Muramyl dipeptide induces acute joint inflammation in the mouse   总被引:1,自引:0,他引:1  
Acute joint inflammation was produced in BALB/c mice by a single intravenous injection of synthetic muramyl dipeptide (MDP), its stereoisomers and 6-O-acyl derivatives of MDP. Four adjuvant-active, but not five adjuvant-inactive MDP analogs induced acute swelling and erythema of the ankles and wrists which were detected around 6-10 hr, reached the maximum severity by 18-24 hr and subsided by days 3 to 4 after injection. Introduction of the stearoyl group, but not the alpha-branched long chain fatty acid group into the C-9 hydroxyl group of MDP enhanced and prolonged the joint lesions compared with MDP.  相似文献

109 malignant lymphomas were surveyed by Southern blot analysis and polymerase chain reaction (PCR) for Epstein-Barr virus (EBV) DNA and compared with 16 examples of non-neoplastic lymphadenopathy and 4 normal thymuses. In specimens positive by the method of Southern and PCR, in situ hybridization studies were performed on formalin-fixed, paraffin-embedded sections. By Southern blot analysis, two of seven Hodgkin's disease samples (29%) (one of mixed cellularity and the other of lymphocyte predominance type), three of 56 B-cell lymphomas (5.6%) and five of 46 T-cell lymphomas (11%) demonstrated EBV DNA. However, the 16 examples of lymphadenitis and the 4 normal thymuses showed no EBV DNA. With PCR, EBV DNA was identified in one B-cell lymphoma, nine T-cell lymphomas, ten lymphadenitis specimens and two of the normal thymus, in addition to the positive specimens determined by the Southern blotting method. These results indicate that the presence of EBV DNA is not related to lymphoid malignancy, but enhancement of the DNA is demonstrated in some neoplastic conditions. By in situ hybridization, EBV genomes were not detected in all PCR-positive cases, but only in those positive by Southern blot analysis.  相似文献

Two trisaccharides, two tetrasaccharides, one penta-, one hexa-, two hepta-, one deca- and two undeca-saccharides were isolated from several Japanese black bear milk samples by chloroform/methanol extraction, gel filtration and preparative thin-layer chromatography. An assay for total hormone-sensitive lipase (HSL) in rat fat cells was devised in which fat-associated HSL was solubilized with ether, and triolein or cholesteryloleate was used as substrate. Norepinephrine (NE) caused marked release of glycerol from fat cells but did not activate HSL as estimated using triolein or cholesteryloleate as substrate. Propranolol, a beta-blocker, inhibited NE-induced lipolysis in fat cells without a concomitant reduction in HSL activity. The antilipolytic action of insulin on NE-induced lipolysis could not be explained by a decrease in HSL activity. Neither ACTH-induced lipolysis in fat cells nor its inhibition by insulin was accompanied by matching fluctuations in HSL activity. These results indicate that neither NE and ACTH-induced lipolysis in fat cells, nor the antilipolytic actions of propranolol and insulin, involve fluctuations in HSL activity.  相似文献

Peptide(s) produced from degraded soybean protein by an alkaline protease from Bacillus circulans HA12 (degraded soybean-meal products; DSP) increased the number of both the root hair cells (trichoblasts) and hairless cells (atrichoblasts) of Brassica rapa by about 4.4 times and 1.9 times, respectively. To identify the root hair-promoting peptide(s) in DSP, the origin protein of the root hair-promoting peptide(s) was identified as Kunitz trypsin inhibitor (KTI). The root hair-promoting peptide in the degraded products of KTI was purified and produced a signal of 1,198.2 Da with matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI–TOF MS) analysis. A search of the amino acid sequence of KTI located the peptide GGIRAAPTGNER, which had a molecular weight identical to 1,198.2 Da. The peptide GGIRAAPTGNER was chemically synthesized, and the synthetic peptide possessed root hair-promoting activity. Thus, it is concluded that this peptide in DSP is the foreign bioactive peptide promoting the differentiation of root hairs.  相似文献

This study examined the time of hatching of the desert locust Schistocerca gregaria Forskål (Orthoptera: Acrididae) in the laboratory to test the effect of eggs within a pod versus individualized eggs. The pod organization of eggs is thought to play a role in controlling hatching time and to facilitate synchronous hatching at constant temperatures. In the present study, we examined the hatching times of eggs in a pod and individualized eggs under 24-h thermocycles and simulated field temperatures. We tested two patterns of thermocycles consisting of a 12-h thermoperiod (35 or 30 °C) and 12-h cryoperiod (low temperature period; 30 or 25 °C), and two patterns of field temperatures observed in a natural habitat, Mauritania, in May and September. The majority of eggs hatched during low temperature periods in all patterns tested. In addition, the variances of hatching times for individualized eggs were significantly greater than for egg pods in which a clear peak of time of hatching was observed. We show that egg condition influences hatching time under thermocycles of constant and fluctuating temperatures in the laboratory, and may play a role in the adaptive time of hatching.  相似文献

Perennial grasses can sequester soil organic carbon (SOC) in sustainably managed biofuel systems, directly mitigating atmospheric CO2 concentrations while simultaneously generating biomass for renewable energy. The objective of this study was to quantify SOC accumulation and identify the primary drivers of belowground C dynamics in a zero‐tillage production system of tropical perennial C4 grasses grown for biofuel feedstock in Hawaii. Specifically, the quantity, quality, and fate of soil C inputs were determined for eight grass accessions – four varieties each of napier grass and guinea grass. Carbon fluxes (soil CO2 efflux, aboveground net primary productivity, litterfall, total belowground carbon flux, root decay constant), C pools (SOC pool and root biomass), and C quality (root chemistry, C and nitrogen concentrations, and ratios) were measured through three harvest cycles following conversion of a fallow field to cultivated perennial grasses. A wide range of SOC accumulation occurred, with both significant species and accession effects. Aboveground biomass yield was greater, and root lignin concentration was lower for napier grass than guinea grass. Structural equation modeling revealed that root lignin concentration was the most important driver of SOC pool: varieties with low root lignin concentration, which was significantly related to rapid root decomposition, accumulated the greatest amount of SOC. Roots with low lignin concentration decomposed rapidly, but the residue and associated microbial biomass/by‐products accumulated as SOC. In general, napier grass was better suited for promoting soil C sequestration in this system. Further, high‐yielding varieties with low root lignin concentration provided the greatest climate change mitigation potential in a ratoon system. Understanding the factors affecting SOC accumulation and the net greenhouse gas trade‐offs within a biofuel production system will aid in crop selection to meet multiple goals toward environmental and economic sustainability.  相似文献
