For a proper development of the placenta, maternal NK cells should not attack the fetal extravillous cytotrophoblast cells. This inhibition of maternal NK cells is partially mediated via the nonclassical MHC class I molecule HLA-G. Recently, we demonstrated that HLA-G forms disulfide-linked high molecular complexes on the surface of transfected cells. In the present study, we demonstrate that HLA-G must associate with beta(2)m for its interaction with CD85J/leukocyte Ig-like receptor-1 (LIR-1). Although HLA-G free H chain complexes are expressed on the surface, they are not recognized and possibly interfere with CD85J/LIR-1 and HLA-G interaction. The formation of these complexes on the cell surface might represent a novel mechanism developed specifically by the HLA-G protein aimed to control the efficiency of the CD85J/LIR-1-mediated inhibition. We also show that endogenous HLA-G complexes are expressed on the cell surface. These findings provide novel insights into the delicate interaction between extravillous cytotrophoblast cells and NK cells in the decidua. 相似文献

Positive response of plant species to plant growth-promoting rhizobacteria have led to an increased interest in their use as bacterial inoculants. However, the introduction of exogenous bacteria into natural ecosystems may perturb bacterial populations within the microbial community and lead to the disruption of indigenous populations performing key functional roles. In this study the effect of Azospirillum brasilense inoculation on maize (Zea mays) rhizosphere Actinobacteria, Bacteroidetes, alpha-Proteobacteria, Pseudomonas and Bdellovibrio spp. was assessed using a polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis (DGGE) approach in conjunction with group-specific primers. The DGGE fingerprints analysis revealed that the introduction of A. brasilense did not alter or disrupt the microbial system at the group-specific level. However, some communities such as the alpha-Proteobacteria and Bdellovibrio were influenced by plant age while the other bacterial groups remained unaffected. Based on these as well as previous data, it can be inferred that inoculation with A. brasilense does not perturb the natural bacterial populations investigated. 相似文献

The crystal structure of the light-harvesting phycobiliprotein, c-phycocyanin from the thermophilic cyanobacterium Synechococcus vulcanus has been refined to 1.6 A resolution based on the previously determined lower resolution structure (PDB entry 1I7Y). The improved data was collected using synchrotron radiation at 100 K. The significantly improved crystallographic data has lead to improved calculated electron density maps, allowing the unambiguous positioning of all protein and co-factor atoms and the positioning of 377 solvent molecules. The positions of solvent molecules at specific sites important for stabilization of different levels of self-assembly of the phycobilisome structure were identified and the bonding network is described. The presence of solvent molecules in the vicinity of the co-factors and in intermolecular spaces is identified and their possible roles are suggested. All three of the phycocyanobilin co-factors bind water molecules at specific sites between the propionic acid side chains. Molecular dynamic (MD) simulations support that these special waters have a role in stabilization of this conformation. On the basis of the crystal packing reported here and in comparison to other phycobiliprotein crystal forms, we have analyzed the roles of specific sites on the formation of the phycobilisome complex. 相似文献

Early survival and growth of some plants in arid environments depends on facilitation by a nurse plant. Amelioration of soil
temperature extremes through shading and accumulation of mineral nutrients near nurse-plants are mechanisms of facilitation.
We investigated the effects of shading (soil temperature) and soil type on survival and growth of the giant columnar cactus,
cardon (Pachycereus pringlei). Cardon was grown either in a sandy clay-loam soil obtained from resource islands formed under mature mesquite (Prosopis articulata) or in the loamy-sand soil from plant-free bare areas that surround the islands. Seedlings were potted in these soils and
the pots were buried to ground level in the open. We also determined plant responses to fertilization with N, P, K or NPK
in the bare-area soils. Enhancement of survival and growth in the resource-island soils compared to that in the bare-area
soils was highly significant. Plants survived and grew better in resource-island soils than in bare-area soil, an effect that
was enhanced by shading (one-half of full sun). Greater root/shoot ratios of plants grown in bare-area soil indicated increased
resource allocation to roots under limiting conditions. Significant interactions (analysis of variance) indicated that the
soil and sun factors of the experiment were not independent of one another. Plant growth in bare-area soil improved considerably
(>200%) in response to N fertilization (screenhouse conditions), and approximated that of plants in resource-island soil without
N amendment. The growth response to P was small (<50%), while K did not affect growth significantly. Responses to NPK were
similar to those to N alone. The results suggested that shading and nutritional effects interact in determining early survival
and growth of cardon in different soils.

Complex formation among transforming growth factor-β (TGF-β) receptors and its modulation by coreceptors represent an important level of regulation for TGF-β signaling. Oligomerization of ALK5 and the type II TGF-β receptor (TβRII) has been thoroughly investigated, both in vitro and in intact cells. However, such studies, especially in live cells, are missing for the endothelial cell coreceptor endoglin and for the ALK1 type I receptor, which enables endothelial cells to respond to TGF-β by activation of both Smad2/3 and Smad1/5/8. Here we combined immunoglobulin G–mediated immobilization of one cell-surface receptor with lateral mobility studies of a coexpressed receptor by fluorescence recovery after photobleaching (FRAP) to demonstrate that endoglin forms stable homodimers that function as a scaffold for binding TβRII, ALK5, and ALK1. ALK1 and ALK5 bind to endoglin with differential dependence on TβRII, which plays a major role in recruiting ALK5 to the complex. Signaling data indicate a role for the quaternary receptor complex in regulating the balance between TGF-β signaling to Smad1/5/8 and to Smad2/3. 相似文献

The effects of NaCl on the kinetic properties of desalted phosphoenolpyruvate carboxylase (PEP carboxylase, EC 4.1.1.31) from two halophytes, Suaeda monoica Forssk. ex. J.F. Gmel and Chloris gayana Kunth. were investigated. The tolerance of PEP carboxylase to NaCl in the reaction medium depends on the enzyme pre-conditioning as well as on the concentration of its substrate PEP in the assay medium. Addition of PEP to the extraction and the storage medium, stabilizes the enzyme. Such a pre-treated enzyme is inhibited by NaCl in the presence of low concentrations of PEP in the assay medium but is activated by NaCl in the presence of PEP at concentrations above 1.0 m M . NaCl modifies the nH value, K' and Vmax , and seems to act as an allosteric effector. 相似文献

Students of Israeli politics have stressed cultural factors in explaining the success of right‐wing parties among Oriental Jewish voters. My argument is that closer attention must be paid to labour market relations in trying to explain both the anti‐Arab sentiments of Oriental Jews and their proclivity for right‐wing politics. Oriental Jews compete with both citizen and non‐citizen Palestinians for jobs at the lowest end of the occupational ladder. This competition, I argue, can explain a great deal of their political attitudes. The data to support the argument are derived from an attitude survey conducted in 1988 in eight ‘development towns’ ‐ small working‐class communities populated mainly with Orientals and characterized by high unemployment rates and pervasive social and economic ills. These towns, and the sociologically similar slum neighbourhoods of major cities, provided Rabbi Meir Kahane, Israel's most vociferously racist politician, with the bulk of his electoral support when he was elected to the Knesset in 1984. In 1988 I found support for Kahane in development towns was almost three times as high as it was in 1984. (Kahane was barred from running for the Knesset in 1988 for being a racist.) Analysis of the data showed that this support was disproportionately concentrated among respondents who suffered most from the effects of labour market friction with Palestinian workers. 相似文献

Defects in ribosome biogenesis and function are present in a growing list of human syndromes associated with cancer susceptibility. One example is X‐linked dyskeratosis congenita (X‐DC) in which the DKC1 gene, encoding for an enzyme that modifies ribosomal RNA, is found to be mutated. How ribosome dysfunction leads to cancer remains poorly understood. A critical cellular response that counteracts cellular transformation is oncogene‐induced senescence (OIS). Here, we show that during OIS, a switch between cap‐ and internal ribosome entry site (IRES)‐dependent translation occurs. During this switch, an IRES element positioned in the 5′untranslated region of p53 is engaged and facilitates p53 translation. We further show that in DKC1m cells, p53 IRES‐dependent translation is impaired during OIS ex vivo and on DNA damage in vivo. This defect in p53 translation perturbs the cellular response that counteracts oncogenic insult. We extend these findings to X‐DC human patient cells in which similar impairments in p53 IRES‐dependent translation are observed. Importantly, re‐introduction of wild‐type DKC1 restores p53 expression in these cells. These results provide insight into the basis for cancer susceptibility in human syndromes associated with ribosome dysfunction. 相似文献
