(1) Neurogenesis driven by neural stem cells (NSCs) is regulated by physiological and pathological factors. Melatonin (MT)
has profound neurotrophic and neuroprotective effects. Hence, we studied the role of MT in regulating the viability and differentiation
of NSCs derived from rat ventral midbrain. (2) NSCs were isolated from the rat ventral midbrain. The viability of NSCs was
determined by 3-(4,5-dimethylthiazol-2-yl)-5-(3-carboxymethoxyphenyl)-2-(4-ulfophenyl)-2H-tetrazolium assay. The differentiation
of NSCs was examined by analyzing the expression of the neural markers, MT receptors, brain derived neurotropic factor (BDNF)
and glial cell line-derived neurotrophic factor (GDNF) with semi-quantitative RT-PCR, immunofluorescence cytochemistry, and
Western blot. (3) Our results showed that MT could promote the viability of NSCs. In addition, MT could significantly elevate
the mRNA and protein levels of tyroxine hydroxylase (TH), a marker of dopaminergic neurons, and decrease the expression of
the astrocytes maker glial fibrillary acidic protein (GFAP). MT also increased the production of BDNF and GDNF in the cultured
NSCs. Meanwhile, we first found that two subtypes of MT receptors, MT1 and MT2, were expressed in the ventral midbrain NSCs.
(4) These results demonstrated that MT could induce NSCs to differentiate into dopaminergic neurons and decrease astrocyte
production. These findings also suggest that MT could offer a beneficial tool in guiding directional differentiation of NSCs. 相似文献

铜绿假单胞菌是临床上常见致病菌, 传统的检测方法有各种弊端。本研究对该细菌的ETA基因用生物信息学方法加以分析, 选取相对保守且高度特异的DNA序列, 设计一对特异性引物和一个TaqMan探针, 建立FQ-PCR (fluorescence quantitative PCR)检测PA的方法。通过对梯度浓度的铜绿假单胞菌基因组DNA样品进行FQ-PCR检测和对多种细菌的DNA进行扩增, 来检测其灵敏度和验证引物和探针的特异性。试验结果表明, 对比现有的检测方法, 以ETA基因为靶基因, 基于TaqMan探针的快速FQ-PCR检测技术有更高的灵敏度和更好的特异性等优点, 具有很好的研究价值和应用前景。 相似文献

Juvenile hemochromatosis is a severe hereditary iron overload disease caused by mutations in the HJV (hemojuvelin) and HAMP (hepcidin) genes. Hepcidin is an important iron regulatory hormone, and hemojuvelin may regulate hepcidin synthesis via the multifunctional membrane receptor neogenin. We explored the expression of murine hemojuvelin and neogenin mRNAs and protein. Real-time RT-PCR analysis of 18 tissues from male and female mice was performed to examine the mRNA expression profiles. To further study protein expression and localization we used immunohistochemistry on several tissues from three mouse strains. Mouse Neo1 mRNA was detectable in the 18 tissues tested, the highest signals being evident in the ovary, uterus, and testis. Neogenin protein was observed in the brain, skeletal muscle, heart, liver, stomach, duodenum, ileum, colon, renal cortex, lung, testis, ovary, oviduct, and uterus. The spleen, thymus, and pancreas were negative for neogenin. The highest signals for Hjv mRNA were detectable in the skeletal muscle, heart, esophagus, and liver. The results indicate that Neo1 mRNA is widely expressed in both male and female mouse tissues with the highest signals detected in the reproductive system. Moreover, Hjv and Neo1 mRNAs are simultaneously expressed in skeletal muscle, heart, esophagus, and liver. 相似文献

The transwell chamber migration assay and CCD digital camera imaging techniques were used to investigate the relationship between regulatory volume decrease (RVD) and cell migration in nasopharyngeal carcinoma cells (CNE-2Z cells). Both migrated and non-migrated CNE-2Z cells, when swollen by 47% hypotonic solution, exhibited RVD which was inhibited by extracellular application of chloride channel blockers adenosine 5‘-triphosphate (ATP), 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) and tamoxifen. However, RVD rate in migrated CNE-2Z cells was bigger than that of non-migrated cells and the sensitivity of migrated cells to NPPB and tamoxifen was higher than that of nonmigrated cells. ATP, NPPB and tamoxifen also inhibited migration of CNE-2Z cells. The inhibition of migration was positively correlated to the blockage of RVD, with a correlation coefficient (r) = 0.99, suggesting a functional relationship between RVD and cell migration. We conclude that RVD is involved in cell migration and RVD may play an important role in migratory process in CNE-2Z cells. 相似文献

为了研究盐酸双氟沙星(difloxacin, DIF)在异育银鲫体内血浆蛋白结合率的变化及其与药代动力学之间的关系, 实验采用超滤法测定了DIF在异育银鲫体内血浆蛋白结合率, 运用HPLC测定其对应时间点药物浓度, 并分析了血浆蛋白结合率变化对DIF体内处置的影响。实验以感染嗜水气单胞菌的异育银鲫为感染组, 健康异育银鲫为对照组。结果显示: 感染组各时间点DIF血浆蛋白结合率均高于对照组, 感染组与对照组DIF血浆蛋白结合率与总药物浓度呈对数关系: y=-9.01ln x+74.34和y=-4.81ln x+65.15, DIF血浆蛋白结合率与游离药物浓度的对数关系式分别为: y=-6.36ln x+64.91和y=-4.36ln x+ 60.63; 感染组和对照组血浆药时曲线均可使用开放性二室模型描述; 感染组DIF的吸收和消除慢于对照组, 其表观分布容积和曲线下面积大于对照组。结果显示异育银鲫体内感染嗜水气单胞菌促使DIF血浆蛋白结合率升高; 血浆蛋白结合率升高导致药物以结合药物的形式储存于血液中可能是导致药物组织分布受限、消除缓慢、长时间滞留于血液原因之一。 相似文献

运用双向聚丙烯酰胺凝胶电泳(2DPAGE)分析未交配小鼠子宫内膜和妊娠第五天(D5)小鼠子宫内膜胚泡黏附时植入位点及其旁组织蛋白质组。差异蛋白质组学显示,等电点(isoelectric point,pI)约7.1、分子量(molecular weight,Mw)约18kDa的蛋白质点在D5小鼠子宫内膜特别是植入位点表达上调。对此蛋白质点用基质辅助激光解析电离飞行时间质谱(matrix-assisted laser desorption/ionization time of flying mass spectrometry,MALDI—TOF—MS)测定其胶内酶解后的肽质量指纹谱(Peptide Mass Fingerprint,PMF),经Mascot:Peptide Mass Fingerprint中SWISS-PROT数据库查询后,鉴定该蛋白质为鼠源性nm23-M2/NDPKB。RT—PCR和免疫组织化学结果也显示D5小鼠子宫内膜nm23-M2/NDPK B mRNA和蛋白表达明显增加。提示nm23-M2/NDPKB参与胚泡着床这一重要生命活动过程。 相似文献

本文对金缕梅科弗特吉族(Molinadendron和Matudaea除外)的细胞核核糖体DNA ITS片段进行了序列分析,在此基础上对该族的演化关系、尤其是中国特有属银缕梅属的系统位置进行了最简约分支分析。基本结果如下:其一,蚊母族和狭义的弗特吉族均不形成自己的单系分支,因此,支持Endress的处理,即合并两族为广义的弗特吉族;其二,银缕梅属同帕罗堤属有着密切的亲缘关系,支持形态学和解剖学结论;其三,根据该片段核苷酸序列的演化速率推算出的银缕梅属和帕罗堤属间分化时间为晚中新世。此结论大致与化石记录相符。 相似文献
