High-mannose type N-linked glycan with 6 mannosyl residues, termed "M6Gn2", displayed clear binding to the same M6Gn2, conjugated with ceramide mimetic (cer-m) and incorporated in liposome, or coated on polystyrene plates. However, the conjugate of M6Gn2-cer-m did not interact with complex-type N-linked glycan with various structures having multiple GlcNAc termini, conjugated with cer-m. The following observations indicate that hamster embryonic fibroblast NIL-2 K cells display homotypic autoadhesion, mediated through the self-recognition capability of high-mannose type glycans expressed on these cells: (i) NIL-2 K cells display clear binding to lectins capable of binding to high-mannose type glycans (e.g., ConA), but not to other lectins capable of binding to other carbohydrates (e.g. GS-II). (ii) NIL-2 K cells adhere strongly to plates coated with M6Gn2-cer-m, but not to plates coated with complex-type N-linked glycans having multiple GlcNAc termini, conjugated with cer-m; (iii) degree of NIL-2 K cell adhesion to plates coated with M6Gn2-cer-m showed a clear dose-dependence on the amount of M6Gn2-cer-m; and (iv) the degree of NIL-2 K adhesion to plates coated with M6Gn2-cer-m was inhibited in a dose-dependent manner by α1,4-L-mannonolactone, the specific inhibitor in high-mannose type glycans addition. These data indicate that adhesion of NIL-2 K is mediated by self-aggregation of high mannose type glycan. Further studies are to be addressed on auto-adhesion of other types of cells based on self interaction of high mannose type glycans.  相似文献

The goal of this study is to quantify and determine the way in which the emotional response to music is reflected in the electrical activities of the brain. When the power spectrum of sequences of musical notes is inversely proportional to the frequency on a log-log plot, we call it 1/f music. According to previous research, most listeners agree that 1/f music is much more pleasing than white (1/f 0) or brown (1/f 2) music. Based on these studies, we used nonlinear methods to investigate the chaotic dynamics of electroencephalograms (EEGs) elicited by computer-generated 1/f music, white music, and brown music. In this analysis, we used the correlation dimension and the largest Lyapunov exponent as measures of complexity and chaos. We developed a new method that is strikingly faster and more accurate than other algorithms for calculating the nonlinear invariant measures from limited noisy data. At the right temporal lobe, 1/f music elicited lower values of both the correlation dimension and the largest Lyapunov exponent than white or brown music. We observed that brains which feel more pleased show decreased chaotic electrophysiological behavior. By observing that the nonlinear invariant measures for the 1/f distribution of the rhythm with the melody kept constant are lower than those for the 1/f distribution of melody with the rhythm kept constant, we could conclude that the rhythm variations contribute much more to a pleasing response to music than the melody variations do. 目的 海南岛地处热带,具有丰富的龟鳖物种多样性.为了有效保护这一珍贵资源,对该地区龟鳖的多样性、养殖现状及保护管理需求进行总结和讨论.方法 根据有关文献和我们的调查结果,从龟鳖多样性、养殖现状、保护和可持续利用的管理等方面进行分析.结果 海南岛现有龟鳖6科14属18种,占中国龟鳖的50%,其中有1种野外灭绝,有5种处于极度濒危状态,有11种处于濒危状态;国家一级重点保护的有1种,二级保护的有9种.海南岛是我国龟鳖养殖种类最多、规模最大的地区,有龟鳖养殖户330多户,养殖龟鳖120余种.结论 海南岛龟鳖资源丰富,养殖规模巨大.应进一步加强野生龟鳖资源的保护和管理,将龟鳖纳入国家重点保护的野生动物范围,继续开展保护繁育放归活动.同时,从多方面加强龟鳖养殖的产业化发展,促进龟鳖资源的可持续利用.  相似文献

Background and Aims

Great attention has been paid to N2O emissions from paddy soils under summer rice-winter wheat double-crop rotation, while less focus was given to the NO emissions. Besides, neither mechanism is completely understood. Therefore, this study aimed at evaluating the relative importance of nitrification and denitrification to N2O and NO emissions from the two soils at different soil moisture contents

Methods

N2O and NO emissions during one winter wheat season were simultaneously measured in situ in two rice-wheat based field plots at two different locations in Jiangsu Province, China. One soil was neutral in pH with silt loam texture (NSL), the other soil alkaline in pH with a clay texture (AC). A 15?N tracer incubation experiment was conducted in the laboratory to evaluate the relative importance of nitrification and denitrification for N2O and NO emissions at soil moisture contents of 40 % water holding capacity (WHC), 65 % WHC and 90 % WHC.

Results

Higher N2O emission rates in the AC soil than in the NSL soil were found both in the field and in the laboratory experiments; however, the differences in N2O emissions between AC soil and NSL soil were smaller in the field than in the laboratory. In the latter experiment, nitrification was observed to be the more important source of N2O emissions (>70 %) than denitrification, regardless of the soils and moisture treatments, with the only exception of the AC soil at 90 % WHC, at which the contributions of nitrification and denitrification to N2O emissions were comparable. The ratios of NO/N2O also supported the evidence that the nitrification process was the dominant source of N2O and NO both in situ and in the laboratory. The proportion of nitrified N emitted as N2O (P N2O ) in NSL soil were around 0.02 % in all three moisture treatments, however, P N2O in the AC soil (0.04 % to 0.10 %) tended to decrease with increasing soil moisture content.

Conclusions

Our results suggest that N2O emission rates obtained from laboratory incubation experiments are not suitable for the estimation of the true amount of N2O fluxes on a field scale. Besides, the variations of P N2O with soil property and soil moisture content should be taken into account in model simulations of N2O emission from soils.  相似文献

重组腺相关病毒(Recombinant adenovirus-associated virus,rAAV)载体源于腺相关病毒(Adeno-associated virus,AAV),以其无致病性、低免疫原性、可定点整合及在宿主细胞内长期表达等优势,广泛应用于肿瘤和遗传疾病等基因治疗研究,被视为最有前途的基因治疗载体之一。但是人体内广泛存在AAV中和抗体,以及rAAV对体内特定组织或细胞的靶向性欠理想,限制了其在临床上的应用。经化学修饰的rAAV可以抵御血清中和抗体,提高转导靶向性,还可实现rAAV体内动态监测,这为解决当前rAAV载体的问题提供了新的思路和方法。本文对化学修饰的rAAV展开系统阐述,并对其未来发展趋势作出展望。  相似文献

目的:通过克隆LC3.I基因,体外原核表达LC3-I蛋白后制备抗LC3单克隆抗体,作为自噬研究中的标记分子检测自噬的发生和发展过程。方法:RT.PCR方法从RAW264.7细胞基因组中克隆LC3基因,亚克隆至pQE80L原核表达载体后转化E.cobDH5a进行诱导表达,SDS—PAGE电泳及Westemblot鉴定表达蛋白。蛋白纯化后免疫BALB/c小鼠。采用淋巴细胞杂交瘤技术,制备分泌抗LC3.I杂交瘤细胞株,体内诱生腹水制备mAb,间接ELISA法测定其效价,辛酸一硫酸铵沉淀法及亲和层析法纯化mAb。结果:成功克隆了LC3一I基因,并对其在E.coilDH5a进行诱导表达,SDS-PAGE分析表明在相对分子量Mr为20×10^3有特异条带,Westernblot验证表达产物具有一定的生物学活性。建立了3株稳定分泌特异性抗LC3-ImAb的杂交瘤细胞株,诱导产生的腹水获得的抗体效价在10^5-10^7之间,结论:在E.coli中对LC3-I进行表达,并制备特异性较强抗LC3-I蛋白的单克隆抗体。为自噬研究提供了良好的标记分子,可对自噬形成和发展进行有效的检测。  相似文献

Baculovirus chitinase gene (chiA) is a late gene essential for liquefying the host insect at a late stage of infection for its hydrolyzing chitin function. In a previous report, baculovirus ChiA has been shown to offer many interesting new opportunities for pest control. Recently, a putative chiA gene was identified in the Korean isolate of the Spodoptera litura nucleopolyhedorvirus (SpliMNPV‐K1) genome. The open reading frame (ORF) contains 1692 nucelotides and encodes a protein of 563 amino acids with a predicted molecular weight of about 62.6 kDa. To study the insecticidal activity of ChiA from SpliMNPV‐K1, we constructed a recombinant AcMNPV, Ap‐SlChiA, which is designed to express the ChiA under the control of a polyhedrin promoter. Western blot analysis indicated that ChiA was successfully expressed by this recombinant virus. Chitinase assay revealed that the chitobiosidase and endochitinase activity of the recombinant virus was 2.5‐ and 3.9‐flods higher than those of wild‐type AcMNPV, respectively. In addition, the recombinant virus showed higher evident insecticidal activity against 3rd instar larvae of Spodotera exigua than that of the AcMNPV. These results suggest that the chiA gene from SpliMNPV‐K1 could be successfully applied to improve pathogenicity of baculoviruses.  相似文献
