Vitamin B6 is synthesized by green Cytisus scoparius callus and green Phellodendron amurense callus cultured on Linsmaier and Skoog Agar-medium with 10?5m of ±-naphthaleneacetic acid (NAA) and 10?6 m of 6-benzyladenine (BA). Even when thiamine and inositol were omitted from this medium, the growth and vitamin B6 content of Cytisus scoparius callus did not change. Vitamin B6 contents of clones of the calluses varied and were unstable during long-term subculture. Clonal selection was repeated to obtain stable strains with high vitamin B6 content, and the vitamin B6 content of one strain of green Cytisus scoparius callus became 4-times higher than that of the green leaves. 相似文献

Methyl eicosapentaenoate (methyl 5,8,11,14,17-eicosapentaenoate) was subjected to autoxidation and methylene blue sensitized photooxidation. Methyl eicosapentaenoate monohydroperoxides, the primary products of the autoxidation and photosensitized oxidation, were isolated by silica gel column chromatography, and characterized by ultraviolet, infrared and nuclear magnetic resonance spectra. The isomeric composition of the monohydroperoxides were determined by gas chromatography-mass spectrometry as follows: the 5-, 8-, 9-, 11-, 12-, 14-, 15- and 18-isomers (autoxidation), and the 5-, 6-, 8-, 9-, 11-, 12-, 14-, 15-, 17- and 18-isomers (photosensitized oxidation). Fungicidal activities of S-alkyl S′-p-substituted benzyl N-3-pyridyldithiocarbonimidates toward Coniothyrium diplodiella, Scleotinia sclerotiorum (on agar medium) and Sphaerotheca fuliginea (on pot test) were shown. The activities varied with the change of the S-alkyl group and were maximized at certain numbers of carbon atoms. A different pattern was observed in the activity toward S. fuliginea when the carbon number was 8 or more. Bulkiness of the S-alkyl group also appeared to influence the activities. The steric factor of the p-alkyl substituents mainly influenced the activity toward S. fuliginea up to the tert-butyl analog. The activities toward C. diplodiella and S. sclerotiorum increased with the increase in the bulkiness and the hydrophylicity of the p-substituent. Rm values were determined on reversed phase TLC and the structure-activity correlations were analyzed against hydrophobic (Rm), electronic (σ) and steric (Es) factors on 34 compounds. 相似文献

The tetradecapeptide of a renin substrate, DRVYIHPFHLLVYS, was used as a substrate for assaying several fungal aspartic and acidic proteinases in the acidic pH range. Aspartic and acidic proteinases froll) Phycomycetes, Mucor and Rhizopus, and Deuteromycotina, Aspergillus and Penicillium, cleaved the tetradecapeptide at its tyrosyl4-isoleucyl5 (Y4-I5),histidyI6-proly7 (H6_P7) and leucyl11-valyl12 (L11-V12) bonds in the acidic pH range, while acidic proteinases type B and type A-I from Scytalidium lignicolumn, and those from Cladosporium and Basidiomycetes, Pycnoporus sanguineus, and the yeast, Rhodotorula glutinis; showed slightly different specificities towards the tetradecapeptide. Pepsin primarily cleaved the valy3-tyrosyl4 (V3-Y4) and leucyl10-leucyl11 (L10-L11) bonds. An extracellular phospholipase D from Actinomadura sp. Strain No. 362 was purified about 430-fold from the culture filtrate. The purified enzyme preparation was judged to be homogeneous on polyacrylamide gel electrophoresis. The molecular weight and isoelectric point of the enzyme were estimated to be about 50,000—60,000 and 6.4, respectively. The enzyme was most active at pH 5.5 and 50°C in the presence of Triton X-100, but showed the highest activity at pH 7.0 and 60 — 70°C in its absence. The enzyme was stable up to 30°C at pH 7.2 and also stable in the pH range of 4.0 to 8.0 on 2 hr incubation at 25°C. With regard to substrate specificity, this enzyme hydrolysed lecithin best among the phospholipids tested. It was activated by Fe3 +, Al3+, Mn2 +, Ca2 +, diethyl ether, sodium deoxycholate and Triton X-100, but was inhibited by cetyl pyridinium chloride and dodecylsulfate. 相似文献

A new binary vector, pZT4B, containing the UDP-N-acetylglucosamine: dolichol phosphate N-acetylglucosamine-1-P transferase (GPT) gene as a selection marker, was constructed. The green fluorescent protein (GFP) gene was inserted into pZT4B, and the resulting plasmid was used in the transformation of Arabidopsis. N-d-Glucosyl-p-aminobenzoic acid has been found to form melanoidins in methanol solution acidified with hydrogen chloride at 25°. From the reaction mixture 5-hydroxymethyl-furfural (HMF) has been isolated and identified as its 2,4-dinitrophenylhydrazone. So, comparisons between the browning reaction of HMF or furfural with aromatic amine and that of the corresponding n-glycosides have been made under the same condition. From the results obtained, it has been shown that, under the described condition, furfural is almost inactive for browning, while on the contrary, HMF is active and plays an important role in the browning reaction. 相似文献

An indole derivative having blue fluorescence was produced in cooked soybean digested at 37°C for 24 hr with an acid proteinase Molsin (optimum pH: 2.8) from Aspergillus saitoi or a usual acid proteinase pepsin (optimum pH: 1.6) from beef stomach. This indole derivative was identical with a condensation product from l-tryptophan and n-hexanal. Based on MS, NMR, IR and UV spectrometry, the condensation product was identified as l-pentyl-2, 3, 4, 9-tetrahydro-lH-pyrido [3, 4-b]-indole-3-carboxylic acid [trivial name: 1-pentyl-l, 2, 3, 4-tetrahydro-2-carboline carboxylic acid-(3)].Data were presented of the formation of the above indole derivative and of the resulting consumption of l-tryptophan and n-hexanal.The possible ocurrence of the formation of Harmala alkaloids, i.e. 2-carboline derivatives, through in vitro digestion of soybean with acid proteinases was discussed.A carbonyl-trapping ability of l-tryptophan was suggested. 相似文献

Soy proteins were incubated with a microbial acid protease (Molsin) under the following condition: substrate concentration, 1%; enzyme-substrate ratio (by weight), 1/100; pH, 2.8; and temperature, 40°C—flavor components and related impurities are removable from crude soy-protein concentrates by their incubation for 2 hr under the above condition. The acid-precipitated fraction of soy protein incubated for 2 hr with Molsin (i.e. 2 hr-proteolyzate) showed the following composition: 10% trichloroacetic acid (TCA) insoluble fraction, 47.52%; 10% TCA soluble peptide fraction, 52.02%; and free amino acid fraction, 0.46%. Gel filtration of the 2 hr-proteolyzate gave an elution pattern showing its molecular weight distribution.In the process of the incubation of the acid-precipitated protein, the 10% TCA insoluble fraction showed increase in amino nitrogen content, its solubility in a phosphate buffer increased to change at 6 hr, and a hydrophobic amino acid share in this fraction increased gradually.In vitro digestibility of the acid-precipitated fraction were improved and the lipoxygenase activity in this fraction decreased through the Molsin treatment.Ultracentrifugal analysis showed a decreasing tendency of the cold-insoluble fraction of soy protein during its incubation with Molsin. Optical rotatory dispersion and circular dichroism study elucidated conformational changes in this fraction during its incubation either with or without Molsin. 相似文献
