以云南西双版纳国家级自然保护区尚勇子保护区内亚洲象种群为研究对象,通过自动红外照相技术,估算了该地区亚洲象的最小种群数量。本研究于2016年1月,在研究区域内共布设了27个相机位点,野外安放时间为4个月。调查期间,每台相机的有效捕获日为9-52d不等(均值为24d),红外相机有效捕获日621d,拍摄到亚洲象照片共1944张。通过红外相机照片开展个体识别,最终估算出尚勇保护区内亚洲象的最小种群数量为69头(其中成年象38头,亚成象15头,幼象16头)。拍到7头活动于中国-老挝边境区域的跨境象群。本文探讨利用红外相机拍摄的亚洲象照片进行个体识别的方法,指出与常规调查方法相比较的优势和不足,作为快速、实时有效的种群评估方法的价值。研究结果丰富了保护区内亚洲象种群数据库,为研究、制定和开展亚洲象保护行动提供重要支撑。 相似文献

将合成的人胰高血糖素样肽-1(glucagon-like peptide-1,GLP-1)突变体基因与IgG4抗体的Fc部分进行融合获得GLP-1-IgG4-Fc片段,获得的基因片段与pXC17.4载体进行连接,用电转化方法将线性化质粒稳定转染CHO-K1细胞,通过Clone Pix 2筛选出高表达细胞株,产量达1.5g/L。收集培养上清并经Protein A和Source 30Q纯化,得到的GLP-1-IgG4-Fc融合蛋白,SDS-PAGE纯度高于95%,高效液相色谱(high performance liquid chromatography,HPLC)纯度和毛细管区域凝胶电泳(capillary zone electrophoresis,CZE)纯度均不低于80%,尺寸排阻层析(size-exclusion chromatography,SEC)纯度高于99%。经质谱和肽图谱测定,分子量与理论值一致,肽图谱序列与对照品高度一致。生物学活性分析表明,GLP-1-IgG4-Fc融合蛋白具有促进表达有GLP-1受体的HEK293细胞分泌环磷酸腺苷(cyclic adenosine monophosphate,c AMP)的活性,并且该活性与对照品高度相似。 相似文献

Cell-wall deposition of cellulose microfibrils is essential for plant growth and development. In plant cells,cellulose synthesis is accomplished by cellulose synthase complexes located in the plasma membrane. Trafficking of the complex between endomembrane compartments and the plasma membrane is vital for cellulose biosynthesis;however, the mechanism for this process is not well understood. We here report that, in Arabidopsis thaliana,Rab-H_1b, a Golgi-localized small GTPase, participates in the trafficking of CELLULOSE SYNTHASE 6(CESA6) to the plasma membrane. Loss of Rab-H_1b function resulted in altered distribution and motility of CESA6 in the plasma membrane and reduced cellulose content. Seedlings with this defect exhibited short, fragile etiolated hypocotyls.Exocytosis of CESA6 was impaired in rab-h1 b cells, and endocytosis in mutant cells was significantly reduced as well. We further observed accumulation of vesicles around an abnormal Golgi apparatus having an increased number of cisternae in rab-h1 b cells, suggesting a defect in cisternal homeostasis caused by Rab-H_1b loss function. Our findings link Rab GTPases to cellulose biosynthesis, during hypocotyl growth, and suggest Rab-H_1b is crucial for modulating the trafficking of cellulose synthase complexes between endomembrane compartments and the plasma membrane and for maintaining Golgi organization and morphology.e 相似文献

采用石蜡切片法对马哈利樱桃大孢子发生和雌配子体发育过程进行观察研究。结果表明:(1)马哈利樱桃雌配子体发育早期,在单室子房内可以看到2个倒生胚珠,但在后期其中一个退化,另一个发育为种子;其胚珠具双珠被,为厚珠心。(2)大孢子母细胞减数分裂形成直线型四分体,功能大孢子位于合点端;胚囊发育为蓼型,成熟胚囊为七细胞八核。(3)根据不同时间花的外部形态特征与内部解剖学对比的观察结果,在陕西关中地区,三月下旬是马哈利樱桃雌性生殖细胞分化和发育的重要时期,果园在此期间应加强肥水管理。 相似文献

The toxic metal ion cadmium (Cd2+) induces pleiotropic effects on cell death and survival, in part through effects on cell signaling mechanisms and cytoskeletal dynamics. Linking these phenomena appears to be calmodulin‐dependent activation of the Ca2+/calmodulin‐dependent protein kinase II (CaMK‐II). Here we show that interference with the dynamics of the filamentous actin cytoskeleton, either by stabilization or destabilization, results in disruption of focal adhesions at the ends of organized actin structures, and in particular the loss of vinculin and focal adhesion kinase (FAK) from the contacts is a result. Low‐level exposure of renal mesangial cells to CdCl2 disrupts the actin cytoskeleton and recapitulates the effects of manipulation of cytoskeletal dynamics with biological agents. Specifically, Cd2+ treatment causes loss of vinculin and FAK from focal contacts, concomitant with cytoskeletal disruption, and preservation of cytoskeletal integrity with either a calmodulin antagonist or a CaMK‐II inhibitor abrogates these effects of Cd2+. Notably, inhibition of CaMK‐II decreases the migration of FAK‐phosphoTyr925 to a membrane‐associated compartment where it is otherwise sequestered from focal adhesions in a Cd2+‐dependent manner. Fatty acyl reductases (FARs) are key enzymes that participate in sex pheromone biosynthesis by reducing fatty acids to fatty alcohols. Lepidoptera typically harbor numerous FAR gene family members. Although FAR genes are involved in the biosynthesis of sex pheromones in moths, the key FAR gene of Spodoptera litura remains unclear. In this work, we predicted 30 FAR genes from the S. litura genome and identified a domain duplication within gene SlitFAR3, which exhibited high and preferential expression in the sexually mature female pheromone glands (PGs) and a rhythmic expression pattern during the scotophase of sex pheromone production. The molecular docking of SlitFAR3, as predicted using a 3D model, revealed a co-factor NADPH binding cavity and 2 substrate binding cavities. Functional expression in yeast cells combined with comprehensive gas chromatography indicated that the SlitFAR3 gene could produce fatty alcohol products. This study is the first to focus on the special phenomenon of FAR domain duplication, which will advance our understanding of biosynthesis-related genes from the perspective of evolutionary biology. 相似文献

Background
Glucose-6-phosphate dehydrogenase (G6PD) deficiency and hemoglobinopathies were the inherited conditions found mostly in African. However, few epidemiological data of these disorders was reported in Equatorial Guinea (EQG). This study aimed to assess the prevalence and healthy effects of G6PD deficiency and hemoglobinopathies among the people on malaria endemic Bioko Island, EQG.Materials and Methods
Blood samples from 4,144 unrelated subjects were analyzed for G6PD deficieny by fluorescence spot test (FST), high-resolution melting assay and PCR-DNA sequencing. In addition, 1,186 samples were randomly selected from the 4,144 subjects for detection of hemoglobin S (HbS), HbC, and α-thalassemia deletion by complete blood count, PCR-DNA sequencing and reverse dot blot (RDB).Results
The prevalence of malaria and anemia was 12.6% (522/4,144) and 32.8% (389/1,186), respectively. Overall, 8.7% subjects (359/4,144) were G6PD-deficient by FST, including 9.0% (249/2,758) males and 7.9% (110/1,386) females. Among the 359 G6PD-deficient individuals molecularly studied, the G6PD A- (G202A/A376G) were detected in 356 cases (99.2%), G6PD Betica (T968C/A376G) in 3 cases. Among the 1,186 subjects, 201 cases were HbS heterozygotes, 35 cases were HbC heterozygotes, and 2 cases were HbCS double heterozygotes; 452 cases showed heterozygous α-thalassemia 3.7 kb deletion (-α3.7 kb deletion) and 85 homozygous - α3.7 kb deletion. The overall allele frequencies were HbS 17.1% (203/1186); HbC, 3.1% (37/1186); and –α3.7 kb deletion 52.4% (622/1186), respectively.Conclusions
High G6PD deficiency in this population indicate that diagnosis and management of G6PD deficiency is necessary on Bioko Island. Obligatory newborn screening, prenatal screening and counseling for these genetic disorders, especially HbS, are needed on the island. 相似文献999.
