Furin is a member of the proprotein convertase family, which is capable of cleaving the precursors of a wide variety of substrates including membrane-type 1 matrix metalloproteinase (MT1-MMP) proenzyme. c-Src is activated by growth factors, and has been linked with a poor prognosis in pancreatic cancer (PCa). Both c-Src and Furin play crucial roles in tumorigenesis, and the mechanism controlling their association is not understood. Modulation of the association between Furin and pro-MT1-MMP by c-Src inhibitor PP2 was evaluated by western blotting, assay of in vitro enzyme, co-immunoprecipitation (co-IP), and confocal immunofluorescence microscopy. Human platelet-derived growth factor BB (PDGF-BB) activated c-Src and induced c-Src-dependent association of Furin with pro-MT1-MMP in HPAC pancreatic cancer cells. Co-IP and confocal immunofluorescence assays revealed that c-Src interacts with Furin in vivo. The SH2 domain appeared to be important for c-Src interaction with Furin. In addition, we showed that Furin protein is tyrosine phosphorylated. Association between Furin and MT1-MMP is regulated by the tyrosine kinase c-Src.  相似文献

本研究根据其它植物Actin基因的保守序列设计一对简并性引物,以拒盐型盐生植物小花碱茅根部总RNA为模板,采用RT-PCR的方法扩增出Actin基因片段并克隆到PUCm-T载体,阳性克隆经PCR检测后进行测序,在GenBank中注册;序列分析结果表明:该片段长约600 bp,编码198个氨基酸;所得序列与GenBank中注册的其它植物Actin基因序列同源性均在84%以上,与其它肌动蛋白的氨基酸序列同源性达94%以上.  相似文献

遥感的空间性、实时性与作物生长模型的过程性、机理性优势互补,将两者有效耦合已成为提高作物生长监测预测能力的重要手段之一。提出了一种基于地空遥感信息与生长模型耦合的冬小麦预测方法,该方法基于初始化/参数化策略,以不同生育时期的小麦叶面积指数(LAI)和叶片氮积累量(LNA)为信息融合点将地面光谱数据(ASD)及HJ-1 A/B CCD、Landsat-5 TM数据与冬小麦生长模型(WheatGrow)耦合,反演得到区域尺度生长模型运行时难以准确获取的部分管理措施参数(播种期、播种量和施氮量),在此基础上实现了对冬小麦生长的有效预测。实例分析结果表明,LNA较LAI对模型更敏感,以之作为耦合点的反演效果较好。另外,抽穗期是遥感信息与生长模型耦合的最佳时机,对播种期、播种量和施氮量反演的RMSE值分别达到5.32 d、14.81 kg/hm2、14.11 kg/hm2。生长模型与遥感耦合后的模拟结果很好地描述了冬小麦长势和生产力指标的时空分布状况,长势指标的模拟相对误差小于0.25,籽粒产量模拟的相对误差小于0.1。因此研究结果可为区域尺度冬小麦生长的监测预测提供重要理论依据。  相似文献

小麦冠层反射光谱与植株水分状况的关系   总被引:21,自引:3,他引:21  
研究了不同土壤水、氮条件下小麦冠层光谱反射特征与叶片和植株水分状况的相关性.结果表明,在小麦主要生育期,冠层叶片含水率与460~510、610~680和1480~1500nm波段范围内的光谱反射率有较高的相关性,植株含水率与810~870nm波段范围内的光谱反射率密切相关.在整个生长期内,小麦冠层叶片含水率与460~1500nm波段范围内的光谱反射率均有良好相关性,植株含水率与560~1480nm波段范围内光谱反射率的相关性均达到极显著水平.冠层叶片(CL)、上层叶(UL)和下层叶片(LL)含水率与光谱指数的相关程度为CL>LL>UL.冠层叶片和植株含水率与比值指(R(610,560))和光谱指数(R(610,560)/ND(810,610))呈极显著线性负相关,与归一化指数((R810-R610)/(R810+R610))呈极显著线性正相关.其中,用光谱指数(R(610,560)/ND(810,610))监测不同生育期小麦冠层叶片和植株含水率的效果最好。  相似文献

The unit firing activities of neurons in the preoptic area (POA) of ground squirrel hypothalamic tissue slices were recorded and the metabolism of NA in hypothalamus was measured with high performance liquid chromatography (HPLC). Thermosensitivity, proportions, the critical temperature (Tc) and the lowest temperature (TL) of firing activity of the above-mentioned neurons, and NA metabolism in hypothalamus were compared in different seasons and hibernating phases. In comparison with that in summer euthermar, it was shown that (i) the percentage and thermosensitivity of the POA neurons varied respectively in the hibernating phases; (ii) TL and Tc of the POA neurons in winter, both euthermar and hibernation, were markedly decreased; (iii) the POA neurons in hibernation became much more sensitive to NA, and the response of cold-sensitive neurons to NA changed from inhibiting pattern in summer to exciting one in hibernation; (iv) the contents and metabolism of NA in hypothalamus decreased significantly in the entering phase and deep hibernation phase, while the metabolism of NA increased remarkably in the arousal phase. These changes might explain the regulatory mechanism how ground squirrel actively decreases body temperature (Tb) in entering into hibernation and quickly recovers body temperature in arousal phase. In this paper, we investigate a simple protein sequence conservation measure which takes amino acid similarity into account. Instead of grouping 20 amino acids into disjoint sets in previous methods, we consider ten overlapping classes. The method is based on the assumption that a column in a multiple sequence alignment is evolved from an identical column in the evolutionary history. Two ten-dimensional vectors are constructed for each position to denote frequencies of ten classes in a column and the corresponding hypothetical identical column. Then the cosine function of the angle between these two vectors is considered as a measure of divergence of stereochemical properties at this position. This divergence, combining with other conservation scores, is used as conservation measure of the column. Finally, we evaluate our methods by identifying catalytic sites, using rank analysis criterion and receiver operator characteristic analysis criterion.  相似文献
