use at vegetative stage and then increase the water availability during grain filling and finally the terminal drought tolerance. Hundred and thirteen recombinant inbred lines developed from a cross between H77/833-2 and PRLT2/89-33 (terminal drought-sensitive?×?tolerant genotype) were evaluated to map transpiration rate (Tr, a proxy for canopy conductance), organ weights, leaf area and thickness and to study their interactions. Transpiration rate was increased by two H77/833-2 and two PRLT2/89-33 alleles on linkage group (LG) 2, whose importance depended on the vapor pressure deficit. The two H77/833-2 and one PRLT2/89-33 alleles co-mapped to a previously identified major terminal drought tolerance quantitative trait locus (QTL), although in a much smaller genetic interval. The other Tr allele from H77/833-2 also enhanced biomass dry weight and co-located with a formerly identified stover and tillering QTL. Leaf characteristics were linked to two loci on LG7. Plant water use was increased and decreased by different loci combinations for Tr, tillering and leaf characteristics, whose respective importance depended on the environmental conditions. Therefore, different alleles influence plant water use and have close interactions with one another and with the environment, so that different ideotypes for plant water use exist or could be designed from specific allele combinations conferring particular physiological characteristics for specific adaptation to a range of terminal drought conditions. 相似文献

The effects of synthetic phytohormones, such as jasmonic acid (JA) and methyl jasmonate (MeJA), on induced plant defenses and protection against herbivores have been well documented. Previous research on tomatoes has shown that exogenous JA and MeJA can elevate the activities of defensive enzymes, like polyphenol oxidases (PPOs) and proteinase inhibitors (PIs), and reduce the performance of herbivores. While such defenses are believed to depend on the availability of resources for the plant, data on the relationship between nutrient availability and tomato JA defenses are scarce. For this study, we grew cultivated tomatoes under different nutrient regimes, that is, high, moderate, or low, and sprayed them with 1.5?mM MeJA or a control solution to assess (1) the oviposition preference of Helicoverpa armigera moths for these plants; (2) the plant??s total nitrogen and protein content; (3) the plant??s PPO and PI activity; and finally (4) the performance of H. armigera larvae on these plants. The results revealed that adult moths selecting an oviposition site did not discriminate between the treatments. Plants had a higher N-content when grown under a high-nutrient regime, but did not differ in total protein, irrespective of the MeJA treatment. Also, PPO activity was the same across treatments, but trypsin inhibitor (TPI) activity was highest in plants grown under a moderate fertilization regime. MeJA-induced TPI activity in all treatments was high but equal. Larvae had the highest weight gain on plants grown under a moderate fertilization regime. Interestingly, they stopped feeding on plants induced with MeJA, resulting in 100?% mortality. Our results indicate that the plant??s normal TPI activity is maximal under moderate-nutrient conditions and that this, surprisingly, coincides with maximal larval weight gain. In contrast, induction of TPI activity via MeJA is much stronger and independent of the plant??s fertilization history yet suffices to make the larvae stop feeding. We speculate that a moderate induction of TPI activity may result in a compensatory feeding response while inducing a high level of TPI activity will lead to enhanced protection. 相似文献

A simple, sensitive and high throughput liquid chromatography/positive-ion electrospray ionization mass spectrometry (LC–ESI-MS/MS) method has been developed for the simultaneous determination of valacyclovir and acyclovir in human plasma using fluconazole as internal standard (IS). The method involved solid phase extraction of the analytes and IS from 0.5 mL human plasma with no reconstitution and drying steps (direct injection of eluate). The chromatographic separation was achieved on a Gemini C18 analytical column using isocratic mobile phase, consisting of 0.1% formic acid and methanol (30:70 v/v), at a flow-rate of 0.8 mL/min. The precursor → product ion transition for valacyclovir (m/z 325.2 → 152.2), acyclovir (m/z 226.2 → 152.2) and IS (m/z 307.1 → 220.3) were monitored on a triple quadrupole mass spectrometer, operating in the multiple reaction monitoring (MRM) mode. The method was validated over the concentration range 5.0–1075 ng/mL and 47.6–10225 ng/mL for valacyclovir and acyclovir respectively. The mean recovery of valacyclovir (92.2%), acyclovir (84.2%) and IS (103.7%) from spiked plasma samples was consistent and reproducible. The bench top stability of valacyclovir and acyclovir was extensively evaluated in buffered and unbuffered plasma. It was successfully applied to a bioequivalence study in 41 healthy human subjects after oral administration of 1000 mg valacyclovir tablet formulation under fasting condition. 相似文献

Chromosomes have been studied in seven species of bruchids belonging to the subfamilies Bruchinae and Amblycerinae. The diploid number of chromosomes is twenty in Bruchidius multilineolatus (Pic.), B. aureus Arora, B. saundersi (Jek.) and Conicobruchus indicus (Pic.); Bruchus pisorum L. and Specularius maindroni (Pic.) possess a diploid number of twenty-two whereas Spermophagus latescenta Arora has a diploid number of thirty-two. The male sex chromosome mechanism, however, is uniformly Xyp in all these species. The usual diplotene and diakinesis stages are not discrete in Bruchidius multilineolatus and Specularius maindroni. Implication of different dietary specific lipids such as phytantriol (PT) and glyceryl monooleate (GMO) on enhancing the oral bioavailability of amphotericin B (AmB) was examined. Liquid crystalline nanoparticles (LCNPs) were prepared using hydrotrope method, followed by in vitro characterization, Caco-2 cell monolayer uptake, and in vivo pharmacokinetic and toxicity evaluation. Optimized AmB-LCNPs displayed small particle size (<?210 nm) with a narrow distribution (~?0.2), sustained drug release and high gastrointestinal stability, and reduced hemolytic toxicity. PLCNPs presented slower release, i.e., ~?80% as compared to ~?90% release in case of GLCNPs after 120 h. Significantly higher uptake in Caco-2 monolayer substantiated the role of LCNPs in increasing the intestinal permeability followed by increased drug titer in plasma. Pharmacokinetic studies demonstrated potential of PT in enhancing the bioavailability (approximately sixfold) w.r.t. of its native counterpart with reduced nephrotoxicity as presented by reduced nephrotoxicity biomarkers and histology studies. These studies established usefulness of PLCNPs over GLCNPs and plain drug. It can be concluded that acid-resistant lipid, PT, can be utilized efficiently as an alternate lipid for the preparation of LCNPs to enhance bioavailability and to reduce nephrotoxicity of the drug as compared to other frequently used lipid, i.e., GMO. 相似文献
