It is generally accepted that copper toxicity is a consequence of the generation of reactive oxygen species (ROS) by copper ions via Fenton or Haber-Weiss reactions. Copper ions display high affinity for thiol and amino groups occurring in proteins. Thus, specialized proteins containing clusters of these groups transport and store copper ions, hampering their potential toxicity. This mechanism, however, may be overwhelmed under copper overloading conditions, in which copper ions may bind to thiol groups occurring in proteins non-related to copper metabolism. In this study, we propose that indiscriminate copper binding may lead to damaging consequences to protein structure, modifying their biological functions. Therefore, we treated liver subcellular membrane fractions, including microsomes, with Cu2+ ions either alone or in the presence of ascorbate (Cu2+/ascorbate); we then assayed both copper-binding to membranes, and microsomal cytochrome P450 oxidative system and GSH-transferase activities. All assayed sub-cellular membrane fractions treated with Cu2+ alone displayed Cu2+-binding, which was significantly increased in the presence of Zn2+, Hg2+, Cd2+, Ag+1 and As3+. Treatment of microsomes with Cu2+ in the μM range decreased the microsomal thiol content; in the presence of ascorbate, Cu2+ added in the nM concentrations range induced a significant microsomal lipoperoxidation; noteworthy, increasing Cu2+ concentration to ≥50 μM led to non-detectable lipoperoxidation levels. On the other hand, μM Cu2+ led to the inhibition of the enzymatic activities tested to the same extent in either presence or absence of ascorbate. We discuss the possible significance of indiscriminate copper binding to thiol proteins as a possible mechanism underlying copper-induced toxicity. 相似文献

Proteomics was used to identify a protein encoded by ORF 3a in a SARS-associated coronavirus (SARS-CoV). Immuno-blotting revealed that interchain disulfide bonds might be formed between this protein and the spike protein. ELISA indicated that sera from SARS patients have significant positive reactions with synthesized peptides derived from the 3a protein. These results are concordant with that of a spike protein-derived peptide. A tendency exists for co-mutation between the 3a protein and the spike protein of SARS-CoV isolates, suggesting that the function of the 3a protein correlates with the spike protein. Taken together, the 3a protein might be tightly correlated to the spike protein in the SARS-CoV functions. The 3a protein may serve as a new clinical marker or drug target for SARS treatment. 相似文献

Populus euphratica has been used as a plant model to study resistance against salt and osmotic stresses, with recent studies having characterized
the tonoplast and the plasma membrane ATPases, and two Na+/H+ antiporters, homologs of the Arabidopsis tonoplast AtNHX1, were published in databases. In the present work we show that P. euphratica suspension-cultured cells are highly tolerant to high salinity, being able to grow with up to 150 mM NaCl in the culture
medium without substantial modification of the final population size when compared to the control cells in the absence of
salt. At a salt concentration of 300 mM, cells were unable to grow but remained highly viable up to 17 days after subculture.
The addition of a 1-M-NaCl pulse to unadapted cells did not promote a significant loss in cell viability within 48 h. In tonoplast
vesicles purified from cells cultivated in the absence of salt and from salt-stressed cells, vacuolar H+-pyrophosphatase (V-H+-PPase) seemed to be the primary tonoplast proton pump; however, there appears to be a decrease in V-H+-PPase activity with exposure to NaCl, in contrast to the sodium-induced increase in the activity of vacuolar H+-ATPase (V-H+-ATPase). Despite reports that in P. euphratica there is no significant difference in the concentration of Na+ in the different cell compartments under NaCl stress, in the present study, confocal and epifluorescence microscopic observations
using a Na+-sensitive probe showed that suspension-cultured cells subject to a salt pulse accumulated Na+ in the vacuole when compared with control cells. The influence of various Ppd genes on the beginning and duration of photoperiodic responce has been investigated in near isogenic lines of winter bread wheat Mironovskaya 808. During ontogenesis the photoperiodic responce is ascertained from the middle 2nd stage of ontogenesis according to Kuperman (usually a week later after vernalization completion in winter genotypes) and it is completed to the late 5th stage (2-3 weeks before heading). Different Ppd alleles do not affect the photoreaction intensity however they have an influence on its duration through the rate of development. Ppd-A1a and Ppd-B1a genes manifest shorter duration of expression when compared to the recessive alleles in the initial Mironovskaya 808 cultivar. Studies conducted along the southern Iberian coastline validate macrobenthic community analyses at taxonomic levels higher than that of species. Twelve studies on littoral benthic communities, carried out by the same research team, were selected spanning both a variety of sampling strategies (spatial, temporal, spatio-temporal) and substrate/habitat types (sediment, rock, algae). In order to establish differences between the results obtained at the taxonomic levels of species, family and order, similarities among stations were calculated using Spearman’s coefficient for ranges. A subset of three studies was selected to investigate possible differences in ‘best-explaining’ environmental variables with taxonomic level. The environmental variables selected at species level were the same as those found at levels of family and order. It is concluded that studies at the different levels of taxonomic resolution (species, family, order) lead to similar results both with regard to relative community distributions and the environmental variables associated with these. The importance of this result for monitoring similar benthic communities is discussed. 相似文献

Background and Aims Although density-specific stiffness,E/, (where E is Young's modulus and is wood density) is oftenassumed constant by the elastic similarity model, and in determinationof critical buckling height (Hcrit), few studies have testedthis assumption within species. Here this assumption is testedfor Pinus radiata growing across an environmental gradient,and theory is combined with data to develop a model of Young'smodulus. Methods Analyses use an extensive series of environmentalplots covering the range of climatic and edaphic conditionsover which P. radiata is grown in New Zealand. Reduced majoraxis regression was used to determine scaling exponents betweenloglog plots of Hcrit vs. groundline diameter (D), andE/ vs. D. Path analysis was used to identify significant directand indirect (through stem slenderness) edaphic and climaticinfluences on E. Key Results Density-specific stiffness exhibited 3-foldvariation. As E/ scaled positively with D, the exponent of 0·95between Hcrit and D exceeded the assumed value of 0·67under constant E/. The final path analysis model included meanair temperature in early autumn (Taut) and slenderness as significant(P < 0·05) positive direct influences on E. Tree leafarea index and Taut were indirectly associated with E throughtheir significant (P < 0·05) positive direct relationshipwith stem slenderness. Young's modulus was most sensitive toTaut, followed by stem slenderness then leaf area index, andthe final model explained 76 % of the variance in E. Conclusions The findings suggest that within speciesE/ variation may influence Hcrit and the scaling exponent betweenD and Hcrit so important in assumptions regarding allometricrelationships. Azithromycin is a macrolide antibiotic known to bind to lipids and to affect endocytosis probably by interacting with lipid membranes [Tyteca, D., Schanck, A., Dufrene, Y.F., Deleu, M., Courtoy, P.J., Tulkens, P.M., Mingeot-Leclercq, M.P., 2003. The macrolide antibiotic azithromycin interacts with lipids and affects membrane organization and fluidity: studies on Langmuir-Blodgett monolayers, liposomes and J774 macrophages. J. Membr. Biol. 192, 203-215]. In this work, we investigate the effect of azithromycin on lipid model membranes made of 1,2-dioleoyl-sn-glycero-3-phosphocholine (DOPC) or 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC). Thermal transitions of both lipids in contact with azithromycin are studied by (31)P NMR and DSC on multilamellar vesicles. Concerning the DPPC, azithromycin induces a suppression of the pretransition whereas a phase separation between the DOPC and the antibiotic is observed. For both lipids, the enthalpy associated with the phase transition is strongly decreased with azithromycin. Such effects may be due to an increase of the available space between hydrophobic chains after insertion of azithromycin in lipids. The findings provide a molecular insight of the phase merging of DPPC gel in DOPC fluid matrix induced by azithromycin [Berquand, A., Mingeot-Leclercq, M.P., Dufrene, Y.F., 2004. As toxic pollutants commonly found in tobacco (Nicotiana tabacum L.) products, lead (Pb) and cadmium (Cd) can enter the human body via smoking and thus pose a potential health risk to smokers. We conducted a greenhouse experiment to study the effects of arbuscular mycorrhizal (AM) inoculation with Glomus intraradices BEG 141 and organic amendment with cattle manure, alone or in combination, on the growth, P nutrition, and heavy-metal uptake by tobacco plants grown in soil to which was added Pb-Cd at 0/0, 350/1, 500/10, and 1,000/100?mg?kg?1, respectively. In general, AM colonization and plant growth were greatly reduced by Pb-Cd contamination, whereas organic amendment alleviated Pb-Cd stress and showed some beneficial effects on AM symbiosis and some soil parameters. AM inoculation, alone or in combination with organic amendment, increased plant dry weights and improved P nutrition significantly at all Pb-Cd addition levels, and, in most cases, it decreased Pb and Cd concentrations in tobacco plants and DTPA-extractable concentrations in soil. AM inoculation increased total glomalin-related soil protein (GRSP) concentrations in soil to which Pb-Cd was added. The higher soil pH and GRSP contents and the lower DTPA-extractable Pb and Cd concentrations contributed by AM inoculation and/or organic amendment may be contributing factors that lead to higher growth promotion and lower metal toxicity and uptake by plants. Our findings suggest that AM inoculation in combination with organic manure may be a potential method for not only tobacco production but phytostabilization of Pb-Cd-contaminated soil. 相似文献
