Ge/Si heterostructure with fully strain-relaxed Ge film was grown on a Si (001) substrate by using a two-step process by ultra-high vacuum chemical vapor deposition. The dislocations in the Ge/Si heterostructure were experimentally investigated by high-resolution transmission electron microscopy (HRTEM). The dislocations at the Ge/Si interface were identified to be 90° full-edge dislocations, which are the most efficient way for obtaining a fully relaxed Ge film. The only defect found in the Ge epitaxial film was a 60° dislocation. The nanoscale strain field of the dislocations was mapped by geometric phase analysis technique from the HRTEM image. The strain field around the edge component of the 60° dislocation core was compared with those of the Peierls–Nabarro and Foreman dislocation models. Comparison results show that the Foreman model with a = 1.5 can describe appropriately the strain field around the edge component of a 60° dislocation core in a relaxed Ge film on a Si substrate.  相似文献

A deep understanding of the molecular interactions of carbon nanodots with biomacromolecules is essential for wider applications of carbon nanodots both in vitro and in vivo. Herein, nitrogen and sulfur co‐doped carbon dots (N,S‐CDs) with a quantum yield of 16% were synthesized by a 1‐step hydrothermal method. The N,S‐CDs exhibited a good dispersion, with a graphite‐like structure, along with the fluorescence lifetime of approximately 7.50 ns. Findings showed that the fluorescence of the N,S‐CDs was effectively quenched by bovine hemoglobin as a result of the static fluorescence quenching. The mentioned quenching mechanism was investigated by the Stern‐Volmer equation, temperature‐dependent quenching, and fluorescence lifetime measurements. The binding constants, number of binding sites, and the binding average distance between the energy donor N,S‐CDs and acceptor bovine hemoglobin were calculated as well. These findings will provide for valuable insights on the future bioapplications of N,S‐CDs.  相似文献

The agricultural soils near a copper smelter in southeast China were found to be highly contaminated with Cu, Pb, Zn, and Cd. Metal migration from the soil to groundwater presents an environmental risk that depends on the physicochemical properties of the contaminated soils. Soil solution samples were obtained using lysimeters from a loam soil with multiple metal pollutions over a period of about 1 yr. A field lysimeter study was also conducted to examine the potential use of (S, S')-ethylenediamine-N, N'-disuccinic acid trisodium salt (EDDSNa3) in chelate-enhanced phytoremedation and to evaluate the leaching of heavy metals. The average heavy metal concentrations in the soil solution (without the addition of EDDS) were high (e.g., 0.15 mg Pb L(-1) at a 50-cm depth) compared to the upper limit for protection of groundwater in China, but varied during the sampling period. Cu concentrations were not correlated with pH or dissolved organic carbon (DOC), but Zn and Cd concentrations were related to soil solution pH. EDDS enhanced metal solubility in the soil, but plant metal uptake by Elsholtzia splendens Nakai did not increase accordingly. There may be an increasing risk of groundwater pollution by Cu and the EDDS enhanced phytoremediation technique needs to be carefully applied to minimize this side effect.  相似文献

A flexible asymmetric supercapacitor (ASC) with high energy density is designed and fabricated using flower‐like Bi2O3 and MnO2 grown on carbon nanofiber (CNF) paper as the negative and positive electrodes, respectively. The lightweight (1.6 mg cm?2), porous, conductive, and flexible features make the CNF paper an ideal support for guest active materials, which permit a large areal mass of 9 mg cm?2 for Bi2O3 (≈85 wt% of the entire electrode). Thus, the optimal device with an operation voltage of 1.8 V can deliver a high energy density of 43.4 μWh cm?2 (11.3 W h kg?1, based on the total electrodes) and a maximum power density of 12.9 mW cm?2 (3370 W kg?1). This work provides an example of large areal mass and flexible electrode for ASCs with high areal capacitance and high energy density, holding great promise for future flexible electronic devices.  相似文献

DNA replication in all eukaryotes starts with the process of loading the replicative helicase MCM2–7 onto chromatin during late mitosis of the cell cycle. MCM2–7 is a key component of the prereplicative complex (pre-RC), which is loaded onto chromatin by the concerted action of origin recognition complex, Cdc6, and Cdt1. Here, we demonstrate that And-1 is assembled onto chromatin in late mitosis and early G1 phase before the assembly of pre-RC in human cells. And-1 forms complexes with MCM2–7 to facilitate the assembly of MCM2–7 onto chromatin at replication origins in late mitosis and G1 phase. We also present data to show that depletion of And-1 significantly reduces the interaction between Cdt1 and MCM7 in G1 phase cells. Thus, human And-1 facilitates loading of the MCM2–7 helicase onto chromatin during the assembly of pre-RC.  相似文献

IFN-β is induced via a c-fos dependent mechanism that is present downstream of the receptor activator of NF-κB ligand (RANKL)-RANK signal transduction cascade during osteoclast differentiation. Increased production of IFN-β in turn inhibits osteoclastogenesis. However, the mechanism by which IFN-β exerts its suppressive function remains unclear. In the present study, we found that miR-155, an IFN-β-induced miRNA, mediated the suppressive effect of IFN-β on osteoclast differentiation by targeting SOCS1 and MITF, two essential regulators of osteoclastogenesis. These findings have not only demonstrated that miR-155 inhibits osteoclast differentiation, but also provided a new therapeutic target for treatment of osteoclast-mediated diseases.  相似文献

The study aims to characterize the pharmacokinetic, tissue distribution, excretion, and antiviral activity properties of a novel pegylated recombinant human consensus interferon-α variant (PEG-IFN-SA) following a single subcutaneous administration to monkeys, rats and guinea pigs. Studies included: (1) pharmacokinetic properties of PEG-IFN-SA and comparison with those of non-pegylated IFN-SA in rhesus monkeys and rats; (2) tissue distribution and urinary, fecal, and biliary excretion patterns of (125)I-PEG-IFN-SA in guinea pigs; and (3) antiviral activity assessment of PEG-IFN-SA in cynomolgus monkeys. The pegylated protein exhibited improved pharmacokinetic properties compared to IFN-SA in both monkeys and rats, with a 12-fold and 15-fold increase in elimination half-life, and a 100-fold and 10-fold decrease in serum clearance, as well as a 2.5-fold and 10-fold increase in the time to reach peak serum concentration, respectively. (125)I-PEG-IFN-SA was found to be distributed to most of the tissues examined and has character of targeting special distribution, and urinary appeared to be a major route for the excretion of PEG-IFN-SA in guinea pigs. Serum sample analysis from PEG-IFN-SA-treated monkeys showed dose-dependent antiviral activity for one week. These findings demonstrate that pegylation of IFN-SA results in more desirable pharmacokinetic properties, enhanced drug exposure and sustained-efficacy of in vivo antiviral action.  相似文献

利用DNAStar分析了GenBank中所有8株羊痘病毒全基因序列,选取位于山羊痘病毒(AY077835)基因组gp064区域约64bp的基因片段,设计并合成了一对PCR引物和一条TaqMan-MGB探针,建立了FQ-PCR和标准曲线,并利用该方法对山羊痘临床皮肤痘疹材料和人工感染动物材料进行GPV核酸检测。结果表明,构建的FQ-PCR具有良好的敏感性、特异性、稳定性和临床应用性。该方法的建立为山羊痘临床快速高效地诊断和山羊痘病毒感染羊只的发生、发展及转归研究提供了一种有效的手段。  相似文献

SUMO is a protein modifier that is vital for multicellular development. Here we present the first system-wide analysis, combining multiple approaches, to correlate the sumoylated proteome (SUMO-ome) in a multicellular organism with the developmental roles of SUMO. Using mass-spectrometry-based protein identification, we found over 140 largely novel SUMO conjugates in the early Drosophila embryo. Enriched functional groups include proteins involved in Ras signaling, cell cycle, and pattern formation. In support of the functional significance of these findings, sumo germline clone embryos exhibited phenotypes indicative of defects in these same three processes. Our cell culture and immunolocalization studies further substantiate roles for SUMO in Ras signaling and cell cycle regulation. For example, we found that SUMO is required for efficient Ras-mediated MAP kinase activation upstream or at the level of Ras activation. We further found that SUMO is dynamically localized during mitosis to the condensed chromosomes, and later also to the midbody. Polo kinase, a SUMO substrate found in our screen, partially colocalizes with SUMO at both sites. These studies show that SUMO coordinates multiple regulatory processes during oogenesis and early embryogenesis. In addition, our database of sumoylated proteins provides a valuable resource for those studying the roles of SUMO in development.  相似文献

Carbon supplementation, soil moisture and soil aeration are believed to enhance in situ bioremediation of PAH-contaminated soils by stimulating the growth of indigenous microorganisms. However, the effects of added carbon and nitrogen together with soil moisture and soil aeration on the dissipation of PAHs and on associated microbial counts have yet to be fully assessed. In this study the effects on bioremediation of carbon source, carbon-to-nitrogen ratio, soil moisture and aeration on an aged PAH-contaminated agricultural soil were studied in microcosms over a 90-day period. Additions of starch, glucose and sodium succinate increased soil bacterial and fungal counts and accelerated the dissipation of phenanthrene and benzo(a)pyrene in soil. Decreases in phenanthrene and benzo(a)pyrene concentrations were effective in soil supplemented with glucose and sodium succinate (both 0.2 g C kg−1 dry soil) and starch (1.0 g C kg−1 dry soil). The bioremediation effect at a C/N ratio of 10:1 was significantly higher (P < 0.05) than at a C/N of either 25:1 or 40:1. Soil microbial counts and PAH dissipation were lower in the submerged soil but soil aeration increased bacterial and fungal counts, enhanced indigenous microbial metabolic activities, and accelerated the natural degradation of phenanthrene and benzo(a)pyrene. The results suggest that optimizing carbon source, C/N ratio, soil moisture and aeration conditions may be a feasible remediation strategy in certain PAH contaminated soils with large active microbial populations.  相似文献
