Background

Allergic disease can be characterized as manifestations of an exaggerated inflammatory response to environmental allergens triggers. Mite allergen Der-p2 is one of the major allergens of the house dust mite, which contributes to TLR4 expression and function in B cells in allergic patients. However, the precise mechanisms of Der-p2 on B cells remain obscure.

Methodology/Principal Findings

We investigated the effects of Der-p2 on proinflammatory cytokines responses and Toll-like receptor-4 (TLR4)-related signaling in human B cells activation. We demonstrated that Der-p2 activates pro-inflammatory cytokines, TLR4 and its co-receptor MD2. ERK inhibitor PD98059 significantly enhanced TLR4/MD2 expression in Der-p2-treated B cells. Der-p2 markedly activated mitogen-activated protein kinase (MAPK) phosphatase-1 (MKP-1) and decreased p38 phosphorylation in B cells. MKP-1-siRNA downregulated TLR4/MD2 expression in Der-p2-treated B cells. In addition, Der-p2 significantly up-regulated expression of co-stimulatory molecules and increased B cell proliferation. Neutralizing Der-p2 antibody could effectively abrogate the Der-p2-induced B cell proliferation. Der-p2 could also markedly induce NF-κB activation in B cells, which could be counteracted by dexamethasone.

Conclusions/Significance

These results strongly suggest that Der-p2 is capable of triggering B cell activation and MKP-1-activated p38/MAPK dephosphorylation-regulated TLR4 induction, which subsequently enhances host immune, defense responses and development of effective allergic disease therapeutics in B cells.  相似文献

Effects of ammonium on the photosynthetic recovery of Nostoc flagelliforme Berk. et M. A. Curtis were assayed when being rehydrated in low‐K+ or high‐K+ medium. Its photosynthetic recovery was K+ limited after 3 years of dry storage. The potassium absorption of N. flagelliforme reached the maximum after 3 h rehydration in low‐K+ medium but at 5 min in high‐K+ medium. The K+ content of N. flagelliforme rehydrated in high‐K+ medium was much higher than that in low‐K+ medium. The maximal PSII quantum yield (Fv/Fm) value of N. flagelliforme decreased significantly when samples were rehydrated in low‐K+ medium treated with 5 mM NH4Cl. However, the treatment of 20 mM NH4Cl had little effect on its Fv/Fm value in high‐K+ medium. The relative Fv/Fm 24 h EC50 (concentration at which 50% inhibition occurred) value of NH4+ in high‐K+ medium (64.35 mM) was much higher than that in low‐K+ medium (22.17 mM). This finding indicated that high K+ could alleviate the inhibitory action of NH4+ upon the photosynthetic recovery of N. flagelliforme during rehydration. In the presence of 10 mM tetraethylammonium chloride (TEACl), the relative Fv/Fm 24 h EC50 value of NH4+ was increased to 46.34 and 70.78 mM, respectively, in low‐K+ and high‐K+ media. This observation suggested that NH4+ entered into N. flagelliforme cells via the K+ channel. Furthermore, NH4+ could decrease K+ absorption in high‐K+ medium.  相似文献

在纯化棕色棉纤维色素的基础上,测定了天然棕色棉纤维色素的紫外-可见光谱(UV光谱)、红外光谱(IR光谱)以及分析了pH值、浓度、诊断试剂对天然棕色棉纤维色素UV光谱的影响,发现天然棕色棉纤维色素UV光谱随着pH值和浓度不同而变化,诊断试剂对纤维色素UV光谱也有影响,但并不符合黄酮类化合物对诊断试剂的反应规律,红外光谱的主要吸收波数是3554、3477、3414、3289、1638、1618、1386、1073、617、478cm^-1。结合化学性质鉴定,以及和已知的白色棉种皮色素比较。初步推断棕色棉色素的化学结构,认为棕色棉纤维中的色素是由单宁物质氧化形成的醌类化合物。最后解释了棕色棉纤维具有颜色是因为棉纤维发育后期缩合单宁接触空气后氧化的结果。  相似文献

A specific, sensitive and widely applicable reversed-phase high-performance liquid chromatography with fluorescence detection (RP-HPLC-FLD) method was developed for the simultaneous determination of thiamphenicol (TAP), florfenicol (FF) and florfenicol amine (FFA) in eggs. Samples were extracted with ethyl acetate-acetonitrile-ammonium hydroxide (49:49:2, v/v), defatted with hexane, followed by RP-HPLC-FLD determination. Liquid chromatography was performed on a 5 μm LiChrospher C(18) column using a mobile phase composed of acetonitrile (A), 0.01 M sodium dihydrogen phosphate containing 0.005 M sodium dodecyl sulfate and 0.1% triethylamine, adjusted to pH 4.8 by 85% phosphoric acid (B) (A:B, 35:65 v/v), at a flow rate of 1.0 mL/min. The fluorescence detector of HPLC was set at 224 nm for excitation wavelength and 290 nm for emission wavelength. Limits of detection (LODs) were 1.5 μg/kg for TAP and FF, 0.5 μg/kg for FFA in eggs; limits of quantitation (LOQs) were 5 μg/kg for TAP and FF, 2 μg/kg for FFA in eggs. Linear calibration curves were obtained over concentration ranges of 0.025-5.0 μg/mL for TAP with determination coefficients of 0.9997, 0.01-10.0 μg/mL for FF with determination coefficients of 0.9997 and 0.0025-2.50 μg/mL for FFA with determination coefficients of 0.9998, respectively. The recovery values ranged from 86.4% to 93.8% for TAP, 87.4% to 92.3% for FF and from 89.0% to 95.2% for FFA. The corresponding intra-day and inter-day variation (relative standard deviation, R.S.D.) found to be less than 6.7% and 10.8%, respectively.  相似文献

[目的] 三裂叶豚草是中国重点防控的入侵植物,研究其潜在的适生区分布格局变化对预警防控具有重要意义。[方法] 基于MaxEnt优化模型预测当前及未来(RCP2.6、RCP4.5、RCP8.5)气候情景下三裂叶豚草潜在的地理分布格局。[结果] 当调控倍频(RM)=3.5,FC(特征组合)=LQH(线性、二次型、片段化组合)时为最优模型,MaxEnt模型的预测结果较准确,受试者工作特征曲线下的面积(AUC值)达0.9287;综合8种途径评估出人类活动因子(hfp,贡献率69%)、等温性(bio3,贡献率8.9%)、温度季节变动系数(bio4,贡献率2.9%)、年降水量(bio12,贡献率7.3%)和海拔(ELEV,贡献率3.7%)是制约三裂叶豚草潜在地理分布的主要环境因子;在当前气候情景下,三裂叶豚草的高适生区主要集中在东北平原、江淮地区、京津冀、山东半岛等经济发达地区。[结论] 在未来气候变化情景下,三裂叶豚草在我国的潜在适生区范围总体呈现不同程度的缩减,即丧失率大于增加率;三裂叶豚草将有向南迁移的趋势;最冷月最低温度(bio6)是最不相似变量分析中出现频次最高的因子,表明温度因素对三裂叶豚草潜在地理分布变迁起到重要制约作用。本研究结果可为三裂叶豚草的预防和控制提供理论基础。  相似文献

Salintiy and temperature are two important ecological factors which affect the distribution and abundance of Spartina alerniflora Loisel. To find out how cordgrass adapts to the environmental conditions in the introduced range, we studied the dynamics of a series of important physiological components including superoxide dismutase (SOD), guaiacol peroxidase (POD), catalase (CAT), malondialdehyde (MDA), soluble sugar and free proline under different salinity and temperature stresses. The results showed that low NaCl concentration (lower than 100 mmol·L−1) enhanced the growth of S. alterniflora. On the contrary, high NaCl concentration (higher than 100 mmol·L−1) inhibited the growth of S. alterniflora. To a certain extent, S. alterniflora was able to be acclimated to the osmotic pressure created by external solution concentration by adjusting the activities of POD, SOD and CAT, and the contents of free proline and soluble sugar. S. alterniflora varied in its responses to environment in different parts of the plant under 5°C and 38°C temperature stress. Compared with roots, leaves accumulated more soluble sugar, and CAT activities in leaves were higher, whereas SOD and POD activities in leaves were much lower than those in roots.  相似文献

A hairpin RNA-encoding construct targeting gmFAD2-1 was transformed into soybean, and an optimised Agrobacterium-mediated embryonic tip system was employed. A novel intergeneric grafting method using transgenic plantlets as scions was used instead of the conventional rooting method. Compared with traditional acclimatisation, the survival ratio of cleft grafts increased by 70%, and the culture period was shortened by about 40 days. The regeneration frequency of the grafted shoots in this embryonic tip system was approximately 74.6%. Soybean transformants were confirmed by Southern and Northern blot hybridisation analyses. The fatty acid composition of the T1 and T2 seeds from the transformed plants was determined by gas chromatography. The resulting downregulation of the gmFAD2-1 gene substantially increased the level of oleic acid from 16% to 55% as indicated by the oleic desaturation proportion (ODP). The ratio of plants with high ODP, moderate ODP and low ODP was about 1:2:1, which was consistent with a single-gene segregation pattern.  相似文献
