BMP2/7 heterodimer enhances osteogenic differentiation of rat BMSCs via ERK signaling compared with respective homodimers

Bone morphogenetic protein (BMP)2/7 heterodimer shows greater efficacy in enhancing bone regeneration. However, the precise mechanism and the role of mitogen-activated protein kinase (MAPK) signaling network in BMP2/7-driven osteogenesis remain ambiguous. In this study, we evaluated the effects of BMP2/7 heterodimers on osteoblastic differentiation in rat bone marrow mesenchymal stem cells (BMSCs), with the aim to elaborate how MAPKs might be involved in this cellular process by treatment of rat BMSCs with BMP2/-7 with a special signal-pathway inhibitor. We found that BMP2/7 heterodimer induced a much stronger osteogenic response in rat BMSCs compared with either homodimer. Most interestingly, extracellular signal-regulated kinase (ERK) demonstrated a highly sustained phosphorylation and activation in the BMP2/7 heterodimer treatment groups, and inhibition of ERK cascades using U0126 special inhibitor that significantly reduced the activity of ALP and calcium mineralization to a substantial degree in rat BMSCs treated with BMP2/7 heterodimers. Collectively, we demonstrate that BMP2/7 heterodimer shows a potent ability to stimulate osteogenesis in rat BMSCs. The activated ERK signaling pathway involved in this process may contribute partially to an increased osteogenic potency of heterodimeric BMP2/7 growth factors.     

Antibody Profiling by Proteome Microarray with Multiplex Isotype Detection Reveals Overlap between Human and Aotus nancymaae Controlled Malaria Infections

The development of vaccines against malaria and serodiagnostic tests for detecting recent exposure requires tools for antigen discovery and suitable animal models. The protein microarray is a high‐throughput, sample sparing technique, with applications in infectious disease research, clinical diagnostics, epidemiology, and vaccine development. We recently demonstrated Qdot‐based indirect immunofluorescence together with portable optical imager ArrayCAM using single isotype detection could replicate data using the conventional laser confocal scanner system. We developed a multiplexing protocol for simultaneous detection of IgG, IgA, and IgM and compared samples from a controlled human malaria infection model with those from controlled malaria infections of Aotus nancymaae, a widely used non‐human primate model of human malaria. IgG profiles showed the highest concordance in number of reactive antigens; thus, of the 139 antigens recognized by human IgG antibody, 111 were also recognized by Aotus monkeys. Interestingly, IgA profiles were largely non‐overlapping. Finally, on the path toward wider deployment of the portable platform, we show excellent correlations between array data obtained in five independent laboratories around the United States using the multiplexing protocol (R²: 0.60–0.92). This study supports the use of this platform for wider deployment, particularly in endemic areas where such a tool will have the greatest impact on global human health.

体温对沙鼠前脑缺血/再灌注Ca~(2 )/钙调素依赖性蛋白激酶Ⅱ活性变化的影响

目的和方法 :本文以蒙古沙土鼠双侧颈动脉夹闭 (BCAO)形成前脑缺血模型 ,观察不同体温 (3 6.5℃、3 0 .0℃、3 9.0℃ )对脑缺血 /再灌注海马、皮质、纹状体、小脑Ca2 /CaMPKⅡ活性变化的影响。结果 :①除小脑外 ,海马、皮质、纹状体Ca2 /CaMPKⅡ活性在缺血后均有不同程度下降 ,该下降对缺血过程中体温变化十分敏感 :如分别于 3 6.5℃、3 0 .0℃、3 9.0℃时同样缺血 10min ,海马酶活性分别为对照组的 3 2 .2 %、10 1.3 %、9.1% ;②持续一定时间的低体温再灌注可显著促进海马、皮质、纹状体该酶活性的恢复 :如 3 6.5℃和 3 0 .0℃再灌注相同时间后 ,海马酶活性分别为对照组的 5 1.3 %和 5 3 .2 % (4h ,P >0 .0 5 )、5 8.0 %和 68.9% (8h ,P <0 .0 5 )、67.4 %和 84 .1% (16h ,P <0 .0 5 )。结论 :低体温可显著保护脑缺血 /再灌注缺血敏感组织Ca2 /CaMPKⅡ活性 ,这可能与其保护缺血性脑损伤关系密切。

近海潮间带植物生境的微生物是最富有生物多样性的海洋种群,其生境的独特性决定了该区域微生物较高的药用价值.本文介绍了我国南北方海域近海潮间带植物生境和微生物的特性及药用前景,为该区域微生物的开发利用提供参考.     

桃ERF转录因子家族生物信息学分析及 芽萌发相关基因筛选

以中油四号油桃(Prunus persica var. nectarina)为研究对象, 利用MEGA 6.0、MEME、GSDS和DNAMAN 6.0等软件对桃ERF家族数据进行生物信息学分析, 鉴定得到102个ERF转录因子家族基因, 并通过构建系统进化树将这102个基因分为10个子家族(I-X)。基因结构分析表明, 有81个基因不含内含子, 20个基因含有1个内含子, 有1个基因与其它成员差异较大, 含有5个内含子。保守元件分析表明, ERF家族包含20个保守元件, 其中Motif 1、Motif 2和Motif 4都属于AP2/ERF结构域, 同一个保守元件主要出现在同一个子家族中, 并且大部分保守元件的功能未知。VIII子家族基因的荧光定量PCR分析表明, 在桃叶芽处于不同的发育状态时, PpeERF068的表达量存在较大差异, 光照培养箱中培养的桃芽在萌发过程中各时期表达量变化趋势进一步表明该基因可能与叶芽萌发有关, 将其命名为PpeEBB1。该研究为进一步揭示PpeEBB1的分子机制奠定了基础, 并为桃树的栽培管理和熟期调控了提供理论指导。     

鄂东花马湖水生高等植物研究：Ⅰ.水生植物区系

花马湖位于北纬30°15′—30°18′、东经114°58′—115°1′,面积约27.5km~2。湖中分布82种2变种水生维管束植物,隶属于34科66属,其中资源植物众多。花马湖水生高等植物大型属少,小型属较多,单型属相对突出。区系地理成分以温带成分占优势,计34种,占全湖种类的40.48%。起源古老的成分也较多。通过与有关湖泊水生植物区系的比较,及区系成分的分析,说明花马湖水生植物区系具有从暖温带至中亚热带过渡的特点。     

国产蕺菜的染色体数目变异及核穿壁现象

蕺菜 Houttuynia cordata Thunb.属三白草科蕺菜属,俗称鱼腥草、侧耳根等,主要分 布于 亚洲东部和东南部,我国以长江流域及其以南各省区常见。蕺菜药蔬兼用,为极具开发前景 的植物资源。部分学者曾对蕺菜染色体数目进行过报道,但报道的染色体数目不尽一致（梁汉兴,1991,1995; Hsu,1968; Nilsamranchit et al, 1999）,并且对此 现象未作较为详细而深入的解释。本文对国内部分省区收集的蕺菜种质资源进行染色体数目分析,旨在从细胞学角度探讨蕺菜的遗传多样性,为
