Methicillin‐resistant Staphylococcus aureus (MRSA) emerged via acquisition of a mobile element, staphylococcal cassette chromosome mec (SCCmec). Integration and excision of SCCmec is mediated by an unusual site‐specific recombination system. Most variants of SCCmec encode two recombinases, CcrA and CcrB, that belong to the large serine family. Since CcrA and CcrB are always found together, we sought to address their specific roles. We show here that CcrA and CcrB can carry out both excisive and integrative recombination in Escherichia coli in the absence of any host‐specific or SCCmec‐encoded cofactors. CcrA and CcrB are promiscuous in their substrate choice: they act on many non‐canonical pairs of recombination sites in addition to the canonical ones, which may explain tandem insertions into the SCCmec attachment site. Moreover, CcrB is always required, but CcrA is only required if one of the four half‐sites is present. Recombinational activity correlates with DNA binding: CcrA recognizes only that half‐site, which overlaps a conserved coding frame on the host chromosome. Therefore, we propose that CcrA serves as a specificity factor that emerged through modular evolution to enable recognition of a bacterial recombination site that is not an inverted repeat.  相似文献

Abstract

The feasibility of using reversed-phase high performance liquid chromatography (RP-HPLC) for the separation of metallothioneins (MTs) and subsequent determination of cadmium in MTs by graphite furnace atomic absorption spectrometry (GFAAS) in rabbit kidney and liver has been studied. RP-HPLC was used to isolate, characterise and quantitate liver and kidney MT isoforms. The MTs were eluted from a radially compressed C18 column with a neutral sodium phosphate buffer and detected by UV absorbance at 254 nm. Rabbit liver MTs was found to be comprised of seven distinct isoforms with five of which were found to be subspecies of the MT-I isoform. Rabbit kidney MTs exhibited only two predominant isoforms. A standard calibration curve was constructed using purified rabbit kidney MT-I and MT-II which demonstrated excellent linear correlation between peak height and the quantity of MT injected into the column. Recovery of MT from RP-HPLC was found to exceed 90%. Kidney and liver tissues from rabbit by feeding low levels of cadmium in diets was assayed using the RP-HPLC analysis of cytosol samples. Feeding stable cadmium in the diet resulted in the deposition of MT in the kidney rather than in the liver. The cadmium content in MT isoforms was determined by GFAAS. Less than 10% of the total cadmium in kidney was associated with MTs.  相似文献

Measuring yeast biomass is important in the processes of microbial fermentations. It has been demonstrated that synchronous light scattering (SLS) signals could be applied for the quantification of model bioparticles such as Saccharomyces cerevisiae. In this study, an improved synchronous light scattering method was developed for yeast biomass estimation. The settlement of yeast cells during SLS signals measuring process was studied, and hydrolysis anionic polyacrylamide was added into yeast suspensions to increase the stability of the cells in liquid environment. By simultaneously scanning both the excitation and emission monochromators of a common spectrofluorometer with same starting excitation and emission wavelength (namely, ?λ = 0), the SLS intensity was found to be proportional to the yeast concentration in the range from 0 to 4.9 × 106 cell/mL (R 2 = 0.9907), the detection limit is 8.1 × 103 cell/mL. The developed method exhibited good stability and sensitivity in the recovery test and growth curve drawing process, demonstrating the potential of the method in practical application of biomass estimation.  相似文献

Spinach has long been used as a model for genetic and physiological studies of sex determination and expression. Although trisomic analysis from a cross between diploid and triploid plants identified the XY chromosome as the largest chromosome, no direct evidence has been provided to support this at the molecular level. In this study, the largest chromosomes of spinach from mitotic metaphase spreads were microdissected using glass needles. Degenerate oligonucleotide primed polymerase chain reaction was used to amplify the dissected chromosomes. The amplified products from the Y chromosome were identified using the male-specific marker T11A. For the first time, the largest spinach chromosome was confirmed to be a sex chromosome at the molecular level. PCR products from the isolated chromosomes were used in an in situ probe mixture for painting the Y chromosome. The fluorescence signals were mainly distributed on all chromosomes and four pair of weaker punctate fluorescence signal sites were observed on the terminal region of two pair of autosomes. These findings provide a foundation for the study of sex chromosome evolution in spinach.  相似文献

The investigation and development of the cancer stem cell (CSC) model has received much focus during these years. CSC is characterized as a small fraction of cancer cells that have an indefinite ability for self-renewal and pluripotency and are responsible for initiating and sustaining of the bulk of cancer. So, whether current treatment strategies, most of which target the rapid division of cancer cells, could interfere with the slow-cycling CSCs is broadly questioned. Meanwhile, however, the new understanding of tumorigenesis has led to the development of new drug screening strategies. Both stem cells and mesenchymal stem cells have been vigorously used in pre-clinical studies of their anti-tumor potential, mainly due to their inherent tumoritropic migratory properties and their ability to carry anti-tumor transgenes. Here, based on the tumorigenic and tumoritropic characteristics of CSCs, we proposed two hypotheses exploring possible usage of CSCs as novel anti-tumor agents and potential sources for tissue regeneration. Further experimental validation of these hypotheses may unravel some new research topics.  相似文献

Two samples (YC7, YC27) of Nicotiana tabacum showing leaf curling, vein swelling and enations on undersides of leaves were collected in the Fujian Province of China in 2007. Virus isolates YC7‐1 and YC7‐2 (associated with betasatellite, YC7‐2β) were detected in both samples. The complete DNA‐A sequence of YC7‐1 (FJ869907) comprised 2741 nucleotides (nt). The complete DNA‐A (FJ869908) and betasatellite (FJ869909) sequence of YC7‐2 consisted of 2754 and 1344 nt, respectively. YC7‐1 had the highest nucleotide sequence identity (97.3%) with Papaya leaf curl Guangdong virus (PaLCuGuV‐[CN:Gd2:02], AJ558122). YC7‐2 had the highest sequence identity (90.1%) with Ageratum yellow vein virus (AYVV‐TW[TW:Tai:99], AF307861) and its betasatellite (96.5%) with Ageratum yellow vein betasatellite (AYVB‐[TW:CHu:02], AJ542495). These indicate that YC7‐1 and YC7‐2 are isolates of PaLCuGuV and AYVV, respectively. Symptoms including leaf curling, vein swelling and enations on undersides of leaves were observed in N. tabacum and N. glutinosa when infected by whiteflies with sample YC7 as the viral source under greenhouse conditions. PCR results showed that these infected plants contained both YC7‐1 and YC7‐2/YC7‐2β. To our knowledge, this is the first report of PaLCuGuV and AYVV/AYVB co‐infecting N. tabacum in China.  相似文献

目的探讨微卫星在转基因和基因突变小鼠中的变化,为基因修饰和遗传突变动物的遗传检测和表型分析提供理论依据和技术手段。方法根据文献报道,从GenBank中选取198个等位基因数量多、富含多态性的微卫星位点,以野生型动物为对照,对6种近交系遗传背景的转基因小鼠和5种自然基因突变的近交系小鼠进行微卫星多态性检测,选用1.5%琼脂糖凝胶电泳和STR扫描技术,比较分析微卫星不稳定性。结果共有40个微卫星位点在转基因和基因突变小鼠中表现出多态性。在基因突变小鼠中,微卫星不稳定性有55.6%(10/18)是由纯合变为杂合(Ⅰ型),有3个位点(16.6%,3/18)是纯合突变(Ⅱ型),有5个位点同时存在2种类型的突变。但是在转基因动物中,大多数的微卫星多态性为Ⅰ型突变(87.5%,28/32),只有2个位点(6.2%,2/32)是Ⅱ型突变。另外有2个位点同时存在2种类型的突变。结论基因修饰或基因突变可引起小鼠相关微卫星发生不稳定性,而且某些微卫星位点对基因改变敏感性较高。  相似文献

RT-PCR扩增猕猴黄嘌呤脱氢酶/氧化酶(XDH/XO)基因片段,为进一步开展相关研究提供实验资料。方法提取猕猴新鲜肝脏组织总RNA,用RT-PCR二步法进行XDH/XO基因片段扩增,对获得的目的片段进行序列测定,与GenBank上发表的人类(Homosapiens)、小鼠(Musmusculus)、家鼠(Rattusnorvegicus)、野猪(Susscrofa)等物种XDH/XO基因进行该序列同源性比对分析,DNAMAN软件预测该段核苷酸的氨基酸序列,Inter-ProScan及SWISS-MODEL工具进行XDH/XO的编码蛋白结构域及功能预测及三维结构构建。结果RT-PCR产物电泳检测得到了与设计大小相一致的目的条带,序列测定共测到683个核苷酸,DNAMAN软件预测该段核苷酸的氨基酸序列包括了1个编码53个氨基酸的开放阅读框(ORF),通过该软件包中Multiplealignment对目的基因片段的核苷酸序列与NCBI报道的人类、小鼠、家鼠、野猪XDH/XO基因mRNA互补的cDNA核苷酸序列同源性进行同源性比较分析,结果显示所扩增得到的目的片段与人类同源性最高,为95.6%,与小鼠、家鼠、野猪的同源性分别为85.2%、84.3%、86.1%,说明获得的基因片段是猕猴的XDH/XO基因片段,且该基因在物种间具有较高的相似性。生物信息学预测该段XDH/XO编码蛋白含有醛氧化/脱氢酶的钼喋呤结合点结构域及黄嘌呤脱氢酶结构域。结论在体外成功扩增出猕猴XDH/XO基因片段,为进一步开展高尿酸血症致病机理研究,抗高尿酸血症新药研发奠定工作基础。  相似文献

观察联合应用siRNA对HepG2.2.15细胞中HBV抗原表达和复制的抑制作用。应用ELISA方法检测HBeAg和HBsAg;HBVDNA水平用实时定量PCR测定;用RT—PCR检测HBVmRNA水平。结果显示,实验中应用的HBV特异性siRNA均具有明显的抗HBV抗原表达和病毒复制作用;联合应用siRNA较单独应用具有更强的抗HBV作用。可见,HepG2.2.15细胞中联合应用siRNA对HBV复制的抑制作用比单独应用siRNA更有效。  相似文献

microRNAs是一种小分子非编码RNA,广泛参与细胞的生长发育、分化和凋亡等生物学过程. microRNA-21(miR-21)是一个在实体肿瘤中高表达的miRNA,可促进癌细胞增殖. 为了研究miR-21在金华猪生长过程中的作用,本试验选取了金华猪80日龄胚胎的心、肝、脾、肺、肾、小肠、胰脏、皮脂组织,进行miR-21组织表达谱研究,结果发现miR-21在肝脏中的表达最高. 进一步以肝脏为对象,研究miR-21在金华猪不同生长阶段中的表达,结果显示胚胎80日龄的miR-21表达量最高(P<0.01),其次是胚胎期的60日龄和105日龄,出生后1至120日龄miR-21的表达量均维持在较低的水平且相互之间差异不显著(P>0.05). 利用KEGG软件对miR-21预测的靶基因进行通路归类分析表明miR-21参与细胞代谢、生长等过程,从中选择两个抑制细胞增殖的靶基因,增强子结合蛋白(CCAAT/enhancer binding protein,Cebpa) 和原肌球蛋白1基因(tropomyosin 1,Tpm1),qRT-PCR检测二者在不同生长阶段的mRNA表达量,结果其表达趋势与miR-21相反,其中胚胎期80天表达量最低(P<0.05). 综上表明miR-21可能具有促进胚胎肝脏生长的作用.  相似文献
