When Hiproly barley callus was placed on an auxin-free Murashige and Skoog’s medium, adventitious roots were formed. The contents of S-adenosylmethionine (SAM) and 1-methylhistidine increased gradually in the callus throughout the root differentiation, along with increases in the levels of their precursors, methionine, histidine, and adenosine 5′-triphosphate. In the callus after auxin withdrawal, the addition of methionine resulted in rapid increases in the content of SAM and the incorporation of [methyl-14C]methionine into SAM. The accumulation of SAM was closely related to the elevation in methionine level in the callus. 1-Methylhistidine also increased greatly in the callus treated with methionine, but the addition of histidine had little effect on the change in 1-methylhistidine in the callus. 相似文献

Seasonal fluctuations in herbivore densities are affected by various factors. Solidago altissima was introduced from North America and has spread widely over Japan. Then, an aphid Uroleucon nigrotuberculatum and a lace bug Corythucha marmorata, both associated with S. altissima, invaded Japan in the late 1980s and 2000, respectively. In Miyagi, northeastern Japan, U. nigrotuberculatum and C. marmorata were established in the 1990s and 2012, respectively. We conducted periodic surveys of U. nigrotuberculatum in 2003–2004 and 2015–2016, and of C. marmorata in 2016 in Miyagi. Then, a competition experiment was performed to examine the relationship between them. In 2003, the average maximum daily temperature did not exceed 30 °C and the aphid was continuously confirmed on S. altissima during the census period. In other census years, the temperature exceeded 30 °C and the aphid disappeared in summer. At these sites, C. marmorata was continuously found on S. altissima. The presence of C. marmorata did not negatively affect U. nigrotuberculatum in the competition experiment. In addition, peak densities of U. nigrotuberculatum were higher in 2015–2016 than in 2003–2004. Based on these results, we conclude that U. nigrotuberculatum populations were at least partly affected by high temperature during the summer, but not by C. marmorata invasion in Miyagi. 相似文献

To improve the reliability of sensory evaluation, it is necessary to minimize response bias. In this study, we investigated the effect of a warm‐up sample on time–intensity evaluation of after‐flavor. Untrained panelists evaluated continuously perceived intensity of bitterness and retronasal aroma, using four types of samples (two brands of coffee beverages × two types of lids). Half of participants evaluated bitterness intensity in the first session and retronasal aroma intensity in the second session, whereas the other half evaluated them in the opposite order. Each session consisted of four trials, and we regarded the first trial sample of each session as a warm‐up. Results revealed that perceived intensity was significantly lower in the first trial than in the following trials, and that the time courses of the second, third, and fourth trials were similar except when participants evaluated bitterness intensity in the second session. In other words, the use of a warm‐up sample stabilized the performance of time–intensity evaluation of bitterness and retronasal aroma in the first session. Furthermore, no warm‐up sample was needed in the second session when participants evaluated bitterness intensity, but a warm‐up was needed when they evaluated retronasal aroma intensity.
Practical application
Based on the results of this study, we concluded that the use of a warm‐up sample stabilized performance in time–intensity evaluation of after‐flavor by untrained panelists. We proposed that in order to obtain reliable performance in time–intensity evaluation of after‐flavor, untrained panelists should be provided a training trial using warm‐up sample before staring the test trials. 相似文献164.
We examined the effects of environmentally friendly (EF) farming and landscape factors on the abundances of major spider guilds and a rice pest, the small brown planthopper (SBPH), Laodelphax striatellus (Fallén), in a Japanese rice paddy ecosystem. The abundances of all spider guilds increased by EF farming, whereas different spider guilds showed contrasting responses to the size of the forest within 200 m of the fields. The abundances of ground spiders and horizontal web weavers increased with increasing forest area, unlike small ground spiders, whose abundance decreased. The abundances of SBPH nymphs and adults decreased by EF farming but responded differently to landscape: nymphs decreased with increasing forest area, but adults did not. Further analysis indicated a potential negative relationship between web-weaving spiders and SBPH nymphs. Our findings suggest that the size of the forest adjacent to fields is an important determinant of spider guild composition and pest abundance. 相似文献

The effect of 1-hexanol on spectral properties and the processes of energy transfer of the green gliding photosynthetic bacterium Chloroflexus aurantiacus was investigated with reference to the baseplate region. On addition of 1-hexanol to a cell suspension in a concentration of one-fourth saturation, a specific change in the baseplate region was induced: that is, a bleach of the 793-nm component, and an increase in absorption of the 813-nm component. This result was also confirmed by fluorescence spectra of whole cells and isolated chlorosomes. The processes of energy transfer were affected in the overall transfer efficiency but not kinetically, indicating that 1-hexanol suppressed the flux of energy flow from the baseplate to the B806-866 complexes in the cytoplasmic membranes. The fluorescence excitation spectrum suggests a specific site of interaction between bacteriochlorophyll (BChl) c with a maximum at 771 nm in the rod elements and BChl a with a maximum at 793 nm in the baseplate, which is a funnel for a fast transfer of energy to the B806-866 complexes in the membranes. The absorption spectrum of chlorosomes was resolved to components consistently on the basis, including circular dichroism and magnetic circular dichroism spectra; besides two major BChl c forms, bands corresponding to tetramer, dimer, and monomer were also discernible, which are supposed to be intermediary components for a higher order structure. The DNA-binding activity of a tobacco heat shock factor (HSF) was induced by heat treatment (37–40 °C) of a cell-free extract that contained extra-nuclear fraction, but not in an extract of isolated nuclei. These observations suggest that an inactive form of HSF can directly recognize and transduce the heat shock signal and that such transduction requires components of the extranuclear fraction. Addition of ATP or of most other nucleoside triphosphates reduced the binding of the HSF to the heat shock element (HSE) in the same extract, and removal of ATP by dialysis from the extract restored the ability of the HSF to bind to DNA. The restored activity of the HSF could be eliminated again by a second addition of ATP. Our observations provide the first example of the involvement of ATP in the regulation of the reversible changes in HSF that control its ability to bind to HSEs in a cell-free extract.Abbreviations AMP-PNP

The binding of IgM from a rheumatoid factor (RF-IgM) to IgG from 12 animal species was analyzed by an ELISA system. The RF-IgM bound various animal IgG with dissimilar affinities. The binding of RF-IgM to animal IgG was inhibited by addition of protein A, which binds some animal IgG by recognizing the junctional site on CH2-CH3 domains in the Fc region. As previously reported, no significant correlation was observed between the binding of RF-IgM to IgG and the content of galactose-free oligosaccharides, which is increased in IgG of rheumatoid arthritis patients or autoimmune mice. We suggest that the crucial epitope of IgG for RF-IgM binding is not the oligosaccharide structure generated specifically in IgG of autoimmune diseases but that RF-IgM may recognize a certain protein conformation of a region in IgG near the binding site of protein A. 相似文献

We examined effects of fibroblasts of different origin on long-term maintenance of xenotransplanted human epidermal keratinocytes. A suspension of cultured epidermal cells, originating from adult human trunk skin, was injected into double mutant immunodeficient (BALB/c nu/scid) mice subcutaneously, with or without cultured fibroblastic cells of different origin. At one week after transplantation, the epidermal cells generated epidermoid cysts consisting of human epidermis-like tissue. When the epidermal cells were injected alone or together with fibroblastic cells derived from human bone marrow, muscle fascia, or murine dermis, organized epidermoid cysts regressed within 6 weeks. In contrast, when the epidermal cells were injected together with human dermal fibroblasts, generated epidermoid cysts were maintained in vivo for more than 24 weeks. Histological examination showed that the reorganized epidermis, after injection of both epidermal keratinocytes and dermal fibroblasts, retained normal structures of the original epidermis during 6 to 24 weeks after transplantation. The results indicate that human dermal fibroblasts facilitate the long-term maintenance of the reorganized epidermis after xenotransplantation of cultured human epidermal keratinocytes by supporting self renewal of the human epidermal tissue in vivo. 相似文献

Recently, evidence has accumulated that mutations in DNA repair genes might be associated with certain steps in carcinogenesis. The DNA polymerase gene is one of the DNA repair genes, and mutations in it have been detected in 83% of human colorectal cancers. To assess the involvement of polymerase gene mutations in the development of human prostate cancers, we performed sequence analyses of human DNA samples.
