Earlier studies have reported a role for lipooligosaccharides (LOSs) in sliding motility, biofilm formation, and infection of host macrophages in Mycobacterium marinum. Although a LOS biosynthetic gene cluster has recently been identified in this species, many structural features of the different LOSs (LOS-I–IV) are still unknown. This clearly hampers assessing the contribution of each LOS in mycobacterial virulence as well as structure-function-based studies of these important cell wall-associated glycolipids. In this study, we have identified an M. marinum isolate, M. marinum 7 (Mma7), which failed to produce LOS-IV but instead accumulated large amounts of LOS-III. Local genomic comparison of the LOS biosynthetic cluster established the presence of a highly disorganized region in Mma7 compared with the standard M strain, characterized by multiple genetic lesions that are likely to be responsible for the defect in LOS-IV production in Mma7. Our results indicate that the glycosyltransferase LosA alone is not sufficient to ensure LOS-IV biosynthesis. The availability of different M. marinum strains allowed us to determine the precise structure of individual LOSs through the combination of mass spectrometric and NMR techniques. In particular, we established the presence of two related 4-C-branched monosaccharides within LOS-II to IV sequences, of which one was never identified before. In addition, we provided evidence that LOSs are capable of inhibiting the secretion of tumor necrosis factor-α in lipopolysaccharide-stimulated human macrophages. This unexpected finding suggests that these cell wall-associated glycolipids represent key effectors capable of interfering with the establishment of a pro-inflammatory response.A key feature of all members of the genus Mycobacterium is a cell wall of unique and complex structure, which plays an important role in antibiotic resistance and pathogenesis of mycobacteria by modulating the host immune system and phagocytic cell functions (1). The mycobacterial cell wall includes essentially two types of lipids, the mycolic acids, which are very long chain fatty acids covalently bound to the arabinogalactan polysaccharide attached to a peptidoglycan backbone (2), and a vast array of extractable lipids/glycolipids (3). The latter include the ubiquitous trehalose dimycolate (TDM)3 and phosphatidyl mannosides (PIM) (4) as well as a vast array of species-specific lipids such as phenol glycolipids (5), phthiocerol dimycocerosates (5), sulfolipids (4), glycopeptidolipids, and lipooligosaccharides (LOSs).LOSs were found and described in Mycobacterium kansasii (68), Mycobacterium gastri (8, 9), Mycobacterium szulgai (10), Mycobacterium malmoense (11), Mycobacterium gordonae (12), Mycobacterium butyricum (13), Mycobacterium mucogenicum (14), the Canetti variant of Mycobacterium tuberculosis (15) and, more recently in Mycobacterium marinum (Mma) (16). However, they remain among the less studied mycobacterial glycolipids at a biosynthetic, structural, and functional point of view. To date, only three genes have been experimentally demonstrated to be involved in the late steps of LOS biosynthesis in M. marinum (16, 17), and one gene encodes a polyketide synthase responsible for the synthesis of the polymethyl-branched fatty acid in the Mycobacterium smegmatis LOS (18).LOSs represent highly antigenic glycoconjugates exposed to the cell surface and useful target molecules for serotyping in a given mycobacterial species. Their precise role in mycobacteria virulence as well as in the colony morphology remains unclear (19, 20). Early studies demonstrated that rough variants of M. kansasii, devoid of all LOSs, induce chronic systemic infections in mice, whereas smooth variants containing LOSs are rapidly cleared from the organs of infected animals (19, 21). It was therefore proposed that LOSs may act as avirulence factors by masking other cell wall-associated virulence factors. Accordingly, LOSs are absent in most clinical isolates of M. tuberculosis as well as in the laboratory strain H37Rv. A recent genetically based comparison of the LOS biosynthetic cluster in M. marinum and M. tuberculosis revealed that only about one-third of the genes are conserved between the two species, with the genetic locus of M. tuberculosis H37Rv containing fewer genes (17). Although recent studies suggested a possible role of LOSs in sliding motility, biofilm formation, and infection of macrophages by M. marinum (17), the precise contribution of LOSs to M. marinum pathogenesis or virulence is seriously hampered by the restricted number of isogenic strains deficient in their production and the lack of precise structural data of LOS variants. LOSs from different mycobacterial species exhibit considerable variations in the glycan core. A previous work identified the presence of four major LOS variants in M. marinum, designated LOS-I to LOS-IV (16). Through partial characterization, the structure of LOS-I was previously established as 3-O-Me-Rhap-(1–3)-Glcp-(1–3)-Glcp-(1–4)-Glcp-(1–1)-Glcp. Although all LOSs were shown to contain this common oligosaccharidic core substituted by an additional Xylp unit, LOS-II, -III, and -IV are further substituted by other unidentified monosaccharides, designated X and YZ, which leave their exact sequence largely unknown (16).In this study, we report the identification of a natural mutant of M. marinum, devoid of LOS-IV production, which allowed the production of large amounts of LOS-III and the determination of the fine structure of all LOSs. In addition, the availability of all LOS variants with defined structures has opened the possibility to undertake structure-function relationship studies. These molecules were therefore used in in vitro assays to uncover their potent biological roles.  相似文献

Caseinolytic (Clp) proteases are widespread energy-dependent proteases; the functional ATP-dependent protease is comprised of multimers of proteolytic and regulatory subunits. Mycobacterium tuberculosis has two ClpP proteolytic subunits (ClpP1 and ClpP2), with both being essential for growth in vitro. ClpP1 and clpP2 are arranged in an apparent operon; we demonstrated that the two genes are co-expressed under normal growth conditions. We identified a single promoter region for the clpP1P2 operon; no promoter was detected upstream of clpP2 demonstrating that independent expression of clpP1 and clpP2 was highly unlikely. Promoter activity was not induced by heat shock or oxidative stress. We identified a regulatory region upstream of the promoter with a consensus sequence matching the ClgR regulator motif; we determined the limits of the region by mutagenesis and confirmed that positive regulation of the promoter occurs in M. tuberculosis. We developed a reporter system to monitor ClpP1 and ClpP2 enzymatic activities based on LacZ incorporating ssrAtag sequences. We showed that whilst both ClpP1 and ClpP2 degrade SsrA-tagged LacZ, ClpP2 (but not ClpP1) degrades untagged proteins. Our data suggest that the two proteolytic subunits display different substrate specificities and therefore have different, but overlapping roles in M. tuberculosis.  相似文献

Intraspecific nest parasitism in two colonies of Spotless Starling Sturnus unicolor breeding in nestboxes was studied in central Spain from 1991 to 1994. Nests were monitored regularly and three criteria were used to detect nest parasitism: the appearance of more than one egg per day during the laying period of the host; the appearance of an egg after the start of incubation; eggs with unusual shape or pigmentation. The proportion of parasitized nests in first clutches (37%) was twice that of intermediate (19%) or second (20%) clutches in colony B, whereas parasitism occurred in first (35%) and intermediate (12%) but not in second clutches in colony A. Most clutches (52–70%) were parasitized during the host's laying period and received one parasitic egg. In 10% of the parasitized clutches in colony B, one of the host's eggs disappeared on the day the parasitic egg was added, suggesting that the parasitic female removed this egg. Although parasitism increased clutch size significantly, it led to a decrease in host breeding success, mainly through the removal of eggs and the loss of host nestlings and the survival of parasitic chicks. Observations suggested that parasitic females were young individuals without their own nests and/or those whose breeding attempt had been disrupted while laying in their own nest.  相似文献

Climate change is predicted to cause changes in species distributions and several studies report margin range shifts in some species. However, the reported changes rarely concern a species' entire distribution and are not always linked to climate change. Here, we demonstrate strong north‐eastwards shifts in the centres of gravity of the entire wintering range of three common waterbird species along the North‐West Europe flyway during the past three decades. These shifts correlate with an increase of 3.8 °C in early winter temperature in the north‐eastern part of the wintering areas, where bird abundance increased exponentially, corresponding with decreases in abundance at the south‐western margin of the wintering ranges. This confirms the need to re‐evaluate conservation site safeguard networks and associated biodiversity monitoring along the flyway, as new important wintering areas are established further north and east, and highlights the general urgency of conservation planning in a changing world. Range shifts in wintering waterbirds may also affect hunting pressure, which may alter bag sizes and lead to population‐level consequences.  相似文献

Infection of the zebrafish with Mycobacterium marinum is regarded as a well-established experimental model to study the pathogenicity of Mycobacterium tuberculosis. Herein, a M. marinum transposon mutant library was screened for attenuated M. marinum phenotypes using a Dictyostelium discoideum assay. In one attenuated mutant, the transposon was located within tesA, encoding a putative type II thioesterase. Thin-layer chromatography analyses indicated that the tesA::Tn mutant failed to produce two major cell wall-associated lipids. Mass spectrometry and nuclear magnetic resonance clearly established the nature of missing lipids as phthioglycol diphthioceranates and phenolic glycolipids, respectively, indicating that TesA is required for the synthesis of both lipids. When injected into the zebrafish embryo bloodstream, the mutant was found to be highly attenuated, thus validating the performance and relevance of the Dictyostelium screen. Consistent with these in vivo findings, tesA::Tn exhibited increased permeability defects in vitro, which may explain its failure to survive in host macrophages. Unexpectedly, virulence was retained when bacteria were injected into the notochord. Histological and ultrastructural studies of the infected notochord revealed the presence of actively proliferating mycobacteria, leading to larval death. This work presents for the first time the notochord as a compartment highly susceptible to mycobacterial infection.  相似文献

Background

Worldwide, amoebic liver abscess (ALA) can be found in individuals in non-endemic areas, especially in foreign-born travelers.

Methods

We performed a retrospective analysis of ALA in patients admitted to French hospitals between 2002 and 2006. We compared imported ALA cases in European and foreign-born patients and assessed the factors associated with abscess size using a logistic regression model.

Results

We investigated 90 ALA cases. Patient median age was 41. The male:female ratio was 3.5∶1. We were able to determine the origin for 75 patients: 38 were European-born and 37 foreign-born. With respect to clinical characteristics, no significant difference was observed between European and foreign-born patients except a longer lag time between the return to France after traveling abroad and the onset of symptoms for foreign-born. Factors associated with an abscess size of more than 69 mm were being male (OR = 11.25, p<0.01), aged more than 41 years old (OR = 3.63, p = 0.02) and being an immigrant (OR = 11.56, p = 0.03). Percutaneous aspiration was not based on initial abscess size but was carried out significantly more often on patients who were admitted to surgical units (OR = 10, p<0.01). The median time to abscess disappearance for 24 ALA was 7.5 months.

Conclusions/Significance

In this study on imported ALA was one of the largest worldwide in terms of the number of cases included males, older patients and foreign-born patients presented with larger abscesses, suggesting that hormonal and immunological factors may be involved in ALA physiopathology. The long lag time before developing ALA after returning to a non-endemic area must be highlighted to clinicians so that they will consider Entamoeba histolytica as a possible pathogen of liver abscesses more often.  相似文献

Background

In order to inform meningococcal disease prevention strategies, we analysed the epidemiology of invasive meningococcal disease (IMD) in the province of Quebec, Canada, 10 years before and 10 years after the introduction of serogroup C conjugate vaccination.

Methodology

IMD cases reported to the provincial notifiable disease registry in 1991–2011 and isolates submitted for laboratory surveillance in 1997–2011 were analysed. Serogrouping, PCR testing and assignment of isolates to sequence types (ST) by using multilocus sequence typing (MLST) were performed.

Results

Yearly overall IMD incidence rates ranged from 2.2–2.3/100,000 in 1991–1992 to 0.49/100,000 in 1999–2000, increasing to 1.04/100,000 in 2011. Among the 945 IMD cases identified by laboratory surveillance in 1997–2011, 68%, 20%, 8%, and 3% were due to serogroups B, C, Y, and W135, respectively. Serogroup C IMD almost disappeared following the implementation of universal childhood immunization with monovalent C conjugate vaccines in 2002. Serogroup B has been responsible for 88% of all IMD cases and 61% of all IMD deaths over the last 3 years. The number and proportion of ST-269 clonal complex has been steadily increasing among the identified clonal complexes of serogroup B IMD since its first identification in 2003, representing 65% of serogroup B IMD in 2011. This clonal complex was first introduced in adolescent and young adults, then spread to other age groups.

Conclusion

Important changes in the epidemiology of IMD have been observed in Quebec during the last two decades. Serogroup C has been virtually eliminated. In recent years, most cases have been caused by the serogroup B ST-269 clonal complex. Although overall burden of IMD is low, the use of a vaccine with potential broad-spectrum coverage could further reduce the burden of disease. Acceptability, feasibility and cost-effectiveness studies coupled with ongoing clinical and molecular surveillance are necessary in guiding public policy decisions.  相似文献
