The self-association process of histone H4 molecules into large aggregates is highly cooperative at an ionic strength of 0.03. Carefully prepared aggregates at this ionic strength are ribbon-shaped and take the form of right-handed helices. These ribbons have a uniform width of 20.5nm. The overall diameter of the “cylinder” formed by the helix is ca. 19.5nm, the average pitch of the helices 20nm, and the mean contour length 720nm. The thickness of the ribbon is less than 3nm.  相似文献

Two bacteriolytic enzymes were produced when Hartmanella glebae was grown in the presence of both Enterobacter aerogenes and Alcaligenes faecalis. The identification of enzyme I as N-acetylmuramidase was reported earlier. Enzyme II was purified by gel filtration on a Bio-Gel A column. A recovery of 68.76% with 72.3-fold purification was obtained. It was found that 5 and 10 mM MgCl2 significantly increased the bacteriolytic activity. It is a basic protein. The cell walls of Micrococcus lysodeikticus were lysed by the enzyme, and the products of digestion were purified by Amberlite CG-120 and Sephadex G-15 chromatography to facilitate the detection of amino sugars. After reduction of the oligosaccharides with sodium borohydride and acid hydrolysis, the amino sugars were identified by paper chromatography. It was found that enzyme II cleaved the glycosidic bond between N-acetylmuramic and and N-acetylglucosamine of the peptidoglycan moiety of the cell walls. Thus, the enzyme was identified as endo-beta-N-acetylmuramidase.  相似文献

Continuous survey of the distribution of RNA coliphages in Japan   总被引:1,自引:0,他引:1  
In order to demonstrate the stability and continuity of RNA coliphages (phages) in their natural habitats, we investigated the amount and group types of RNA phages in sewage samples collected continuously from domestic drainage in Japan proper and islands in the seas adjacent to Japan (abbreviated simply as islands, hereafter) over a 5-yr period from 1973 to 1977. It was found that the frequencies of isolation of RNA phages were fairly high and constant. The group types of RNA phages isolated were also stable in the three cities. Choshi, Niigata, and Toyama in Japan proper. The average for the three cities was group II:III = 3:1. The investigation in islands revealed that the frequencies of isolation of RNA phages were fairly high as in the case of the above three cities in Japan proper and the group types of RNA phages isolated were also stable. That is to say, group II phages were predominant on Rishiri Island, Rebun I., Iki I., and Tsushima I., which are located relatively near to mainland Japan, while group III phages were predominant on Amamiohshima I., mainland Okinawa, Ishigakijima I., and Iriomotejima I., which are located south of Kyushu. It can thus be said that the RNA phages in the domestic drainage of Japan proper and islands remained more or less stable over at least the 5-yr period, and an apparent difference in the geographical distribution of RNA phages in Japan exists between Kyushu and Amamiohshima I.  相似文献

D-manno-3-Heptulose (5) was synthesized by dimethyl sulfoxide-phosphorus pentaoxide oxidation of 1,2:3,4:6,7-tri-O-isopropylidene-D-glycero-D-manno-heptitol (3, prepared from volemitol), followed by hydrolysis. D-ido-3-Heptulose (8) was synthesized similarly by oxidation of 1,2:4,5:6,7-tri-O-isopropylidene-D-glycero-l-galacto-heptitol (7, prepared from D-glycero-l-galacto-heptitol, 6). Another tri-O-isopropylidene derivative (11), having a free primary hydroxyl group, was produced in larger amount than 7, and 11 yielded D-glycero-l-galacto-heptose (14). Compound 8 was also synthesized by way of 1,2:4,5.6,7-tri-O-isopropylidene-D-glycero-l-gulo-heptitol (15). The production of 15 from D-glycero-l-gulo-heptitol (13) was accompanied by a larger amount of 2,3:4,5:6,7-tri-O-isopropylidene-D-glycero-D-ido-heptitol (17) which, upon oxidation followed by hydrolysis, yielded D-glycero-D-ido-heptose (18). One of the two tri-O-isopropylidene derivatives obtained by acetonation of perseitol, 2,3:4,5:6,7-tri-O-isopropylidene-D-glycero-D-galacto-heptitol (19), yielded D-glycero-D-galacto-heptose (20).  相似文献

The effect of ethylene on DNA synthesis in potato tuber discsinduced by cutting was examined. Continuous presence of ethylenein the ambient atmosphere of the slices lowered the rate ofinduced DNA synthesis by about 50%, but did not alter the timecourse pattern of development of DNA synthesis. RNA and proteinsyntheses were not affected. The inhibitory effect on DNA synthesiswas observed at as low as 0.01 µl/liter and was due tothe specific action of ethylene, not to a non-specific actionof gaseous hydrocarbons. Ethylene also decreased the numberof cells which could synthesize DNA. The results of ethylenetreatment of various durations at various times after cuttingindicate that a process prerequisite for DNA synthesis and susceptibleto ethylene action starts at about 6 hr after cutting and continuesfor only a limited period. Neuromuscular unit (NMU) spikes activated during the tonic vibration reflex (TVR) in man were studied by means of the cross-correlogram test. Those NMUs which preferredly fired at some definite interval with good correlation with vibration were called "locked" spikes. The amount above zero correlation level seen in the cross-correlogram was expressed as ipsilateral "excitatory frequency", Ef. TVR contraction of the ipsilateral quadriceps femoris muscle was stimulated by voluntary contraction of the contralateral quadriceps femoris. I. Cross-correlogram test of the contralateral voluntary NMU spikes with ipsilateral vibration revealed "contralateral inhibitory modulation". Such inhibitory modulation was observed in 24% of the contralateral NMUs during voluntary tracking contraction with visual feed-back and in 22% of NMUs during blind simulated contraction without visual feed-back. The "subtracted" amount, i.e., contralateral inhibitory modulation, was also expressed as the contralateral "inhibitory frequency", If, which was calculated from the indentations of the cross-correlogram. 2. When varied vibratory frequency was applied to the muscle both Ef and If attained their maximum value around an "optimal frequency" of vibration, i.e., 80-95 HZ. 3. The ratio Ef/If was proposed as the reciprocal co-activation ratio which was fairly independent of the change of vibratory frequency. An average value of Ef/If ratio was calculated at 2.2 +/- 0.9 from 19 experiments.  相似文献

2,4-Dinitrophenol (DNP) was found to cause a "clearing response" of myosin B in a medium in which "superprecipitation" of myosin B would otherwise take place. The effect of actin concentration on Mg-ATPase [EC 3.6.1.3] of HMM was studied in the presence and absence of DNP. The results indicate that DNP causes an increase rather than a decrease in the affinity of HMM for actin, and that it causes a decrease only in the actin-activated portion of the Mg-ATPase activity. Using a light-scattering technique, it was shown that neither the ATP-induced dissociation of acto-HMM nor subsequent reassociation is significantly affected by the presence of DNP. As for the formation of the myosin-phosphate-ADP complex in the myosin-ATPase reaction, it was shown that formation of the reactive complex is not affected by DNP. It can thus be concluded that DNP inhibits the decomposition of the actomyosin-phosphate-ADP complex, which is thought to be coupled with superprecipitation.  相似文献

The observations of Microsporum canis with cryoscanning and scanning electron microscopy without fixation and dehydration were reported. In the former an almost native state was observed through showing some fuzzy outlines due to frost; in the latter it was shown that marked shrinkage and distortion had occured. BackgroundAmebiasis, caused by Entamoeba histolytica, is spreading in developing countries and in many developed countries as a sexually transmitted infection. Here, we evaluated the efficacy of serological screening to identify asymptomatic E. histolytica infection as a potential epidemiological control measure to limit its spread.Methodology/Principal findingsThis cross-sectional study was carried out between January and March 2021 in an HIV-negative men who have sex with men (MSM) cohort at the National Center for Global Health and Medicine. Serological screening was performed using a commercially available ELISA kit. For seropositive individuals, we performed stool polymerase chain reaction (PCR) to determine current E. histolytica infection. We performed E. histolytica serological screening of 312 participants. None had a history of E. histolytica infection prior to the study. The overall E. histolytica seropositivity was 6.7% (21/312), which was similar to that found by the rapid plasma reagin test (17/312). We identified current infection in 8 of 20 seropositive participants (40.0%) by stool PCR.Conclusions/SignificanceOur serological screening approach constitutes a potentially practical epidemiological strategy. Active epidemiological surveys, in combination with an effective screening strategy for asymptomatically infected individuals, should be applied to help reduce sexually transmitted E. histolytica infections.  相似文献

Severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) has been transmitted across all over the world, in contrast to the limited epidemic of genetically- and virologically-related SARS-CoV. However, the molecular basis explaining the difference in the virological characteristics among SARS-CoV-2 and SARS-CoV has been poorly defined. Here we identified that host sialoglycans play a significant role in the efficient spread of SARS-CoV-2 infection, while this was not the case with SARS-CoV. SARS-CoV-2 infection was significantly inhibited by α2-6-linked sialic acid-containing compounds, but not by α2–3 analog, in VeroE6/TMPRSS2 cells. The α2-6-linked compound bound to SARS-CoV-2 spike S1 subunit to competitively inhibit SARS-CoV-2 attachment to cells. Enzymatic removal of cell surface sialic acids impaired the interaction between SARS-CoV-2 spike and angiotensin-converting enzyme 2 (ACE2), and suppressed the efficient spread of SARS-CoV-2 infection over time, in contrast to its least effect on SARS-CoV spread. Our study provides a novel molecular basis of SARS-CoV-2 infection which illustrates the distinctive characteristics from SARS-CoV.  相似文献
