Attempts were made to form 5′-mononucleotides from non-proliferating cells of various kinds of yeasts. It was found that yeasts were devided into four groups according to their ability to excrete compounds absorbing at 260 mμ, the first of which excreted UV-absorbing materials at the acid pH (I), the second at the alkaline pH (II), the third both at the acid and alkaline pHs (III), and the fourth showed weak ability to excrete UV-absorbing materials at both pHs (IV). In general, 3′-mononucleotides were excreted at the acid pH and 5′-mononucleotides at the alkaline pH. The most dominant factor influencing the oxidation-reduction potentials (E) in the cultured system was oxygen tension. H was an useful index to express the degree of oxygen supply in place of dissolved oxygen (PL) under a limited oxygen supply. The conversion of microbial products caused by the change in oxygen supply was clearly analyzed by the use of E value. Bacillus subtilis excreted lactic acid at the E value ?220 mV, 2,3-butyleneglycol at ?195 mV and acetoin at ?160mV as the main product. E also gave the significant information concerning the changes in cell’s respiration. Cyanide at the concentration of 10?5m, azide at 10?3m and 2,4-dinitrophenol (DNP) at 10?2m inhibited cell respiration causing the decrease in E and the increase in PL, and DNP at 0.4×10?3m promoted oxygen uptake of the cells causing the decrease in both E and PL. 相似文献

To establish a novel process for the production of l-glutamic acid from n-paraffins, a glycerol auxotroph GL-21, a new type mutant, was successfully obtained from Corynebacterium alkanolyticum No. 314 by treatment with N-methyl-N′-nitro-N-nitrosoguanidine. This auxotroph required glycerol for its growth regardless of the carbon source used.At 72 hr, this mutant GL-21 produced about 40 mg/ml of l-glutamic acid from n-paraffins in the culture broth at 0.01 per cent addition of glycerol in the absence of penicillin.A thiamine auxotroph, a biotin auxotroph and an oleic acid auxotroph were also obtained by a similar technique, but these auxotrophs were found to be inapplicable for the production of l-glutamic acid from n-paraffins. 相似文献

A novel process for the microbial production of l-glutamic acid on an industrial scale was successfully established by using a glycerol auxotroph.The most suitable carbon source for producing L-glutamic acid was n-paraffins (C13–C15). The production of L-glutamic acid was not affected by a large amount of biotin or oleic acid in the absence of penicillin, and occurred maximally at the glycerol concentration of 0.02% at pH 6.6. The most effective temperature was 28°C.Under optimal conditions in a 200 liter fermentor, the mutant produced 72 g/liter of L-glutamic acid. On the other hand, the parent produced 53 g/liter of L-glutamic acid in the presence of penicillin.It is believed that the low productivity of L-glutamic acid by the parent strain was mainly due to the occurrence of the marked decrease in the viable cell counts at the later phase of the fermentation caused by the action of penicillin added. 相似文献

The combination of flow injection analysis and the electrochemical method was investigated for measuring the organic acid content of citrus fruits. The controlled-potential four-electrode method was used for conductance measurements. The instrumentation of the measuring apparatus used in this study is defined. The optimum conditions of the flow system were as follows: injection volume 35 μ1, mixing coil length 100 cm, flow rate 20ml/min and dilution ratio of samples 1:130. A linear correlation between the peak conductivity and the organic acid content was obtained with high correlation coefficient (r=0.992). It is possible to analyze about 50 samples within an hour using the method, and the coefficient of variation with any given juice for 20 assays was less than 0.5%. 相似文献

The garlic-derived antibacterial principle, alk(en)yl sulfinate compounds, has long been considered as very short-lived substance. However, there are some data showing a rather more stable nature of allicin. We determined here the thermostability of allicin by a systematic analyses employing chemical quantification and an antibacterial activity assay. Allicin in an aqueous extract of garlic was degraded stoichiometrically in proportion to the temperature; we estimated the half-life of allicin to be about a year at 4 °C (from 1.8 mg/ml to 0.9 mg/ml) and 32 d at 15 °C, but only 1 d at 37 °C (from 2.0 mg/ml to 1.0 mg/ml). The half-life values for antibacterial activity showed a similar trend in results: 63 d or more at 4 °C for both antibacterial activities, 14 d for anti-staphylococcal activity, and 26 d for anti-escherichia activity at 15 °C, but only 1.2 d and 1.9 d for the respective activities at 37 °C. Such antibacterial activities were attributable to the major allicin, allyl 2-propenylthiosulfinate. Surprisingly, the decline in the quantity of allicin was not accompanied by its degradation; instead, allicin became a larger molecule, ajoene, which was 3-times larger than allicin. 相似文献

We have examined the distribution of nicotinamide adenine dinucleotide phosphate-diaphorase (NADPH-d) and the calcium binding proteins (CBPs), calbindin D-28k (CB), calretinin (CR) and parvalbumin (PV), in the trigeminal nucleus oralis (Sp5O). NADPH-d was detected by histochemistry while CBP was detected by immunohistochemistry. NADPH-d-positive neurons were distributed in the medial rostro-dorsomedial part (RDMsp5O) and dorsomedial part (DMsp5O) of Sp5O, and the rostrolateral part of the nucleus of the solitary tract (NTS). CB- and CR-positive neurons were mainly distributed in the dorsal part of Sp5O. In contrast, PV-positive neurons were mainly distributed in the ventral part of Sp5O. NADPH-d colocalized with CB (40%) and CR (20%) but not with PV in neurons of DMsp5O/NTS. The mean cell sizes of neurons in RDMsp5O were larger than those in DMsp5O/NTS. PV-positive neurons were larger than NADPH-d-positive neurons. NADPH-d-, CB- and CR-positive neurons were generally small in RDMsp5O and DMsp5O/NTS. Few neurons were retrogradely labeled in RDMsp5O and DMsp5O from the thalamus, when numerous labeled neurons were in the principal and interpolar nuclei. These data indicate that NADPH-d histochemistry and CB, CR and PV immunohistochemistry identify a discrete cell population in Sp5O. Those labeled neurons in RDMsp5O and DMsp5O/NTS were considered to be involved in sensorimotor reflexive function of the intra-oral structures. 相似文献

A lytic enzyme which was capable of lysing cells of Streptococcus mutans was purified from the culture filtrate of Streplomyces griseus H–402 by Amberlite CG–50 treatment, CM-cellulose and hydroxylapatite column chromatographies, and Sephadex G–150 gelfiltration. The lytic enzyme was obtained in a crystalline form which was homogeneous in polyacrylamide gel electrophoresis. The molecular weight was estimated to be 2×104 by the thin-layer gel-filtration method on Sephadex G–75, and 2.3 × 104 by the method of polyacrylamide gel electrophoresis in the presence of sodium dodecyl sulfate. The enzyme was found to be a N-acetylmuramidase whose activity was lost by N-bromosuccinimide as an inhibitor. 相似文献

It has been reported by several persons, including one of the authors, that DPNH obtained by the electrolytic reduction at controlled potentials is not fully active in respect to the reaction with alcohol dehydrogenase system. But statements concening the percentage of activity and the conditions which affect the activity were not identical. This article deals with these problems by reducing DPN electrolytically under different conditions followed by enzymatic examinations. The electrolytic reductions Of TPN and cytochrome c were also performed. Though the data presented are rather complicated, the most active DPNH was prepared in tripolyphosphate buffer with platinum electrode at ?2.0 volt vs. S.C.E., under ice-cooling. During the course of investigation on organophosphorus insecticides, many organophosphorus compounds having methyl-, methyl-chloro-, methyl-nitro-, methyl-cyano-, methyl-thiocyano-, methoxy-nitro-, acyloxy-nitro-, and chloro-cyano-phenyl groups were prepared and their biological activities tested. As the results of these studies, two compounds were found as new low toxic organophosphorus insecticides, namely, O,O-dimethyl-O- (3-methyl-4-nitro-phenyl) and O,O-dimethyl-O- (4-cyanophenyl) phosphorothioates. 相似文献
