Nup98 is a mobile nucleoporin that forms distinct dots in the nucleus, and, although a role for Nup98 in nuclear transport has been suggested, its precise function remains unclear. Here, we show that Nup98 plays an important role in Crm1-mediated nuclear protein export. Nuclear, but not cytoplasmic, dots of EGFP-tagged Nup98 disappeared rapidly after cell treatment with leptomycin B, a specific inhibitor of the nuclear export receptor, Crm1. Mutational analysis demonstrated that Nup98 physically and functionally interacts with Crm1 in a RanGTP-dependent manner through its N-terminal phenylalanine-glycine (FG) repeat region. Moreover, the activity of the Nup98-Crm1 complex was modulated by RanBP3, a known cofactor for Crm1-mediated nuclear export. Finally, cytoplasmic microinjection of anti-Nup98 inhibited the Crm1-dependent nuclear export of proteins, concomitant with the accumulation of anti-Nup98 in the nucleus. These results clearly demonstrate that Nup98 functions as a novel shuttling cofactor for Crm1-mediated nuclear export in conjunction with RanBP3. 相似文献

α-Ionone, α-methylionone, and α-isomethylionone were converted by Aspergillus niger JTS 191. The individual bioconversion products from α-ionone were isolated and identified by spectrometry and organic synthesis. The major products were cis-3-hydroxy-α-ionone, trans-3-hydroxy-α-ionone, and 3-oxo-α-ionone. 2,3-Dehydro-α-ionone, 3,4-dehydro-β-ionone, and 1-(6,6-dimethyl-2-methylene-3-cyclohexenyl)-buten-3-one were also identified. Analogous bioconversion products from α-methylionone and α-isomethylionone were also identified. From results of gas-liquid chromatographic analysis during the fermentation, we propose a metabolic pathway for α-ionones and elucidation of stereochemical features of the bioconversion. 相似文献

Conditions for the production of nitrite and nitrous oxide by an obligate methanotroph, Methylosinus trichosporium (OB 3b), were studied. The rate of nitrite production (V NO2-) was correlated with the concentration of ammonia up to 20 mM in the presence of sufficient amounts of oxygen and inversely correlated with the amounts of methane in the system. The rate of nitrous oxide (N2O) production (V N2O) was correlated positively with V NO2- and the amount of nitrite produced and inversely with the oxygen concentration in the system. Nitrite started to disappear when either oxygen or methane or both were depleted, but only a part of the loss could be accounted for by an increase in N2O. Maximum rates of nitrite and N2O production by Ms. trichosporium were 6.9 X 10(-16) and 2.2 X 10(-17) mol . cell-1 X day-1, respectively. These values are about 0.2 and 1.6% of the values for Nitrosomonas europaea. Therefore, production of nitrite and N2O by methanotrophs in aquatic environments may not be as significant as that of Nitrosomonas. 相似文献

Simple sequence repeat (SSR) markers were developed and characterized for Neolitsea sericea (Bl.) Koidz. (Lauraceae). Out of 196 designed primer pairs, a total of 144 pairs showed amplification, of which 44 had clear and stable chromatograms. Polymorphism of these 44 loci was tested using 32 individuals sampled from a single population of N. sericea. The number of alleles and the polymorphism information content varied from 3 to 12 and 0.271 to 0.853, respectively. A significant departure from the Hardy‐Weinberg equilibrium was observed in one of the 44 loci. These SSR markers are useful for population genetic studies and parentage analysis in N. sericea, which is one of the most common evergreen species in coastal Pinus thunbergii forests in central‐western Japan. 相似文献

Zooplankton collections were made with a multiple opening/closingnet and environmental sensing system (MOCNESS) both day andnight in warm-core ring 82B in the North Atlantic Ocean andat night in the Slope Water between March and August 1982. Speciessuccession of euphausiids in 82B was presented during the lifespanof this warm-core ring, aiming at providing basic informationon possible response of North Atlantic marine ecosystem to globalwarming. Species succession of euphausiids (32 species) in thislong-lived warm-core ring was investigated. Major physical changesof 82B occurred in March to April by convective mixing and thermostadcooling, in April/May by stratification of the surface watersand in August by the interaction with Gulf Stream. Substantialchanges in species composition were observed that correspondedto these physical changes. Four different patterns were foundin temporal change in abundance of warm-water species. Therewere species that decreased in number from March to August,species that decreased from March to June, but increased inAugust, species that increased from Match to August and speciesthat showed no systematic trend. These differences may be attributableto a species tolerance to the thermostad temperature decreaseand their vertical distribution. There was also a large changefrom April to June with ascendance of the transition species,Thysanoessa gregaria. Cold-water species had variable patternsof abundance in 82B and occurred more abundantly in the SlopeWater than in the ring. The monthly percentage decrease in theabundance of warm-water species in 82B was high compared withthat of cold-water species in cold-core rings as a result ofthe more rapid changes in the physical structure and the shorterlifetimes of warm-core rings in the western North Atlantic. 相似文献

Specific binding of antimicrobial peptides to titanium (Ti) surfaces may serve to prevent biofilm formation, leading to a reduction in peri-implantitis. This study evaluated the binding behavior of conjugated molecules consisting of antimicrobial and hexapeptidic Ti-binding peptides (minTBP-1) using the quartz crystal microbalance (QCM-D) technique, and investigated the effect of modification of Ti surfaces with these peptides on the bioactivity of Porphyromonas gingivalis. Four kinds of peptide were prepared: histatin 5 (DSHAKRHHGYKRKFHEKHHSHRGY), minTBP-1 + histatin 5 (RKLPDAPDSHAKRHHGYKRKFHEKHHSHRGY), lactoferricin (FQWQRNMRKVR), and minTBP-1 + lactoferricin (RKLPDAPGGFQWQRNMRKVR). The QCM-D analysis demonstrated that significantly larger increases in peptide adsorption were observed in the conjugated peptides than in antimicrobial peptides alone. In addition, ATP activity in P. gingivalis in peptide-modified specimens significantly decreased compared to that in the Ti control. These results indicate that surface modification with conjugated molecules consisting of antimicrobial and Ti-binding peptides is a promising method for reduction of biofilm formation on Ti surfaces. 相似文献

Our lead compound for a phosphoinositide 3-kinase (PI3K) inhibitor (1) was metabolically unstable because of rapid glucuronidation of the phenol moiety. Based on structure–activity relationship (SAR) information and a FlexSIS docking simulation score, aminopyrimidine was identified as a bioisostere of phenol. An X-ray structure study revealed a hydrogen bonding pattern of aminopyrimidine derivatives. Finally, aminopyrimidine derivatives 33 showed strong tumor growth inhibition against a KPL-4 breast cancer xenograft model in vivo. 相似文献

A decrease in adiponectin secretion leads to the early stage of atherosclerosis. Discoidal high-density lipoproteins (HDL) accept the cholesterol that effluxes from cells expressing the ATP binding cassette transporter A1 (ABCA1) in the first step of reverse cholesterol transport (RCT). Recently, a new therapeutic strategy involving reconstituted (r)HDL has been shown to enhance RCT. Therefore, we hypothesized that adiponectin may increase the efflux associated with ABCA1 and also enhance rHDL-induced efflux in human embryonic kidney 293 (HEK293T) cells. We transfected adiponectin receptor 1 and 2 (AdipoR1 and AdipoR2) cDNA into cells. The transfected cells were labeled with [3H]cholesterol following cholesterol loading with or without adiponectin for 24 h. The levels of cholesterol efflux were analyzed using a liquid scintillation counter. Treatment with adiponectin was associated with significantly higher levels of efflux in AdipoR1- and AdipoR2-transfected cells. Interestingly, rHDL-induced cholesterol efflux was enhanced in the presence of adiponectin. The down-regulation of adiponectin receptors using short-hairpin RNA decreased rHDL-induced cholesterol efflux with the down-regulation of ABCA1. In summary, adiponectin and its receptors increased cholesterol efflux and also enhanced rHDL-induced efflux at least partially through an ABCA1 pathway. These results suggest that adiponectin may enhance the RCT system and induce an anti-atherogenic effect. 相似文献

In Archaea, splicing endonuclease (EndA) recognizes and cleaves precursor RNAs to remove introns. Currently, EndAs are classified into three families according to their subunit structures: homotetramer, homodimer, and heterotetramer. The crenarchaeal heterotetrameric EndAs can be further classified into two subfamilies based on the size of the structural subunit. Subfamily A possesses a structural subunit similar in size to the catalytic subunit, whereas subfamily B possesses a structural subunit significantly smaller than the catalytic subunit. Previously, we solved the crystal structure of an EndA from Pyrobaculum aerophilum. The endonuclease was classified into subfamily B, and the structure revealed that the enzyme lacks an N-terminal subdomain in the structural subunit. However, no structural information is available for crenarchaeal heterotetrameric EndAs that are predicted to belong to subfamily A. Here, we report the crystal structure of the EndA from Aeropyrum pernix, which is predicted to belong to subfamily A. The enzyme possesses the N-terminal subdomain in the structural subunit, revealing that the two subfamilies of heterotetrameric EndAs are structurally distinct. EndA from A. pernix also possesses an extra loop region that is characteristic of crenarchaeal EndAs. Our mutational study revealed that the conserved lysine residue in the loop is important for endonuclease activity. Furthermore, the sequence characteristics of the loops and the positions towards the substrate RNA according to a docking model prompted us to propose that crenarchaea-specific loops and an extra amino acid sequence at the catalytic loop of nanoarchaeal EndA are derived by independent convergent evolution and function for recognizing noncanonical bulge-helix-bulge motif RNAs as substrates. 相似文献

Palmerolides D-G are new bioactive macrolides isolated from the Antarctic tunicate Synoicum adareanum and are related to the melanoma-selective cytotoxin palmerolide A. Most of these palmerolides are potent V-ATPase inhibitors and have sub-micromolar activity against melanoma. Though palmerolide A remains the most potent of this series of natural products against mammalian V-ATPase, recent data suggests that palmerolide D is the most potent against melanoma. A comparison of the bioactivity data obtained for these natural product palmerolides has provided insight into the substructures necessary to retain V-ATPase inhibition and cytotoxic activity. 相似文献
