Synthesis of novel polyamines in Paracoccus, Rhodobacter and Micrococcus

Abstract The Gram-negative facultative chemolithotroph, Paracoccus denitrificans contains putrescine, cadaverine, agmatine, spermidine, aminopropylcadaverine, spermine, thermospermine and aminopentylnorspermidine. This bacterium has the ability to produce norspermidine from supplemented diaminopropane. The halophile, Paracoccus halodenitrificans is devoid of any polyamines. Neither decarboxylation of ornithine, lysine or arginine, nor triamine synthetic activity from diamines was detected in this halophile. Two Gram-negative facultative photoautotrophs, Rhodobacter sphaeroides and Rhodobacter capsulatus contain putrescine, cadaverine, agmatine and spermidine and can produce norspermidine from supplemented diaminopropane. A Gram-negative eubacterium, Micrococcus cryophilus , contains histamine and homospermidine in addition to putrescine, cadaverine and spermidine. Hence, polyamine distribution patterns and polyamine biosynthetic activities were very different among the four groups of Gram-negative eubacteria examined.     

Assembly of intra- and interspecies hybrid apoferritins

An intraspecies hybrid apoferritin was assembled by mixing subunits of horse heart ferritin, which consists mainly of H-type subunits, and horse spleen ferritin, in which L-type subunits predominate. Interspecies hybrid apoferritins were reconstituted from subunits of human liver-horse spleen ferritins and from rat liver-horse spleen ferritins. All the hybrid ferritins migrated as single zones with electrophoretic mobilities intermediate between those of the parent ferritins. Isoelectric focusing data and immunological patterns were consistent with the view that the reassembled apoferritins were composite molecules that contained subunits from each of the interacting forms. Reconstitution occurred in a random manner, as there was no apparent preference for assembly of homologous subunits. These results suggest that intersubunit interaction domains and recognition mechanisms that dictate formation of the highly specific quaternary structure assumed by this protein are common for different species of ferritins.     

In vitro effects of juvenile hormone analog on wing disc morphogenesis under ecdysteroid treatment in the female-wingless bagworm moth Eumeta variegata (Insecta: Lepidoptera, Psychidae)

Female adults of the bagworm moth, Eumeta variegata, lack wings completely, whereas male adults of this species have functional wings. We previously found that ecdysteroid induces apoptotic events in the female wing rudiment of E. variegata in vitro, whereas the male wing discs cultured with 20-hydroxyecdysone (20E) underwent apolysis and then cell differentiation. To investigate whether juvenile hormone (JH) in involved in sex-specific cellular response to ecdysteroid during wing development between sexes of E. variegata, we tested the effects of juvenile hormone analog (JHA), methoprene, and 20E on wing disc morphogenesis between sexes in vitro. Using transmission electron microscopy (TEM), we found that both higher concentration of JHA (5 μg/ml) and 20E (1 μg/ml) addition induced cell death (apoptosis) in the male wing discs but not induced cell death in the female wing rudiments in vitro in E. variegata. These culture experiments clearly detected the differential responses of wing discs to JHA under ecdysteroid treatment between sexes. We propose two important hypotheses: (1) JH is not significantly involved in the suppression of the female wing rudiment morphogenesis under 20E treatment, (2) female wing rudiment has lost the ability for cell proliferation in response to the stimulus of 20E.     

Cultural properties of a luminous mushroom,Mycena chlorophos

Cultural conditions on mycelial growth and fruit-body formation ofMycena chlorophos were studied. The optimum temperature of the mycelial growth was 27°C and the optimum initial pH of medium was 4.0. Peptone agar medium was suitable for the spawn culture. Compost medium containing rice bran at 10% (fw/fw) was appropriate for fruitbody formation in the Petri dish. Light was essential for initiation of primordia, and low-temperature treatment induced fruit-body formation effectively. The optimum conditions for fruit-body formation were found to be the cultivation at 27°C for 4 wk and continued cultivation for 3 wk under illumination at an intensity above 0.2 lx and at 21°C after casing with moist compost powder. In the fruit-bodies obtained, the maximum photosensitive wavelength of luminescence was 522 nm and the optimum temperature for emission was 27°C. The luminescence of a fruit-body was observed for about 3 d consecutively at 21°C.     

Adenosine 5'-(gamma-thio) triphosphate (ATPgammaS) stimulates both P2Y receptors linked to inositol phosphates production and cAMP accumulation in bovine adrenocortical fasciculata cells

The study was aimed to investigate the existence of at least two kinds of P2Y receptors linked to steroidogenesis in bovine adrenocortical fasciculata cells (BAFCs). Extracellular nucleotides facilitated steroidogenesis in BAFCs. The potency order was UTP > adenosine 5'-(gamma-thio) triphosphate (ATPgammaS) > ATP > 2-methylthio ATP (2MeSATP) > adenosine 5'-(beta-thio) diphosphate (ADPbetaS) > alpha,beta-methylene ATP (alpha,beta-me-ATP), beta,gamma-methylene ATP (beta,gamma -me-ATP). ATPgammaS (10-100 microM) remarkably stimulated both total inositol phosphates (IPs) production and cyclic AMP (cAMP) accumulation. Competitive displacement experiments by using [35S]ATPgammaS as a radioactive ligand in BAFCs showed that the potency under these unlabelled ligands was ATPgammaS > ATP > ADPbetaS > 2MeSATP > UTP > alpha,beta-me-ATP, beta,gamma-me-ATP. These suggest that two different binding sites of [35S]ATPgammaS, namely P2Y receptors, exist in BAFCs, and that these receptors are linked to steroidogenesis via distinct second messenger systems in the cells.     

Polyamines of primitive apterygotan insects: springtails, silverfish and a bristletail

Polyamines extracted from whole bodies of four springtails, Tomocerus ishibashii, Hypogastrura communis, Sinella cruviseta and Folsomia candida, a bristletail, Pedetontus nipponicus, and two silverfish, Lepisma saccharina and Thermobia domestica, were analyzed by high-performance liquid chromatography and gas chromatography. All seven apterous insect species contained putrescine, cadaverine and spermidine as the common major polyamines, detected at the level of micromol/g wet mass. T. ishibashii also contained spermine, S. cruviseta contained norspermidine and norspermine and H. communis, F. candida and P. nipponicus contained diaminopropane, norspermidine and norspermine, as minor polyamines above the detection limit (0.01 micromol/g wet mass). The occurrence of diaminopropane, norspermidine, norspermine, spermine and thermospermine was confirmed in L. saccharina and T. domestica. The novel polyamines norspermidine, norspermine and thermospermine, widespread in higher insects, were also distributed within the primitive apterygotan insects.     

Identification of 4-methylspinaceamine--a Pictet-Spengler condensation reaction product of histamine with acetaldehyde--in human urine

This study reports the first identification of 4-methylspinaceamine (4-MSPA)-a Pictet Spengler condensation reaction product of histamine with acetaldehyde-in human urine. 4-MSPA was identified and quantified as follows: the target compound was partially purified by solvent extraction from a urine sample spiked with N-methylpiperazine (N-MP) as an internal standard, then derivatized to a naphthylthiourea derivative with 1-naphthylisothiocyanate (NITC) and finally analyzed by HPLC. For verification, 4-MSPA was also analyzed by ion spray-mass spectrometry (IS-MS), using 4-MSPA-d4 as an internal standard. The amount of 4-MSPA in human urine varied between individuals and from day-to-day, ranging from undetectable to 0.80 nmol/mL.
