The AcrAB-TolC multidrug efflux pump confers resistance to Escherichia coli against many antibiotics and toxic compounds. The TolC protein is an outer membrane factor that participates in the formation of type I secretion systems. The genome of Vibrio vulnificus encodes two proteins homologous to the E. coli TolC, designated TolCV1 and TolCV2. Here, we show that both TolCV1 and TolCV2 partially complement the E. coli TolC function and physically interact with the membrane fusion protein AcrA, a component of the E. coli AcrAB-TolC efflux pump. Using site-directed mutational analyses and an in vivo cross-linking assay, we demonstrated that the α-barrel tip region of TolC homologs plays a critical role in the formation of functional AcrAB-TolC efflux pumps. Our findings suggest the adapter bridging model as a general assembly mechanism for tripartite drug efflux pumps in Gram-negative bacteria.  相似文献

为了调整低山丘陵区低效林林分结构,探明马尾松(Pinus massoniana Lamb.)与乡土阔叶树种凋落叶混合分解过程中的全碳(C)释放规律。本研究以华南广泛分布的马尾松、檫木(Sassafras tzumu(Hemsl.) Hemsl)、香樟(Cinnamomum camphora(Linn) Presl)以及香椿(Toona sinensis(A. Juss.) Roem.)凋落叶为研究对象,将这4个树种凋落叶按照不同树种搭配以及混合比例组合为35个处理后进行野外分解实验,探讨C释放最佳的凋落叶树种组合以及混合比例。研究发现:4个单一树种凋落叶之间,香椿凋落叶的C释放最快,檫木和香樟凋落叶次之,马尾松凋落叶最慢。31个混合凋落叶中,C释放的非加和效应随着分解时间的延长表现出先升增强后减弱的趋势,且相对于其他季节,凋落叶在秋季的非加和效应有所减弱。一针一阔树种组合中,香樟凋落叶占比≥30%的处理:PC73和PC64的协同效应较强;一针两阔和一针三阔组合中,阔叶占比≥30%且含有香椿凋落叶的处理:PST613和712、PCT631和613、PSCT7111和6121的协同效应较强。  相似文献

Endometriosis is a benign gynecological disease of women of reproductive ages, wherein endometrial cells grow ectopically, decreasing their quality of life due to chronic pelvic pain and severe dysmenorrhea. Although surgery and hormone therapies are gold standards for treating endometriosis, side effects are common and the recurrence rate is nearly 50%. Recent studies are exploring phytochemicals as pharmacological adjuvants for treating endometriosis. Delphinidin is an anthocyanin with anti-inflammatory, antioxidative, and anticancerous properties. In this study, delphinidin showed antiproliferative and apoptotic effects on human endometrial cells. Additionally, treatment with delphinidin decreased the mitochondrial membrane potential and increased cytosolic calcium levels in VK2/E6E7 and End1/E6E7 cells. Delphinidin decreased the phosphorylation of proliferative signaling molecules, including ERK1/2, AKT, P70S6K, and S6, while increasing the phosphorylation of P38 MAPK and P90RSK. These results imply that delphinidin is a novel therapeutic agent for treating and managing endometriosis, and has fewer side effects.  相似文献

Bilayer membranes exhibit an elastic resistance to changes in curvature. This resistance depends both on the intrinsic stiffness of the constituent monolayers and on the curvature-induced expansion or compression of the monolayers relative to each other. The monolayers are constrained by hydrophobic forces to remain in contact, but they are capable of independent lateral redistribution to minimize the relative expansion or compression of each leaflet. Therefore, the magnitude of the expansion and compression of the monolayers relative to each other depends on the integral of the curvature over the entire membrane capsule. The coefficient characterizing the membrane stiffness resulting from relative expansion is the nonlocal bending modulus kr. Both the intrinsic (local) bending modulus (kc) and the nonlocal bending modulus (kr) can be measured by the formation of thin cylindrical membrane strands (tethers) from giant phospholipid vesicles. Previously, we reported measurements of kc based on measurements of tether radius as a function of force (Song and Waugh, 1991, J. Biomech. Engr. 112:233). Further analysis has revealed that the contribution from the nonlocal bending stiffness can be detected by measuring the change in the aspiration pressure required to establish equilibrium with increasing tether length. Using this approach, we obtain a mean value for the nonlocal bending modulus kr of approximately 4.1 x 10(-19)J. The range of values is broad (1.1-10.1 x 10(-19)J) and could reflect contributions other than simple mechanical equilibrium. Inclusion of the nonlocal bending stiffness in the calculation of kc results in a value for that modulus of approximately 1.20 +/- 0.17 x 10(-19)J, in close agreement with values obtained by other methods.  相似文献

Previously, we have demonstrated that intrathecally (i.t.) administered corticotropin-releasing factor (CRF) in mice produces stimulus-specific antinociception and modulation of morphine-induced antinociception by mechanisms involving spinal kappa opioid receptors. Recently, we also have found that CRF releases immunoreactive dynorphin A, a putative endogenous kappa opioid receptor agonist, from superfused mice spinal cords in vitro. Dynorphin A administered intracerebroventricularlly (i.c.v.) to mice has been shown to modulate the expression of morphine tolerance. In the present study, the possible modulatory effects of i.t. administered CRF as well as dynorphin A on morphine tolerance were studied in an acute tolerance model. Subcutaneous administration of 100 mg/kg of morphine sulfate (MS) to mice caused an acute tolerance to morphine-induced antinociception. The antinociceptive ED50 of MS was increased from 4.4 mg/kg (naive mice) to 17.9 mg/kg (4 hours after the injection of 100 mg/kg MS). To study the modulatory effects of spinally administered CRF and dynorphin A on the expression of morphine tolerance, CRF and dynorphin A were injected i.t. at 15 min and 5 min, respectively, before testing the tolerant mice by the tail-flick assay. The antinociceptive ED50 of MS in tolerant mice was decreased to 8.8 mg/kg and 7.1 mg/kg, respectively, after i.t. administration of CRF (0.1 nmol) and dynorphin A (0.2 nmol). In contrast, 0.5 nmol of alpha-helical CRF (9-41), a CRF antagonist and 0.4 nmol of norbinaltorphimine, a highly selective kappa opioid receptor antagonist, when administered i.t. at 15 min before the tail-flick test in tolerant mice, increased the antinociceptive ED50 of MS to 56.6 mg/kg and 88.8 mg/kg, respectively. These data confirmed the modulatory effect of dynorphin A on morphine tolerance and suggested that CRF, which releases dynorphin A in several central nervous system regions, also plays a modulatory role in the expression of morphine tolerance.  相似文献

Insects have invaded marine habitats only rarely and secondarily. Recently, we discovered a flightless dipteran species skating rapidly on the surface of seawater ponds at the Pacific coast of eastern China. Morphological analyses initially suggested an isolated position of the non‐biting midge, suggesting the erection of a new genus within Chironomini (Diptera: Chironomidae). However, an analysis of molecular data revealed that the marine species is in fact nested within the species‐rich genus Dicrotendipes. The apparent conflict between molecular and morphological data can be easily explained. It is likely that the new species has evolved a series of autapomorphic adaptations. These traits clearly distinguish the taxon from other species of the genus but do not justify the erection of a new supraspecific taxon, which would render Dicrotendipes paraphyletic. The switch to marine environments was likely a trigger for various morphological modifications resulting from increased selective pressure. Molecular data suggest that the potential speciation event occurred around 19–29 Ma, linked with a migration from freshwater to seawater ponds along the Pacific Ocean. Considering the results of our analysis, we place the flightless marine skater in the genus Dicrotendipes. All life stages of Dicrotendipes sinicus Qi & Lin sp. n. are described and illustrated, associated with larvae obtained by rearing or confirmed through association with DNA barcodes. The biology and ecology of the species are outlined based on collection data and in situ observations. Evolutionary patterns linked with repeated invasions of marine habitats are discussed.  相似文献

Zeolitic imidazolate framework‐8 (ZIF‐8) loading rhodamine‐B (ZIF‐8@rhodamine‐B) nanocomposites was proposed and used as ratiometric fluorescent sensor to detect copper(II) ion (Cu2+). Scanning electron microscopy, Fourier transform infrared spectroscopy, X‐ray powder diffraction, nitrogen adsorption/desorption isotherms and fluorescence emission spectroscopy were employed to characterize the ZIF‐8@rhodamine‐B nanocomposites. The results showed the rhodamine‐B was successfully assembled on ZIF‐8 based on the π‐π interaction and the hydrogen bond between the nitrogen atom of ZIF‐8 and –COOH of rhodamine‐B. The as‐obtained ZIF‐8@rhodamine‐B nanocomposites were octahedron with size about 150–200 nm, had good water dispersion, and exhibited the characteristic fluorescence emission of ZIF‐8 at 335 nm and rhodamine‐B at 575 nm. The Cu2+ could quench fluorescence of ZIF‐8 rather than rhodamine‐B. The ZIF‐8 not only acted as the template to assemble rhodamine‐B, but also was employed as the signal fluorescence together with the fluorescence of rhodamine‐B as the reference to construct a novel ratiometric fluorescent sensor to detect Cu2+. The resulted ZIF‐8@rhodamine‐B nanocomposite fluorescence probe showed good linear range (68.4 nM to 125 μM) with a low detection limit (22.8 nM) for Cu2+ combined with good sensitivity and selectivity. The work also provides a better way to design ratiometric fluorescent sensors from ZIF‐8 and other fluorescent molecules.  相似文献

The room temperature (RT) processability of the photoactive layers in polymer solar cells (PSCs) from halogen‐free solvent along with their highly reproducible power conversion efficiencies (PCEs) and intrinsic thickness tolerance are extremely desirable for the large‐area roll‐to‐roll (R2R) production. However, most of the photoactive materials in PSCs require elevated processing temperatures due to their strong aggregation, which are unfavorable for the industrial R2R manufacturing of PSCs. These limiting factors for the commercialization of PSCs are alleviated by synthesizing random terpolymers with components of (2‐decyltetradecyl)thiophen‐2‐yl)naphtho[1,2‐c:5,6‐c′]bis[1,2,5]thiadiazole and bithiophene substituted with methyl thiophene‐3‐carboxylate (MTC). In contrast to the temperature‐dependent PNTz4T polymer, the resulting random terpolymers (PNTz4T‐MTC) show better solubility, slightly reduced crystallinity and aggregation, and weaker intermolecular interaction, thus enabling PNTz4T‐MTC to be processed at RT from a halogen‐free solvent. Particularly, the PNTz4T‐5MTC‐based photoactive layer exhibits an excellent PCE of 9.66%, which is among the highest reported PCEs for RT and ecofriendly halogen‐free solvent processed fullerene‐based PSCs, and a thickness tolerance with a PCE exceeding 8% from 100 to 520 nm. Finally, large‐area modules fabricated with the PNTz4T and PNTz4T‐5MTC polymer have shown 4.29% and 6.61% PCE respectively, with an area as high as 54.45 cm2 in air.  相似文献

Novel photovoltaic perovskite solar cells (PSCs) with high‐efficient photovoltaic property are largely in thrall to the uncertain perovskite grain size and inevitable defects. Here, inspired by the competitive growth between tree and grass in the forest system, a competitive perovskite grain growth approach via micro‐contact print (MicroCP) method (CD disk as templates) for printing wettability‐patterned substrate is proposed, aiming to achieve large‐grained perovskite and avoid discontinuous perovskite films caused by the low wettability of substrates. A MicroCP process is employed to construct a patterned wettability surface for the perovskite competitive growth mechanism on the electrode surface. This approach modifies the substrates quickly, ensures the uniform coverage of perovskite due to the function of ‐NH2 and Pb2+ bonds, and converts the perovskite films composed of small grains and pinholes into high‐quality perovskite films, free from pinholes and made up of large grains, resulting in efficiencies over 20% for the MicroCP PSCs.  相似文献
