To investigate the association between the polymorphism of P choose element (p. selectin, PS) and soluble P-selectin levels in atrial fibrillation (AF) thromboembolism in Han and Uigur population of Xinjiang. Method: Using ELISA method determination of plasma level of sPs. The frequency distributions of SNP sP-selectin gene promoter (−2123C/G) and SNP in exon region (Thr715Pro) were investigated by polymerase chain reaction (PCR)-restriction fragment length polymorphism and direct DNA sequence analysis among 302 Xinjiang Uigur and 340 age- and sex-matched Han people. Results: Cases sPs exist significant difference serum level and the control group. The frequencies of the −2123C/G allele among the Uigur population had no significant differences from those of the Han population. Thr715Pro did not show any polymorphism in the two populations. Conclusions: The sP-selectin gene polymorphisms are associated with serum sP-selectin levels or thromboembolic events, suggesting that the patients with nonvalvular AF and thromboembolic events may have genetic susceptibility.  相似文献

In China, great differences in economy, social characteristics and hygiene exist between developing and developed regions. A comparative study of infectious diarrhea between two regions was needed. Three groups of diarrheal patients were collected: children ≤5 year-olds from Beijing (developed region) and Henan Province (developing region), and adults over 18 year-olds from Beijing. A questionnaire was used to survey and feces samples were examined for 16 enteropathogens. We enrolled 1422 children and 1047 adults from developed region and 755 children from developing region. Virus positive rates were 32.98% for children and 23.67% for adults in developed region. The most prevalent pathogen for children was rotavirus whereas for adults was norovirus. Bacterial isolation rates were 13.92% for children from developed region, while 29.14% for children from the developing regions. For the greatest difference, Shigella accounted for 50.79% and was the dominant pathogen in the developing region, whereas in the developed region it was only 1.45%. There was no significant relationship between the local levels of development with diarrheogenic Escherichia coli (DEC) categories. But it was seen the notable differences between the population with different age: enteropathogenic E.coli (EPEC) and enteroaggregative E.coli (EAggEC) were the primary classes of DEC in children from both regions, whereas it was enterotoxigenic E.coli (ETEC) in adults. The symptoms of Shigella and Salmonella infection, such as bloody stools, white blood cells (WBC) and red blood cells (RBC) positivity and fever were similar in children, which may lead to the misidentification. Yersinia enterocolitica and shiga toxin-producing E.coli (STEC) infections were firstly reported in Beijing. There was a large difference in etiology of bacterial diarrhea between children in developing and developed regions of China.  相似文献

Extracting a subset of informative genes from microarray expression data is a critical data preparation step in cancer classification and other biological function analyses. Though many algorithms have been developed, the Support Vector Machine - Recursive Feature Elimination (SVM-RFE) algorithm is one of the best gene feature selection algorithms. It assumes that a smaller "filter-out" factor in the SVM-RFE, which results in a smaller number of gene features eliminated in each recursion, should lead to extraction of a better gene subset. Because the SVM-RFE is highly sensitive to the "filter-out" factor, our simulations have shown that this assumption is not always correct and that the SVM-RFE is an unstable algorithm. To select a set of key gene features for reliable prediction of cancer types or subtypes and other applications, a new two-stage SVM-RFE algorithm has been developed. It is designed to effectively eliminate most of the irrelevant, redundant and noisy genes while keeping information loss small at the first stage. A fine selection for the final gene subset is then performed at the second stage. The two-stage SVM-RFE overcomes the instability problem of the SVM-RFE to achieve better algorithm utility. We have demonstrated that the two-stage SVM-RFE is significantly more accurate and more reliable than the SVM-RFE and three correlation-based methods based on our analysis of three publicly available microarray expression datasets. Furthermore, the two-stage SVM-RFE is computationally efficient because its time complexity is O(d*log(2)d}, where d is the size of the original gene set.  相似文献

ObjectivesTo apply contact measurement and reference point system (RPS) alignment techniques to establish a method for 3D reconstruction of the edentulous jaw models with centric relation and to quantitatively evaluate its accuracy.MethodsUpper and lower edentulous jaw models were clinically prepared, 10 pairs of resin cylinders with same size were adhered to axial surfaces of upper and lower models. The occlusal bases and the upper and lower jaw models were installed in the centric relation position. Faro Edge 1.8m was used to directly obtain center points of the base surface of the cylinders (contact method). Activity 880 dental scanner was used to obtain 3D data of the cylinders and the center points were fitted (fitting method). 3 pairs of center points were used to align the virtual model to centric relation. An observation coordinate system was interactively established. The straight-line distances in the X (horizontal left/right), Y (horizontal anterior/posterior), and Z (vertical) between the remaining 7 pairs of center points derived from contact method and fitting method were measured respectively and analyzed using a paired t-test.ResultsThe differences of the straight-line distances of the remaining 7 pairs of center points between the two methods were X: 0.074 ± 0.107 mm, Y: 0.168 ± 0.176 mm, and Z: −0.003± 0.155 mm. The results of paired t-test were X and Z: p >0.05, Y: p <0.05.ConclusionBy using contact measurement and the reference point system alignment technique, highly accurate reconstruction of the vertical distance and centric relation of a digital edentulous jaw model can be achieved, which meets the design and manufacturing requirements of the complete dentures. The error of horizontal anterior/posterior jaw relation was relatively large.  相似文献

Four polyketides, leptosphaerone C (1), penicillenone (2), arugosin I (3) and 9-demethyl FR-901235 (4), as well as five known compounds, bacillosporin A (5), bacillosporin C (6), sequoiamonascin D (7), sequoiatone A (8), and sequoiatone B (9) were isolated from the Penicillium sp. JP-1, an endophytic fungus isolated from Aegiceras corniculatum. Their structures were determined by spectroscopic methods, mainly by 2D NMR spectroscopic analyses. Compound 1 showed cytotoxicity against A-549 cells with an IC50 value of 1.45 microM, while compound 2 showed cytotoxicity against P388 cells with an IC50 value of 1.38 microM.  相似文献

Joint genomic prediction (GP) is an attractive method to improve the accuracy of GP by combining information from multiple populations. However, many factors can negatively influence the accuracy of joint GP, such as differences in linkage disequilibrium phasing between single nucleotide polymorphisms (SNPs) and causal variants, minor allele frequencies and causal variants’ effect sizes across different populations. The objective of this study was to investigate whether the imputed high-density genotype data can improve the accuracy of joint GP using genomic best linear unbiased prediction (GBLUP), single-step GBLUP (ssGBLUP), multi-trait GBLUP (MT-GBLUP) and GBLUP based on genomic relationship matrix considering heterogenous minor allele frequencies across different populations (wGBLUP). Three traits, including days taken to reach slaughter weight, backfat thickness and loin muscle area, were measured on 67 276 Large White pigs from two different populations, for which 3334 were genotyped by SNP array. The results showed that a combined population could substantially improve the accuracy of GP compared with a single-population GP, especially for the population with a smaller size. The imputed SNP data had no effect for single population GP but helped to yield higher accuracy than the medium-density array data for joint GP. Of the four methods, ssGLBUP performed the best, but the advantage of ssGBLUP decreased as more individuals were genotyped. In some cases, MT-GBLUP and wGBLUP performed better than GBLUP. In conclusion, our results confirmed that joint GP could be beneficial from imputed high-density genotype data, and the wGBLUP and MT-GBLUP methods are promising for joint GP in pig breeding.  相似文献

氮是地表水体发生富营养化的主要因子,河流系统是氮输出的主要运移通道,养分在河流生态系统中的持留和趋向控制着污染物的输出。以巢湖流域一个受人为活动严重影响的农田源头溪流——六岔河为研究对象(包括4个渠道型、1个水塘型和3个河口型断面,对应长度分别为1.3km、0.15km和0.36km),设置9个监测点研究总氮(TN)、硝酸盐(NO3-N)、氨态氮(NH4^ -N)和总悬浮物(TSS)在溪流生态系统中的持留,评价人为严重干扰下的农田溪流生态系统在非点源污染物运移中的生态功能。结果表明:TN、NO3-N、NH4^ -N和TSS在溪流中的持留、释放受溪流的河流形态影响,水塘型和河口型断面是污染物持留的主要区域;TN、NO3-N、NH4^ -N和TSS在水塘型和河口型断面内的持留量分别占溪流持留量(基流、径流持留量的和)的61%、47%、75%和56%。降雨-径流过程中发生的持留是污染物持留的主要部分,TN、NO3-N、NH4^ -N和TSS的持留量分别占溪流持留量的93%、97%、89%和96%;渠道型断面是溪流最主要的内在污染源,污染物释放量占溪流释放量的90%以上;受水塘型断面出口处的水坝影响,位于水塘前的渠道型断面在基流和降雨-径流过程中均有效地持留污染物,而其余渠道型断面在不同水文条件下呈现出不同的持留特性。非点源污染物在溪流中的持留和释放的空间动态变化是溪流生态系统对自然和人为干扰的一种综合响应,有必要恢复溪流生态系统功能,控制农业非点源污染。  相似文献

茶多糖是一种从茶叶中提取的酸性糖蛋白,具有良好的抗氧化活性。以自由基清除率为指标,分析皖西南地区夏秋茶多糖的抗氧化活性,基于H2O2和EDTA-Fe2+建立的外源性羟基自由基(·OH)损伤细胞模型和PMA诱导内源性羟基自由基损伤模型,进一步探讨茶多糖对自由基损伤的修复作用机制。结果表明,茶多糖具有良好的体外抗氧化活性,对DPPH·和·OH均具有较强的清除效果,EC50值分别为209.5和535.2μg·mL–1,最大清除效率与Vc相当。细胞增殖实验表明,外源性和内源性自由基氧化损伤模型中细胞存活率均随着茶多糖浓度的增加而升高,在茶多糖浓度为800μg·mL–1时细胞存活率分别高达87.41%和85.84%,且显著高于模型组(47.67%和48.03%)。在修复机制上,利用激光共聚焦显微镜显影细胞内活性氧(ROS)分布以及荧光强度,分析结果显示,与模型组相比,茶多糖对于细胞模型中外源和内源性ROS均具有明显的清除效果,与体外抗氧化实验结果一致。茶多糖在体外表...  相似文献

Many studies reported the influence of wind erosion on soil degradation and the effect of vegetation coverage on preventing wind erosion. However, fewer studies have quantitatively measured the grassland soil particle size fractions and nutrients’ loss caused by wind erosion under different vegetation coverage. Aims: We conducted a field experiments to (1) to explore the effect of vegetation coverage on soil wind erosion; (2) examine quantitatively the effects of wind erosion on soil texture, and determine the most erodible particles fraction of soil; (3) to examine quantitatively the soil carbon, nutrients such as nitrogen and phosphorus loss caused by wind erosion under different vegetation coverage. Methods: Six vegetation coverage treatments (0 %, 15 %, 35 %, 55 %, 75 % and 95 %) were constructed. To be able to monitor wind erosion status under more diverse weather conditions, three consecutive repeat experiments under different weather condition were conducted. Results: The results show that all the residue soil samples after wind erosion became coarser than that of original soil samples. The degree of change for the soil particle size distribution before and after wind erosion gradually increased with the less of vegetation coverage. The critical particle size for distinguishing the original soil sample and the residue soil after wind erosion occurred in the range of 125 μm and 210 μm depending on the vegetation cover. The fractions below or above the critical particle size are either easy to deplete or favoured by wind erosion, respectively. The most reduction occurs between 50 and 90 μm depending on the different weather condition and vegetation coverage. Due to the disproportionately greater amounts of nutrients in the fine soil particles, the preferential depletion of fine particles directly lead to a preferentially significant depletion of organic carbon and nutrients. The organic carbon and nutrient contents in the residue soil after erosion decreased significantly compared to that in the original soil. The soil nutrient loss ratio decrease significantly with the increase of vegetation coverage. Conclusions: Wind erosion is an important factor to affect the evolution of soil texture and soil nutrient. Vegetation coverage has a major impact on both preventing wind erosion and decreasing loss ratio of fine particles and nutrients. If we want to effectively protect the fine particles and nutrients, the vegetation cover should be maintained at least above 35 %.  相似文献
