A method was developed to analyze biogenic amines in extracts of the central nervous system of the tobacco hornworm, Manduca sexta by high-performance liquid chromatography with 16-sensor electrochemical detection (n-EC-HPLC). The amines, precursors, and metabolites were separated in two dimensions. The first dimension involves separation based upon retention time by reversed-phase HPLC, while the second dimension involves separation based upon the characteristic oxidation potentials achieved by n-EC. Biogenic amine identification was based upon maximum oxidation potential and peak height ratios in addition to retention time. The improved resolving power of this method allows for a simplified sample preparation procedure and simultaneous determination of a wide range of compounds, including phenylethylamine, catecholamines, indoleamines, and some of their precursors and metabolites.  相似文献

Currently, knowledge is limited concerning the impact of a Lactobacillus plantarum JL01 diet for weaned piglets on caecal bacteria and metabolite profiles. In our experiments, 24 weaned piglets were randomly divided into two groups; each piglet in the treatment groups (Cec-Lac) was fed a basic diet and administered 10 ml of L. plantarum JL01 (1·0 × 109 CFU per ml) every day. The control group (Cec-Con) was fed a basic diet. After feeding for 28 days, we analysed the parameters of the caecal digesta of weaned piglets. We used 16S rDNA gene sequencing and mass spectrometry (MS)-based metabolomics techniques to investigate the effect of a L. plantarum JL01 diet on intestinal microbial composition and its metabolite profiles in the caecum contents of weaned piglets. The results showed that the richness estimators (ACE and Chao indices) in the caecal bacteria increased in the Cec-Lac group. Prevotella_2 and Desulfovibrio decreased significantly, while Pantoea and Rectale_group increased in the caecum of weaned piglets in the Cec-Lac group. Furthermore, Pearson's correlation analysis revealed that the genus Rectale_group was positively correlated with indole-3-acetic acid (P < 0·05), and the genus Pantoea had the same correlation with 1-palmitoyl lysophosphatidic acid. The metabolomics analysis revealed that the L. plantarum JL01 diet supplementation had significant effects on tryptophan metabolism and fat digestion and absorption. The results indicated that the L. plantarum JL01 dietary supplementation not only altered the microbial composition but also mediated tryptophan metabolism and fat digestion and absorption in the caecum, factors that may further affect the health of the host.  相似文献

Our planet is facing a variety of serious threats from climate change that are unfolding unevenly across the globe. Uncovering the spatial patterns of ecosystem stability is important for predicting the responses of ecological processes and biodiversity patterns to climate change. However, the understanding of the latitudinal pattern of ecosystem stability across scales and of the underlying ecological drivers is still very limited. Accordingly, this study examines the latitudinal patterns of ecosystem stability at the local and regional spatial scale using a natural assembly of forest metacommunities that are distributed over a large temperate forest region, considering a range of potential environmental drivers. We found that the stability of regional communities (regional stability) and asynchronous dynamics among local communities (spatial asynchrony) both decreased with increasing latitude, whereas the stability of local communities (local stability) did not. We tested a series of hypotheses that potentially drive the spatial patterns of ecosystem stability, and found that although the ecological drivers of biodiversity, climatic history, resource conditions, climatic stability, and environmental heterogeneity varied with latitude, latitudinal patterns of ecosystem stability at multiple scales were affected by biodiversity and environmental heterogeneity. In particular, α diversity is positively associated with local stability, while β diversity is positively associated with spatial asynchrony, although both relationships are weak. Our study provides the first evidence that latitudinal patterns of the temporal stability of naturally assembled forest metacommunities across scales are driven by biodiversity and environmental heterogeneity. Our findings suggest that the preservation of plant biodiversity within and between forest communities and the maintenance of heterogeneous landscapes can be crucial to buffer forest ecosystems at higher latitudes from the faster and more intense negative impacts of climate change in the future.  相似文献

本文是1991年11月6—13日横滨第11届国际组织相容性会议(IHWC)协作科研中我国主要少数民族(苗族、布依族、蒙古族、满族、回族、藏族、维吾尔族)和南北汉族样本HLAⅠ、Ⅱ、Ⅲ类抗原多态性分析的联合报告。聚类分析表明,苗、布依与南方汉族聚类,蒙、满、回、藏与北方汉族集群,提示中华民族包含南北两大群体。维吾尔族非常疏远地共聚于高加索人种。高加索人种起源的HLA抗原基因频率(A3、B8等)由西向东又由北向南递减。东南亚蒙古人种起源的HLA抗原基因频率(B46等)由南向北递减。HLA抗原基因频率的这种梯度分布反映我们祖先自远古史前时期以来的不断迁移和相互融合的过程。本文讨论了中华民族包含南北两大群体这一事实对于中华民族起源的启示。  相似文献

Personalized intervention strategies, in particular those that modify treatment based on a participant's own response, are a core component of precision medicine approaches. Sequential multiple assignment randomized trials (SMARTs) are growing in popularity and are specifically designed to facilitate the evaluation of sequential adaptive strategies, in particular those embedded within the SMART. Advances in efficient estimation approaches that are able to incorporate machine learning while retaining valid inference can allow for more precise estimates of the effectiveness of these embedded regimes. However, to the best of our knowledge, such approaches have not yet been applied as the primary analysis in SMART trials. In this paper, we present a robust and efficient approach using targeted maximum likelihood estimation (TMLE) for estimating and contrasting expected outcomes under the dynamic regimes embedded in a SMART, together with generating simultaneous confidence intervals for the resulting estimates. We contrast this method with two alternatives (G-computation and inverse probability weighting estimators). The precision gains and robust inference achievable through the use of TMLE to evaluate the effects of embedded regimes are illustrated using both outcome-blind simulations and a real-data analysis from the Adaptive Strategies for Preventing and Treating Lapses of Retention in Human Immunodeficiency Virus (HIV) Care (ADAPT-R) trial (NCT02338739), a SMART with a primary aim of identifying strategies to improve retention in HIV care among people living with HIV in sub-Saharan Africa.  相似文献

The increasing world population has forced excessive chemical fertilizer and irrigation to complete the global food demand, deteriorating the water quality and nutrient losses. Short-term studies do not compile the evidences; therefore, the study aimed to identify the effectiveness of reduced doses of inorganic fertilizer and water-saving practices, hence, a six-year experiment (2015–2020) was conducted in China to address the knowledge gap. The experimental treatments were: farmer accustomed fertilization used as control (525:180:30 kg NPK ha−1), fertilizer decrement (450:150:15 kg NPK ha−1), fertilizer decrement + water-saving irrigation (450:150:15 kg NPK ha−1), application of organic and inorganic fertilizer + water-saving irrigation (375:120:0 kg NPK ha−1 + 4.5 tones organic fertilizer ha−1), and application of controlled-release fertilizer (80:120:15 kg NPK ha−1). Each treatment was replicated thrice following a randomized complete block design. The results achieved herein showed that control has the highest losses in the six-year study for total nitrogen (225.97 mg L−1), total soluble nitrogen (121.58 mg L−1), nitrate nitrogen (0.93 mg L−1), total phosphorus (0.57 mg L−1), and total soluble phosphorus (0.57 mg L−1) respectively. Reduced fertilizer and water application improved crop nutrient uptake, nitrogen concentration was significantly enhanced with organic and inorganic fertilizer + water-saving irrigation, P concentration was increased with fertilizer decrement + water-saving irrigation, and K concentration was improved with fertilizer decrement + water-saving irrigation. Hence, this study concludes that reduced inorganic fertilizer dose combined with water-saving practices is significantly helpful in reducing nutrient leaching losses and improving nutrient uptake and water pollution. Further studies are needed to explore the impacts of reduced fertilization and water-saving irrigation on leaching losses. The benefits at different climatic conditions, soil types, and fertilizer types with application methods are also a research gap.
