Objectives

To explore potential effects of recombinant human fibroblast growth factor 20 (rhFGF20) in the growth of cultured mouse vibrissal follicles.

Results

The growth of cultured mouse vibrissal follicles was significantly induced by rhFGF20 in a dose dependent pattern in the in vitro vibrissal follicle organ culture model. However, too high concentration of rhFGF20 could inhibit the growth of vibrissal follicles. We further demonstrated that rhFGF20 stimulated the proliferation of hair matrix cells and activated Wnt/β-catenin signaling pathway.

Conclusions

The rhFGF20 might be a potential therapeutic agent to treat hair loss disorders.

To elucidate the effect of selenium (Se) on antioxidant function of mammary glands in dairy cows and the underlying mechanism, an experiment was conducted using a single-factor completely randomized design study. Bovine mammary epithelial cells (BMECs) were randomly divided into four groups: control, Se treatment, 2,4-dinitrochlorobenzene (DNCB) inhibition, and Se prevention. Treatment of BMECs with Se was found to significantly reverse decreased cell proliferation and the expression of thioredoxin reductase (TrxR) after DNCB exposure. DNCB-induced activation of apoptosis signaling kinase-1 (ASK-1), which activates the mitogen-activated protein kinase (MAPK) pathway, was reduced in BMECs treated with Se. Additionally, our results indicated that Se treatment resulted in lower intracellular accumulation of arachidonic acid (ARA) and 15-hydroperoxyeicosatetraenoic acid (15-HPETE) due to suppressed expression of cytosolic phospholipase A2 (cPLA2) regulated by p38MAPK and c-Jun N-terminal kinase (JNK) in DNCB-stimulated BMECs. Taken together, these findings suggest that Se treatment improved the antioxidant function of dairy cow mammary glands and protected cells from oxidative damage primarily by increasing the activity of TrxR, inhibiting the activation of the MAPK signaling pathway, and thus decreasing the content of ARA and its related metabolites.  相似文献

Several low-molecular-weight organic carbon (LMWOC) compounds (acetate, propionate, butyrate, lactate, and glucose) were added to flooded arsenic-rich tailing mine soil to investigate their effect to the mobilization of As/Fe and potential shift of microbial community. A promoting effect to the mobilization and biotransformation of As(V)/Fe(III) in the soils resulting from the supplementation with LMWOCs substrate was indicated compared to the biotic microcosm amended with deionized water alone. During 38-day biotic incubation, more than 2100 μg/L of As(III) and 4.2 mg/L of Fe(II) levels were released from the soils amended with LMWOCs substrates, compared to the levels of As(III) and Fe(II) (less 35 μg/L and 1.82 mg/L) derived from the biotic supplementation with deionized water alone. PCR-DGGE indicated that several LMWOCs-responded bacteria were mostly related to Firmicutes and Proteobacteria. Moreover, a negligible impact on the abundance of Fe(III)-reducing family Geobacteraceae was indicated in the LMWOCs-amended soils. However, an increased abundance of sulfate-reducing bacteria but a decreased abundance of arsenate-respiring bacteria were indicated upon the soils supplemented with acetate alone, compared with other LMWOC amendments. DNA-stable isotope probing analysis demonstrated that the dual roles of acetate was not only served as an electron donor for biotransformation of As(V)/Fe(III) in soil, but also assimilated as a powerful energy source to promote the growth of sulfate-reducing bacteria. The findings suggest that there are specific bacteria that preferentially respond to the additions of LMWOC for controlling the biochemical cycle process of As/Fe in soils.  相似文献

The three-dimensional structure of stromal cell-derived factor-1 (SDF-1) was determined by NMR spectroscopy. SDF-1 is a monomer with a disordered N-terminal region (residues 1-8), and differs from other chemokines in the packing of the hydrophobic core and surface charge distribution. Results with analogs showed that the N-terminal eight residues formed an important receptor binding site; however, only Lys-1 and Pro-2 were directly involved in receptor activation. Modification to Lys-1 and/or Pro-2 resulted in loss of activity, but generated potent SDF-1 antagonists. Residues 12-17 of the loop region, which we term the RFFESH motif, unlike the N-terminal region, were well defined in the SDF-1 structure. The RFFESH formed a receptor binding site, which we propose to be an important initial docking site of SDF-1 with its receptor. The ability of the SDF-1 analogs to block HIV-1 entry via CXCR4, which is a HIV-1 coreceptor for the virus in addition to being the receptor for SDF-1, correlated with their affinity for CXCR4. Activation of the receptor is not required for HIV-1 inhibition.  相似文献

1992-1995年间,我们对苍山洱海自然保护区的小型兽类进行了生态区系的调查,共获标本35种1600余只,参考以往的有关资料,已知当地小兽5目12科30属53种。通过8种主要生态环境的调查结果看出,小兽的物种多样性指数最高为针阔混交林,最低为农耕区,但密度最高为农耕区,最低为高山灌丛草甸。  相似文献

Diazocyclopentadiene (DACP), an inhibitor of ethylene action, was used to investigate the role of ethylene receptor(s) in regulating ethylene biosynthesis and respiration in strawberry fruit. DACP stimulated ethylene production of fruit at all stages of maturity. This stimulation was mainly due to an increase in ACC content in the tissue without significantly changing ACC oxidase activity. DACP did not induce any change in respiration in fruit at various stages of maturity except the early green stage (green I). We also investigated the possible interaction of DACP and IAA in ethylene production. Results indicated that all treatments increased ethylene production compared to the control although the absolute ethylene production differed in the order IAA plus DACP > only DACP > only IAA > control. IAA stimulated ethylene production without change of ACC oxidase activity at 1 day after treatment in strawberry fruit at pink stage. Results suggest that ethylene biosynthesis in nonclimacteric strawberry fruit at various stages of maturity may be regulated by ethylene receptor(s) with inhibition of ethylene production. DACP may release this inhibitory effect, and resulting in increasing ethylene production. Results also indicated that respiration may not be regulated by an ethylene receptor in strawberry fruit at most stages of maturity. DACP and IAA showed interaction in regulation of ethylene production which was caused by an increase in ACC content, not ACC oxidase activity.  相似文献
