Lyophilized horseradish peroxidase (HRP) exhibits poor stereoselectivity in the sulfoxidation of thioanisole when the enzyme is either redissolved in water or suspended in organic solvents. However, when HRP is co-lyophilized in the presence of lyoprotectants or ligands, its stereoselectivity, although still low in most organic solvents, increases up to 4-fold if assayed in secondary or tertiary alcohols (but not in their linear isomers). A mechanistic hypothesis is presented explaining this puzzling phenomenon on the basis of a model of the active site of the enzyme-substrate complex derived from its X-ray crystal structure by means of molecular dynamics and energy minimization.  相似文献

Recently, a system that measures tissue oxygen tension using time-resolved luminescence-based optical sensors has become available commercially (Oxford Optronix, Oxford, England). Two experiments were conducted using this system. First, the oxygen tension distribution was measured in two tumor lines: a spontaneous mouse fibrosarcoma, FSa-II, and a human squamous cell carcinoma xenograft, FaDu. The area in which the pO(2) was equal to or lower than 2.5 mmHg was defined as the hypoxic lesion, and the hypoxic cell fraction was taken as the fraction of these measurements in a tumor. The measured hypoxic cell fractions were compared with those determined by the paired cell survival assay for tumors of various sizes. Second, the tumor tissue pO(2) was measured continuously after administration of two different anesthetics to evaluate the effect of these drugs on tissue pO(2). Results indicated a good agreement between the hypoxic cell fractions measured by this system and those determined by the paired cell survival curve assay for tumors smaller than approximately 500 mm(3). For tumors larger than approximately 500 mm(3), the hypoxic cell fractions measured by the oxygen probe system were higher than those measured by the paired cell survival assay. This may suggest that the hypoxic cell fraction measured by the oxygen probes included both hypoxic and necrotic areas in large tumors where necrotic lesions occupied a significant portion of the tumor. Continuous measurements of pO(2) after anesthesia (Nembutal, or ketamine plus xylazine) showed a consistent rise in the pO(2) during the first 20-30 min of measurement. Subsequently, the pO(2) values became constant or continued to rise slowly. For comparison, the tumor cell survivals were assayed after a dose of 20 Gy given in air at 5, 20 and 60 min after anesthesia. The result showed a decrease in cell survival only in tumors irradiated 20 min after an injection of Nembutal.  相似文献

Glucose sensing mechanism has been intensively studied in pancreatic cells and neurons. Depolarization of membrane potential by closure of KATP , Kv and TASK channel, and subsequently Ca2+ entry via L-type voltage gated Ca2+ channel (VGCC) are implicated to mediate the signal transduction in these cells. However, the mechanism of non-excitable cells, which are lacking VGCC, for sensing glucose remains unclear. In this study, we utilized the calcium ratio measurement and patch clamping technique to study the effects of low glucose on [Ca2+]i and currents in the human embryonic kidney epithelial cells (HEK 293). We found low glucose evoked a significant reversible [Ca2+]i elevation in HEK 293 independent of the closure of Kv channels. This increase of [Ca2+]i was mediated by Ca2+ entry across plasma membrane and exhibited a dosage dependent behaviour to external glucose concentration. The low glucose-induced entry of Ca2+ was characterized as a voltage independent behaviour and had cation permeability to Na+ and Ca2+. The modulation of PLC, AMPK, tyrosine kinase and cADPribose failed to regulate this glucose-sensitive Ca2+ entry. In addition, the entry of Ca2+ was insensitive to nifedipine, 2APB, SKF, La3+, Gd3+, and KBR9743, suggesting a novel signal pathway in mediating glucose sensing.  相似文献

叶色突变体往往伴随着叶绿素含量变化及叶绿体结构异常,是研究叶绿体发育与光合作用相关基因功能的重要材料。该研究通过甲基磺酸乙酯(EMS)诱变籼稻(Oryzasativasubsp.indica)品种华占(HZ)获得黄绿叶突变体,将其命名为ygl18 (yellow-green leaf 18)。与野生型相比,黄绿叶突变体ygl18自三叶期起叶片开始变黄且程度不断加深,同时伴随着光合速率与叶绿素含量下降,且结实率、千粒重及有效穗数均显著降低。透射电镜观察结果显示, ygl18的叶绿体结构紊乱,基质片层疏松,发育受到抑制,与叶片出现黄绿色表型一致。遗传分析表明, ygl18突变性状受1对隐性等位核基因控制,这对等位基因位于水稻第3号染色体长臂标记InDel2和InDel3之间115.2 kb范围内。进一步研究发现该突变体表型是编码铁氧还蛋白FdC2的基因LOC_Os03g48040的5’UTR发生突变所致。通过CRISPR转基因实验验证了该基因对表型的控制作用。研究结果揭示了叶色调控网络的遗传基础,可为今后选育高光效水稻品种提供新线索。  相似文献

We collected a specimen of Chinese Pygmy Dormouse (Typhlomys cinereus)from Nanling Nature Reserve in Guangdong Province in August 2012,analyzed the specimen’s morphological and skull indices,and compared them with the published data for other T. cinereus subspecies located throughout China. The skull of our specimen had four pairs of palatal holes:among them,the two posterior pairs merged together to form two large and long holes,and the posterior edge of the nasals was located vertically below the frontopremaxillary suture. Based on these two unique characteristics,we identified the specimen as the subspecies T. c. daloushanensis. This is the first time this species has been recorded in Guangdong Province.  相似文献

挖掘与稻米蒸煮品质相关的数量性状基因座(quantitative trait locus, QTL),分析候选基因,并通过遗传育种手段改良稻米蒸煮品质相关性状,可有效提升稻米的口感。以籼稻华占(Huazhan, HZ)、粳稻热研2号(Nekken2)及由其构建的120个重组自交系(recombinant inbred lines, RILs)群体为实验材料,测定成熟期稻米的糊化温度(gelatinization temperature, GT)、胶稠度(gel consistency, GC)和直链淀粉含量(amylose content, AC)。结合高密度分子遗传图谱进行QTL定位,共检测到26个与稻米蒸煮品质相关的QTLs (糊化温度相关位点1个、胶稠度相关位点13个、直链淀粉含量相关位点12个),其中最高奇数的可能性(likelihood of odd, LOD)值达30.24。通过实时荧光定量PCR (quantitative real-time polymerase chain reaction, qRT-PCR)分析定位区间内候选基因的表达量,发现6个基因在双亲间的表达量差异显著,推测LOC_Os04g20270LOC_Os11g40100的高表达可能会极大地提高稻米的胶稠度,而LOC_Os01g04920LOC_Os02g17500的高表达以及LOC_Os03g02650LOC_Os05g25840的低表达有助于降低直链淀粉含量。这些结果为培育优质水稻新品种奠定了分子基础,并为揭示稻米蒸煮品质的分子调控机制提供了重要的遗传资源。  相似文献

We investigated the foraging area of three individuals (1 female and 2 males) of Rousettus leschenaulti (Chiroptera, Pteropodidae) in suburban Haikou City, Hainan Province, South China from November 2005 to January 2006 using radio telemetry. These animals left the daytime roosting sites about 90 min after sunset with no significant difference in departure time between the male and female bats. 通过探究水分调亏对河西地区膜下滴灌菘蓝(Isatis indigotica)各项生理指标、产量和水分利用的影响, 为菘蓝高效节水种植提供理论指导。于2016年在河西走廊中部张掖市民乐县益民灌溉试验站进行菘蓝水分调亏研究, 在保持苗期和肉质根成熟期充分灌溉的情况下, 在菘蓝营养生长期和肉质根生长期分别进行轻度、中度和重度的水分亏缺处理, 并测定各项光合生理指标、产量和水分利用率。结果表明, 营养生长期和肉质根生长期的中度与重度水分亏缺显著降低了菘蓝叶片净光合速率、叶面积指数、株高及主根长, 并且随水分亏缺程度加重降幅增大; 而轻度水分亏缺与对照组的差异不显著。营养生长期和肉质根生长期轻度水分亏缺处理的菘蓝产量与水分利用效率最高, 分别达到8 239.56 kg·hm-2和24.11 kg·hm-2·mm-1; 其它水分亏缺处理组产量和水分利用效率均有所降低, 与对照组之间差异显著(P<0.05), 重度水分亏缺处理各项指标均最低。因此, 最优的菘蓝水分调控处理为营养生长期和肉质根生长期的轻度水分调亏, 能够降低菘蓝耗水量, 提高水分利用效率且其产量不会降低。  相似文献

Cryopreservation of callus tissue of Artimisia annua L. was optimized. Two lines of calli were precultured on MS medium with 5% (v/v) dimethyl sulfoxide, and protected by a cryoprotectant containing 15% (v/v) ethylene glycol, 15% (v/v) dimethyl sulfoxide, 30% (v/v) glycerol and 13.6% (w/v) sucrose. The highest survival rate of callus A201 reached 87% after it was pretreated at 25°C, cryopreserved by liquid nitrogen, recovered in water bath at 25°C and reloaded at 25°C with 34% (w/v) sucrose solution, and that of callus A202 reached 78% after it was treated as callus A201, except pretreated at 35°C, recovered at 35°C and reloaded with 47.8% (w/v) sucrose solution.  相似文献
