THE ACTIVATION OF THIAMINE DIPHOSPHATASE BY ATP IN RAT BRAIN 总被引:1,自引:1,他引:0
- 1 Thiamine diphosphatase (TDPase) of brain was activated by a low concentration of ATP.
- 2 In thiamine-deficient rats TDPase activity in the brain increased significantly relative to that in pair-fed animals, while in liver it decreased by the same amount as in the pair-fed controls.
- 3 Liver TDPase was localized almost entirely in the soluble fraction, but in the brain it was bound to insoluble protein. On treatment with Triton X-100 brain TDPase activity increased.
- 4 The ATP content of the brain of deficient rats, but not the ADP or AMP content, was significantly higher than in the control group. The level of inorganic phosphate in the brain and spinal cord of deficient animals was elevated markedly, while that of P-creatine was unchanged.
- 5 The possible roles of brain TDPase in relation to nerve conduction and the blood-brain barrier are discussed.

Light-induced chloroplast rearrangements were studied for leavesof 17 species of terrestrial plants (8 species of monocotyledonaeand 9 of dicotyledonae) by measuring their absorbance changeswith a dual-wavelength positional scanner. Almost all of theseleaves examined underwent two types of absorbance change; absorbanceincrease on illumination with weak blue light and absorbancedecrease with strong light. The maximal increase and decreaseand the light intensities for these maximal responses as wellas the intensity for mutual compensation of these opposite responseswere determined and compared for these species. A new method for isolation of polyamines from tissues was developed and compared with the butanol extraction method which has been widely used for quantitative determination of polyamines. In the new method protein-free tissue extracts are applied to a small Dowex-50 column. The column is washed with appropriate buffers to remove ninhydrin-positive contaminants and the polyamines are eluted. With this method, the overall recovery of polyamines, after separation by paper electrophoresis and subsequent colorimetric determination with ninhydrin, is always over 90% (average 95%). This method is much better than the butanol method, which gives variable recoveries of 70–90%. The new method also has the advantages over the butanol method that the isolated polyamines are purer and the procedure is simpler. 相似文献

The effect of hypophysectomy, growth hormone (GH) and an amino acid-glucose mixture on the regenerative ability of the hypophysectomized Triturus pyrrhogaster yielded the following results:
- 1 The survival time of hypophysectomized newts can be prolonged substantially by the sulfamide application.
- 2 Although the limb regeneration in the hypophysectomized newt is retarded as compared with that of the pituitary intact control, it finally completes morphogenetic process under such conditions of prolonged survival.
- 3 The injection of 100 μg of GH restored the speed of regeneration of pituitary-deprived limbs to almost a normal level.
- 4 Injections of the amino acid-glucose mixture also promoted the limb regeneration in hypophysectomized newts. However, initial delay in regeneration to the time of bud appearance was not restored by the nutrients.
