大豆油中转基因成分的Nested PCR和Semi-nested PCR检测方法研究 总被引:3,自引:0,他引:3
对大豆油中DNA提取方法进行了研究,结果表明CTAB、SDS和Wizard试剂盒提取大豆油DNA均具有良好的效果。利用nested PCR和semi—nested PCR检测大豆(Roundup Ready)油中的转基因成分发现,该方法能够检测到大豆原油中的Lectin基因(112bp)、CaMV35S基因(147bP)和CP4-EPSPS基因(205bp),检测灵敏度达到10^-6ng/μl;但该方法未能从人豆成品油(一级)中扩增到上述基因,当中的转基因成分DNA含量低于10^-12ng/μl。 相似文献

杂交育种依然是我国油菜育种的主要方法,杂种优势的利用仍然是提高产量的重要途径.为了解我国甘蓝型油菜的遗传变异,采用16个EST-SSR标记对近年来推广的91个品种的遗传多样性进行了分析.共扩增到100个条带,其中84个多态性带,多态性比率为84%.平均每对引物扩增的带数和多态性带数分别为6.25个和5.25个.多态性信息含量(PIC)变化在0.022-0.926之间,平均为0.677,所揭示的基因型数变化于2-24之间,平均为12.44个.供试材料间遗传距离变幅较大(0.0530-0.7223之间),说明它们具有广泛的遗传变异.其中,杂交种和2000年以后育成品种的遗传基础较宽,遗传多样性分别明显高于常规品种和2000年以前育成的品种.按非加权成对平均数法(UPGMA)进行的聚类分析显示,在遗传距离为0.313处,参试材料可以分为三大类,其中,包含87份材料的第一大类在遗传距离为0.233处又可进一步分为10个亚类.聚类结果与系谱来源基本一致,比较真实反映了所用材料的遗传变异情况. 相似文献

在滤膜、液体培养基和土壤微宇宙 3种系统中 ,研究了接合型质粒 pLV1 0 1 6由快生型大豆根瘤菌 (Rhizobiumfredii)QB1 1 31向R .frediilux3的水平转移及pLV1 0 1 6由QB1 1 31向土著细菌的转移 .接合培养 1d后 ,分别计算供、受体菌的生长速率和质粒转移速率常数 (γ) .结果表明 ,相同接种浓度下 ,滤膜接合时γ值最高 ,土壤中γ值最低 ,γ值不受土壤是否灭菌和是否有大豆植株的影响 ,γ值与初始接种浓度负相关 ,与供、受体的生长速率正相关 .在未灭菌土中检测到 pLV1 0 1 6可转移到土著细菌 ,土著接合子分别属于根瘤菌属和假单胞菌属 . 相似文献

The feeding habitat selection of blue sheep (Pseudois nayaur) was studied by direct observation method in the Helan Mountains, China during winter (from November to December) and spring
(from April to June) from 2003 to 2004. We established 25 line transects to collect information on feeding habitats used by
blue sheep. Blue sheep in the study area preferred mountain savanna forests, a habitat dominated by Ulmus glaucescens, with medium tree density (<4 individuals / 400 m2), moderate tree height (4–6 m), higher shrub density (> 5 individuals / 100 m2), higher shrub (> 1.3 m), higher food abundance (> 50 g), moderate distance to human disturbance (< 500 m), and mild distance
to bare rock (< 2 m). Such habitats characterized by 12 ecological factors were preferred as feeding areas by blue sheep during
winter. Similar to habitat selection by the species during winter, blue sheep also showed a preference for mountain savanna
with tree dominated by Ulmus glaucescens and medium tree density (< 4 individuals / 400 m2) during spring. Nevertheless, blue sheep preferred medium tree height (< 6 m), moderate tree density (5–10 individuals /
100 m2), medium shrub height (1.3–1.7 m), higher food abundance (> 100 g), moderate altitude (< 2 000 m), moderate distance to water
resource (< 500 m), and medium hiding cover (50%–75%) during spring. Selection of the feeding habitats by sheep showed a significant
difference in vegetation type, landform feature, dominant tree, tree height, shrub density, distance to the nearest shrub,
food abundance, slope direction, slope degree, distance to water resource, and hiding cover between winter and spring. Results
of principal components analysis indicated that the first principal component accounted for 24.493% of the total variance
among feeding habitat variance during winter, with higher loadings for vegetation type, dominant tree, tree height, distance
to the nearest tree, shrub density, shrub height, altitude, distance to water resource, and distance to human disturbance.
In spring, the first principal components explained 28.777% of the variance, with higher loadings for vegetation type, distance
to the nearest tree, shrub height, distance to the nearest shrub, food abundance, altitude, and distance to human disturbance.

Mesenchymal stem cells (MSCs) possess an immunoregulatory capacity and are a therapeutic target for many inflammation‐related diseases. However, the detailed mechanisms of MSC‐mediated immunosuppression remain unclear. In this study, we provide new information to partly explain the molecular mechanisms of immunoregulation by MSCs. Specifically, we found that A20 expression was induced in MSCs by inflammatory cytokines. Knockdown of A20 in MSCs resulted in increased proliferation and reduced adipogenesis, and partly reversed the suppressive effect of MSCs on T cell proliferation in vitro and inhibited tumour growth in vivo. Mechanistic studies indicated that knockdown of A20 in MSCs inhibited activation of the p38 mitogen‐activated protein kinase (MAPK) pathway, which potently promoted the production of tumour necrosis factor (TNF)‐α and inhibited the production of interleukin (IL)‐10. Collectively, these data reveal a crucial role of A20 in regulating the immunomodulatory activities of MSCs by controlling the expression of TNF‐α and IL‐10 in an inflammatory environment. These findings provide novel insights into the pathogenesis of various inflammatory‐associated diseases, and are a new reference for the future development of treatments for such afflictions. 相似文献

Given the potential clinical benefit of inhibiting Na+/Ca2+ exchanger (NCX) activity during myocardial ischemia reperfusion (I/R), pharmacological approaches have been pursued to both inhibit and clarify the importance of this exchanger. SEA0400 was reported to have a potent NCX selectivity. Thus, we examined the effect of SEA0400 on NCX currents and I/R induced intracellular Ca2+ overload in mouse ventricular myocytes using patch clamp techniques and fluorescence measurements. Ischemia significantly inhibited inward and outward NCX current (from -0.04+/-0.01 nA to 0 nA at -100 mV; from 0.23+/-0.08 nA to 0.11+/-0.03 nA at +50 mV, n=7), Subsequent reperfusion not only restored the current rapidly but enhanced the current amplitude obviously, especially the outward currents (from 0.23+/-0.08 nA to 0.49+/-0.12 nA at +50 mV, n=7). [Ca2+]i, expressed as the ratio of Fura-2 fluorescence intensity, increased to 138+/-7% (P<0.01) during ischemia and to 210+/-11% (P<0.01) after reperfusion. The change of NCX current and the increase of [Ca2+]i during I/R can be blocked by SEA0400 in a dose-dependent manner with an EC50 value of 31 nM and 28 nM for the inward and outward NCX current, respectively. The results suggested that SEA0400 is a potent NCX inhibitor, which can protect mouse cardiac myocytes from Ca2+ overload during I/R injuries. 相似文献

为了研究人肺腺癌细胞A 5 49和正常细胞HBE的蛋白质组差异 ,用固相pH梯度双向凝胶电泳分离人肺腺癌细胞系A 5 49和正常细胞HBE的总蛋白质 ,银染显色 ,PDQuest 2 DE软件分析 ,对部分差异蛋白质点进行基质辅助激光解吸电离飞行时间质谱 (MALDI TOF MS)测定其胶内酶解后的肽质指纹图谱 ,用PeptIdent软件查询SWISS PROT数据库。结果获得了分辨率和重复性均较好的双向电泳银染图谱 ,图象分析探测到A 5 492 DE图谱的平均蛋白质点数为 (890± 38)个 ,HBE的平均蛋白质点数为 (75 7± 2 7)个 ,不同胶间蛋白质点的位置偏差在IEF方向为 (2 .85± 0 .48)mm ,在SDS PAGE方向为 (2 .6 9± 0 .37)mm。差异表达分析发现A 5 49和HBE图谱有5 35个蛋白质点相互匹配 ,其中A 5 49有 35 5个未被匹配 ,HBE中有 2 2 2个未被匹配 ;对A 5 49和HBE中的 18个差异蛋白质点分别进行肽质指纹分析 ,经数据库查询 ,初步鉴定为一些与物质代谢、细胞因子、信号转导有关的蛋白质。提示人肺腺癌细胞A 5 49和正常细胞HBE的蛋白质组具有差异 ,这种蛋白质组的差异分析有助于进一步研究肺腺癌的相关蛋白质及分子标记物 相似文献
