The 3-O-sulfotransferase (3-OST) family catalyzes rare modifications of glycosaminoglycan chains on heparan sulfate proteoglycans, yet their biological functions are largely unknown. Knockdown of 3-OST-7 in zebrafish uncouples cardiac ventricular contraction from normal calcium cycling and electrophysiology by reducing tropomyosin4 (tpm4) expression. Normal 3-OST-7 activity prevents the expansion of BMP signaling into ventricular myocytes, and ectopic activation of BMP mimics the ventricular noncontraction phenotype seen in 3-OST-7 depleted embryos. In 3-OST-7 morphants, ventricular contraction can be rescued by overexpression of tropomyosin tpm4 but not by troponin tnnt2, indicating that tpm4 serves as a lynchpin for ventricular sarcomere organization downstream of 3-OST-7. Contraction can be rescued by expression of 3-OST-7 in endocardium, or by genetic loss of bmp4. Strikingly, BMP misregulation seen in 3-OST-7 morphants also occurs in multiple cardiac noncontraction models, including potassium voltage-gated channel gene, kcnh2, affected in Romano-Ward syndrome and long-QT syndrome, and cardiac troponin T gene, tnnt2, affected in human cardiomyopathies. Together these results reveal 3-OST-7 as a key component of a novel pathway that constrains BMP signaling from ventricular myocytes, coordinates sarcomere assembly, and promotes cardiac contractile function.  相似文献

In this report we have examined the ability of ethanol to induce UDP-glucuronyltransferase activity in male rabbits using p-nitrophenol as substrate. The rabbit was found to be an excellent species for studies of ethanol induction since almost 3-fold increases in activity were observed relative to controls. Ethanol induction of p-nitrophenol UDP-glucuronyltransferase activity was even greater than induction by 3-methylcholanthrene, the prototypic inducer of this type of activity. Thus, the rabbit shows promise for studies of UDP-glucuronyltransferase isozymes that are induced by chronic ethanol consumption.  相似文献

The food of a population of Europeans Goldfinches breeding in Orchards was studied. More than 5% of the total foodstuff consumed was met by the seeds of seven food plants. Water, lipid, carbohydrate and protein content was determined from 20 different plants which Goldfinches used during the breeding season in order to calculate energy content. The energy of fresh seeds varied from 1.88 kJ/g in Tussilago farfara to 12.14 kJ/g in Dactylis glomerata. The dependence of the rate of energy intake (profitability) on the pecking rate and the size (weight) of the seeds was calculated. The profitability of various seeds is in agreement with the relative number of birds using them. Calculation of the feeding time that a Goldfinch needs during daylight hours, if it feeds only on a particular plant, indicates that there are several foods that cannot fill the energy demands of the bird during the breeding season. These are eaten only if others with a high energy content are not available. Thus food selection in Goldfinches depends, apart from availability, on seed profitability. The oxygen consumption of a male Goldfinch was significantly higher when the animal was feeding on seed heads of Tussilago farfara and Taraxacum officinale than when it fed on loose dry seeds, and so, different feeding techniques have to be taken into account in calculating net energy gain.  相似文献

We present a molecular phylogeny of the family Raphidiidae including representatives of 21 of the 26 genera. Sequences from the nuclear gene for the large subunit ribosomal RNA (28S rRNA) and the mitochondrial cytochrome c oxidase subunit 3 gene (cox3) were used. For the phylogenetic reconstructions we applied automated and manual approaches for sequence alignment and different evolutionary models and tree building algorithms. The trees based on the two alignment approaches were rather similar in their overall topology. A combination of both marker sequences increased the resolution of the trees. The six clades within the raphidiid family that emerged represent either single genera or groups of genera, namely: (i) the Nearctic genus Agulla Navás, (ii) the Nearctic/Central American genus Alena Navás, (iii) the Central Asiatic and Eastern Palaearctic genus Mongoloraphidia H. Aspöck & U. Aspöck, (iv) the Palaearctic Puncha clade, (v) the western Mediterranean Ohmella clade, and (vi) the Palaearctic Phaeostigma clade. The New World taxa Agulla and Alena are placed as successive out‐groups to a monophyletic Palaearctic clade. The Mongoloraphidia clade is distributed in the eastern Palearctic while the remaining three clades are exclusively (Ohmella clade) or mainly distributed in the western Palaearctic. The early radiation of extant Raphidiidae is interpreted based on the phylogenetic tree obtained in the present study, and the geological and palaeobiological processes around the K–T boundary.  相似文献

The heart of any vertebrate is formed from an apparently symmetric cardiac tube that loops consistently in the same direction along the left-right axis of the embryo. In the amphibian Xenopus laevis, inhibition of proteoglycan synthesis by p-nitrophenyl-beta-D-xylopyranoside during a narrow period of development from late gastrula to early neurula specifically eliminated the looping of the cardiac tube. Most of the proteoglycans synthesized during this period were heparan sulfate proteoglycans. Treatment with p-nitrophenyl-alpha-D-xylopyranoside, an analogue that does not inhibit proteoglycan synthesis, did not interfere with cardiac looping. The critical period for proteoglycan synthesis was coincident with the migration of cardiac primordia to the ventral midline. The inhibition of cardiac looping was further explored in explants of cardiac primordia and anterioventral ectoderm. In recombinate embryos in which half the embryo, and thus one of the two heart primordia, was treated with p-nitrophenyl-beta-D-xylopyranoside, and the other half was untreated, cardiac looping occurred normally. It is proposed that the left-right axis in Xenopus, as reflected in cardiac looping, is established early in development, and that proteoglycan synthesis is involved in the transduction of left-right axial information to the cardiac primordia during migration.  相似文献

In the experiments to be described two congenic inbred lines CB and CC and two recombinant lines CB.R1 and CC.R1 were used. All four lines differ only in regard to the major histocompatibility complex (MHC). To determine the percentage distributions of the two cell subsets in peripheral blood lymphocytes (PBL) in these lines, monoclonal antibodies to these two antigens were used. By FACScan there were more CD4+PBL in CB and CB.R1 lines (share B-F/B-L region, controlling class I/class II antigens with line CB) than CC and CC.R1, while the reverse was true with CD8+ subsets. There were more CD8+ PBL in the CC and CC.R1 lines and less in CB and CB.R1 lines. The ratio of CD4+ to CD8+ in CB chickens was 3.4 +/- 0.2 and in CC chickens 1.6 +/- 0.1.  相似文献

This report describes for the first time the effects of long-term continuous exposures of animals to small air ions and D.C. electric fields. In this study we exposed 200 female NAMRU mice (25/cage) to the following conditions: ± high ions (2×105/cm3), ± low ions (2×103/cm3), ± field only and ground (ion depleted, no field). Specially designed cages provided a defined D.C. field of about 2 kV/meter in ionized environments, with somewhat lower values in the field only cages. Detailed mapping of ion flux originating from a tritium foil generating system (multiple sources in an overhead plate) indicated a well defined, but heterogenous pattern with eight peak areas. Using a 100 cm2 probe, ion flux values ranged from 10–12–10–14 A/cm2, with an average flux of 8.7±6.8×10–13 A/cm2 in high negative ion cages, with good reproducibility between cages.Measurements of serum glucose, cholesterol, and urea nitrogen (samples taken every three months) showed a number of small but consistent and statistically significant differences between animals maintained in different environments during the first year of exposure. Serum globulin and whole blood serotonin, however, did not show any significant environmental effects. Interestingly, pairwise comparisons between high negative and low negative ion conditions, or between high positive and low positive ion conditions, or between the two ground conditions, revealed no significant differences between cages. This argues for a similarity of environmental responses for the mice maintained in each of the compared conditions.The results of a multiple classification analysis for the entire first year showed a preponderence of effects for the ionized cages, although other conditions also had highly significant differences as compared to the grand mean value. While this study has shown effects of only small magnitude (compared to normal physiological variations) in the female NAMRU mice studied here, the significance of these results strongly validates the precept of long-term air ion effects in animals. Given the known variability of different species and strains, future studies will have to determine the actual range of responsiveness of animals exposed on a long-term basis to defined air ion and electromagnetic environments.Deceased December 8, 1982.  相似文献

Transition from symmetry to asymmetry is a central theme in cell and developmental biology. In Xenopus embryos, dorsal-ventral asymmetry is initiated by a microtubule-dependent cytoplasmic rotation during the first cell cycle after fertilization. Here we show that the cytoplasmic rotation initiates differential cytoplasmic polyadenylation of maternal Xwnt-11 RNA, encoding a member of the Wnt family of cell-cell signaling factors. Translational regulation of Xwnt-11 mRNA along the dorsal-ventral axis results in asymmetric accumulation of Xwnt-11 protein. These results demonstrate spatially regulated translation of a maternal cell-signaling factor along the vertebrate dorsal-ventral axis and represent a novel mechanism for Wnt gene regulation. Spatial regulation of maternal RNA translation, which has been established in invertebrates, appears to be an evolutionarily conserved mechanism in the generation of intracellular asymmetry and the consequential formation of the multicellular body pattern.  相似文献
