Plants endemic to oceanic islands represent some of the most unusual and rare taxa in the world. Enzyme electrophoresis was used to assess genetic diversity within and divergence among all endemic species of a small genus of plants on the Canary Islands. Our results show that the genus Tolpis is similar to many other island groups in having generally low allozyme divergence among species, with the highest divergence found among four groups of endemics. The two rare and highly localized species T. glabrescens and T. crassiuscula are each divergent from all other species in the Canaries. Tolpis coronopifolia is also divergent at allozyme loci; this is the only endemic species that is a self-compatible annual (or weak biennial). A large, morphologically variable species complex consisting of T. laciniata and T. lagopoda together with several named and unnamed morphological variants shows low allozyme divergence among its elements. The evolution of polyploidy from diploid ancestors in situ in oceanic archipelagos is uncommon, but the tetraploid T. glabrescens is an exception. Allozyme data do not implicate any extant diploid Tolpis species as parents of the polyploid. It is possible that T. glabrescens originated early in the evolution of Tolpis in the Canary Islands and that its parents are now extinct. The nonendemic T. barbata shows no greater divergence from the Canary Island endemics than some endemics exhibit among themselves. Both changes in allele frequencies and unique alleles are responsible for genetic divergence among species of Tolpis. 相似文献

Sequence data of the chloroplast gene rbcL were used to estimate the time of the well-known eastern Asian–eastern North American floristic disjunction. Sequence divergence of rbcL was examined for 22 species of 11 genera (Campsis, Caulophyllum, Cornus, Decumaria, Liriodendron, Menispermum, Mitchella, Pachysandra, Penthorum, Podophyllum, and Phryma) representing a diverse array of flowering plants occurring disjunctly in eastern Asia and eastern North America. Divergence times of putative disjunct species pairs were estimated from synonymous substitutions, using rbcL molecular clocks calibrated for Cornus. Relative rate tests were performed to assess rate constancy of rbcL evolution among lineages. Corrections of estimates of divergence times for each species pair were made based on rate differences of rbcL between Cornus and other species pairs. Results of these analyses indicate that the time of divergence of species pairs examined ranges from 12.56 ± 4.30 million years to recent (<0.31 million years), with most within the last 10 million years (in the late Miocene and Pliocene). These results suggest that the isolation of most morphologically similar disjunct species in eastern Asia and eastern North America occurred during the global climatic cooling period that took place throughout the late Tertiary and Quaternary. This estimate is closely correlated with paleontological evidence and in agreement with the hypothesis that considers the eastern Asian–eastern North American floristic disjunction to be the result of the range restriction of a once more or less continuously distributed mixed mesophytic forest of the Northern Hemisphere that occurred during the late Tertiary and Quaternary. This implies that in most taxa the disjunction may have resulted from vicariance events. A new chalcone glycoside, okanin 4′-O-diglucoside, is identified from the ray florets of Coreopsis petrophiloides. The known distribution of anthochlors in Coreopsis indicates that two of the most primitive sections produce in their floral parts complex glucosides of butein (and sulfuretin). The more advanced sections synthesize the monoglucosides of okanin (marein), butein (coreospin) and lanceoletin (lanceolin) and their corresponding aurones. The saturated and 2-enoic primary alcohols and aldehydes, ethanol, 1-propanol, 1-butanol, 3-methyl-1-butanol, 1-hexanol, phenylmethanol, 3-phenyl-1-propanol, 2-propen-1-ol, 2-buten-1-ol, 3-methyl-2-buten-1-ol, 2-hexen-1-ol, 3-phenyl-2-propen-1-ol, ethanal, 1-propanal, 1-butanal, 1-hexanal, phenylmethanal, 3-phenyl-1-propanal, 2-propen-1-al, 2-buten-1-al, 2-hexen-1-al, and 3-phenyl-2-propen-1-al, have been compared under uniform conditions as substrates for the alcohol dehydrogenase enzymes from horse and human liver and from yeast. Kinetic constants (Km arid V) have been measured for each of the substrates with each of the enzymes; equilibrium constants for the various alcohol-aldehyde pairs have also been estimated. The results obtained emphasize the similarities of yeast alcohol dehydrogenase to horse and human liver alcohol dehydrogenase, showing the specificity of yeast alcohol dehydrogenase to be less restricted than formerly believed. In general, the 2-enoic alcohols are better substrates for all three alcohol dehydrogenases than their saturated analogs; on the other hand, saturated aldehydes are better substrates than the 2-enoic aldehydes. Based on these various findings, it is suggested that a more likely candidate than ethanol for the physiological substrate of alcohol dehydrogenase in mammalian systems may well be an unsaturated alcohol, although the wide variety of substrates catalyzed at high rates is not incompatible with a general detoxifying function for alcohols or aldehydes, or both, by alcohol dehydrogenase. 相似文献

The introduction of new approaches for characterizing microbial communities and imaging soil environments has benefited soil microbiology by providing new ways of detecting and locating microorganisms. Consequently, soil microbiology is poised to progress from simply cataloguing microbial complexity to becoming a systems science. A systems approach will enable the structures of microbial communities to be characterized and will inform how microbial communities affect soil function. Systems approaches require accurate analyses of the spatio-temporal properties of the different microenvironments present in soil. Plant‐soil feedback (PSF) theory provides a powerful framework for understanding plant dynamics by integrating growth assays into predictions of whether soil communities stabilise plant–plant interactions. However, we lack a comprehensive view of the likelihood of feedback‐driven coexistence, partly because of a failure to analyse pairwise PSF, the metric directly linked to plant species coexistence. Here, we determine the relative importance of plant evolutionary history, traits, and environmental factors for coexistence through PSF using a meta‐analysis of 1038 pairwise PSF measures. Consistent with eco‐evolutionary predictions, feedback is more likely to mediate coexistence for pairs of plant species (1) associating with similar guilds of mycorrhizal fungi, (2) of increasing phylogenetic distance, and (3) interacting with native microbes. We also found evidence for a primary role of pathogens in feedback‐mediated coexistence. By combining results over several independent studies, our results confirm that PSF may play a key role in plant species coexistence, species invasion, and the phylogenetic diversification of plant communities. 相似文献

A genetic map of Ovis aries (haploid n = 27) was developed with 519 markers (504 microsatellites) spanning ∼3063 cM in 26 autosomal linkage groups and
127 cM (female specific) of the X Chromosome (Chr). Genotypic data were merged from the IMF flock (Crawford et al., Genetics
140, 703, 1995) and the USDA mapping flock. Seventy-three percent (370/504) of the microsatellite markers on the map are common
to the USDA-ARS MARC cattle linkage map, with 27 of the common markers derived from sheep. The number of common markers per
homologous linkage group ranges from 5 to 22 and spans a total of 2866 cM (sex average) in sheep and 2817 cM in cattle. Marker
order within a linkage group was consistent between the two species with limited exceptions. The reported translocation between
the telomeric end of bovine Chr 9 (BTA 9) and BTA 14 to form ovine Chr 9 is represented by a 15-cM region containing 5 common
markers. The significant genomic conservation of marker order will allow use of linkage maps in both species to facilitate
the search for quantitative trait loci (QTLs) in cattle and sheep.

V2/V3 conformational epitope antibodies that broadly neutralize HIV-1 (PG9 and PG16) have been recently described. Since an elicitation of previously known broadly neutralizing antibodies has proven elusive, the induction of antibodies with such specificity is an important goal for HIV-1 vaccine development. A critical question is which immunogens and vaccine formulations might be used to trigger and drive the development of memory B cell precursors with V2/V3 conformational epitope specificity. In this paper we identified a clonal lineage of four V2/V3 conformational epitope broadly neutralizing antibodies (CH01 to CH04) from an African HIV-1-infected broad neutralizer and inferred their common reverted unmutated ancestor (RUA) antibodies. While conformational epitope antibodies rarely bind recombinant Env monomers, a screen of 32 recombinant envelopes for binding to the CH01 to CH04 antibodies showed monoclonal antibody (MAb) binding to the E.A244 gp120 Env and to chronic Env AE.CM243; MAbs CH01 and CH02 also bound to transmitted/founder Env B.9021. CH01 to CH04 neutralized 38% to 49% of a panel of 91 HIV-1 tier 2 pseudoviruses, while the RUAs neutralized only 16% of HIV-1 isolates. Although the reverted unmutated ancestors showed restricted neutralizing activity, they retained the ability to bind to the E.A244 gp120 HIV-1 envelope with an affinity predicted to trigger B cell development. Thus, E.A244, B.9021, and AE.CM243 Envs are three potential immunogen candidates for studies aimed at defining strategies to induce V2/V3 conformational epitope-specific antibodies. 相似文献

Saline stress is a major factor that limits crop yield. Nitric oxide (NO) is functional during plant growth, development, and defense responses. In the present study, the protective role of NO in alleviating saline stress in maize at the physiological and proteomic levels was examined. Our results showed that salt treatment quickly induced NO accumulation and addition of the NO donor S-nitroso-N-acetylpenicillamine (SNAP) efficiently eliminated the inhibitory effect of salt on shoot growth and photosynthesis and inhibited salt-inducible H2O2 accumulation. These effects could be reversed by NO metabolic scavengers and inhibitors. Further proteomic and Western blotting analysis revealed that NO induced G-protein-associated protein accumulation and antioxidant enzymes activities, in addition to activation of defense proteins, energy metabolism, and cell structure/division in salt-treated maize seedlings. Controlling the G-protein status with G-protein activators or inhibitors also affected NO generation and root and stem growth in maize seedlings after saline stress. On the basis of these results, we propose that NO enhances salt tolerance in maize seedlings by enhancing antioxidant enzyme activities and controlling H2O2 levels, and these effects are accompanied by diverse downstream defense responses. During this process, G-protein signaling is an early event that works upstream of NO biogenesis. 相似文献
