国际景观生态学研究新进展   总被引:26,自引:13,他引:26  
20世纪90年代中期以来,国际景观生态学发展迅速.景观生态学研究最为活跃的地区集中在北美、欧洲、大洋洲(澳大利亚)、东亚(中国),表明景观生态学理论、方法和应用的广泛性和越来越高的认知度.从研究内容上看,景观生态评价、规划和模拟一直占居主导地位,其次是景观生态保护与生态恢复、景观生态学的理论探讨.随着景观生态学研究的深入,以科学和实践问题为导向的学科交叉与融合不断加强,促进了景观生态学新的学科生长点的形成和发展,主要包括水域景观生态学、景观遗传学、多功能景观研究、景观综合模拟、景观生态与可持续性科学.在全球化背景下,中国的景观生态学研究也已经取得了长足进展,国际同行开始关注并在重要学术刊物上评介中国的景观生态学研究,标志着中国景观生态学已逐步走上国际舞台.然而,中国景观生态学进一步发展的挑战和机遇并存.中国科学家需要抓住人类对可持续发展和可持续性科学的共同夙求,放眼国际前沿、服务本土需求,从中国自身的特色出发,关注受人类干扰的和以人为主导的景观,以景观格局与生态过程的多尺度、多维度耦合研究为核心,区域综合与区内分异并重,推动综合整体性景观生态学的建立和完善.  相似文献

保护区是一种特殊的土地利用类型,以保护物种,生态系统或自然遗产为主要目的,但是保护的成败依赖于保护区管理的有效性和可持续性,这是保护区健康发展的根本保障,功能评价就是促进保护区持续有效管理的重要手段,本文以模糊逻辑和问卷调查为基础,对卧龙自然保护区功能进行了综合评价。评价中,将卧龙自然保护区的整体功能划分为五大方面,即自然保护、宣传教育,科学研究,社会发展和经济发展,其中每项功能再进一步划分为3到5个次级指标,从而形成了一个具有3个层次的评价指标体系,评价结果表明,自然保护功能的评价为“好”,其余4项功能均评价为“一般”,因此,作为5方面功能综合的整体功能状况也为“一般”。卧龙自然保护区五大功能的相对良好次序为:自然保护,科学研究,社会发展,宣传教育和经济发展,这就意味着,卧龙自然保护区的功能具有综合整体性,任何方面的功能失调都会影响到保护区整体功能的发挥和保护区自身的健康发展,改善保护区的整体功能,也必须注重各项功能的相互影响,和彼此协调,只有这样才能解决好影响保护区发展的各种矛盾。克服管理和决策中的各种片面性和低效性,促使保护区的管理不断完善,从而保证保护区的可持续发展,进一步分析卧龙自然保护区各项功能的次级指标,对从各方面寻求改善保护区的功能状况很有帮助。  相似文献

骨骼肌线粒体解耦联蛋白3(uncoupling protein3,UCP3)在低氧时的生理作用尚不清楚。本研究观察了大鼠在耐力训练前后,模拟急性高原低氧各时间点的骨骼肌线粒体UCP3 mRNA和蛋白表达、线粒体呼吸功能、活性氧(reactive oxygen species,ROS)产生速率以及锰超氧化物歧化酶(manganese superoxide dismutase,MnSOD)表达和活性的变化。急性低氧导致线粒体一系列生物能学功能障碍。未训练大鼠UCP3蛋白在4h时比静息时升高了60%,而MnSOD蛋白含量及活性在低氧暴露过程中无显著变化;UCP3蛋白上调通过降低电子传递链耦联程度抑制O2-产生,但同时降低了ATP合成效率。耐力训练显著抑制急性低氧诱导的骨骼肌UCP3蛋白上调(67%;S42%)。训练组大鼠的ROS产生速率在低氧2h、4h和6h时显著低于未训练组;MnSOD蛋白含量及活性分别较术训练组提高了50%和34%。训练组人鼠MnSOD上调可增加线粒体对ROS的耐受力,进而抑制UCP3蛋白表达,从而提高氧化磷酸化效率。急性低氧中,未训练组大鼠呼吸控制比(respiratory control ratio,RCR)和磷氧比(ADP to oxygen consumption ratio,P/O)显著降低,而训练组RCR和P/O保持相对稳定。以上结果提示:(1)模拟急性高原低氧可诱导UCP3 mRNA及蛋白表达升高,从而降低升高了的线粒体膜电位(△ψ),使ROS的产生减少;(2)耐力训练可抑制低氧诱导的UCP3表达上调,提高ROS酶学清除能力,从而提高线粒体氧化磷酸化效率。  相似文献

Vegetation and land-use mosaics of shrub land, grassland, farmland and reforested land are characteristic for residential areas of the Wolong nature reserve in Sichuan province, southwest China. The patterns represent a stage in long-term human disturbance and natural secondary succession since deforestation. The aim of this paper was to elucidate the soil-land cover types, soil-slope position relationships on a typical disturbed slope in the humid mountainous region. Soils were taken from four typical land cover types (shrub land, grassland, farmland and reforested land) under four slope positions (upper slope, middle slope, lower slope, foot slope) to study the distribution of soil nutrients and moisture on the transect scales of the slope. To elucidate the influence of land-use and slope position on soil quality, two integrated quantitative indexes were developed to compare soil quality under different land-use types (QI) and under different slope positions (PI). The results showed shrub land had the highest QI value, while the QI of reforested land and farmland had lower values. Foot slopes and upper slopes had higher soil quality levels compared to middle slopes and lower slopes. The results indicated that on the slope scale shrub land had high soil quality levels. The results also suggested that in the area of China where the climate favors secondary succession, leave nature as it is is a better choice than the policy change farmland to forest land.  相似文献

The cytotoxic and differentiating effects of 10-hydroxycamptothecin (HCPT) in the human promyelocytic leukemia cell line HL-60 were examined. By trypan blue dye exclusion, a 24-h exposure of the cells to 0.1 microM of the drug was found to be cytotoxic. Exposure of the cells to lower concentrations (0.001-0.01 microM) for 3 days reduced cell proliferation and induced cell differentiation. As determined by Wright-Giemsa staining, approximately 25% of promyelocytic cells became metamyelocytes, banded and segmented neutrophils. Electron microscopy demonstrated alterations in the ultrastructure of HCPT-induced HL-60 cells that included the formation of lobulated nuclei and the accumulation of large vesicles and small myelin bodies as well as glycogen-like particles in the cell periphery. Qualitatively similar results were obtained in a subline of HL-60 that is resistant to 4'-(9-acridinylamino)methanesulfon-m-anisidide (m-AMSA); however, the rate and extent of induced nitroblue tetrazolium-positive cells by HCPT and several other agents were greater in the resistant cell line. Under conditions that induced cell differentiation, HCPT sharply inhibited [3H]thymidine incorporation into DNA and increased the rate of protein synthesis without an effect on the rate of RNA synthesis. The measurement of DNA topoisomerase I activity in nuclear extracts from both HCPT- and DMSO-treated cells demonstrated that the enzyme was decreased in mature cells compared to nondifferentiated controls. The data suggest that progressive reduction of DNA topoisomerase I activity may be associated with cell differentiation, but whether HCPT-induced differentiation is mediated by inhibition of the enzyme is inconclusive.  相似文献

The rpoS gene from Pseudomonas sp. M18, which encodes predicted protein (an alternative sigma factor s, σS, or σ38) with 99.5% sequence identity with RpoS from Pseudomonas aeruginosa PAO1, was first cloned. In order to investigate the mechanism of rpoS expression, an rpoS null mutant, named M18S, was constructed with insertion of aacC1 cassette bearing a gentamycin resistance gene. With introduction of a plasmid containing an rpoS′–′lacZ translational fusion (pMERS) to wild-type strain M18 or M18S, it was first found that β-galactosidase activity expressed in strain M18S (pMERS) decreased to fourfold of that expressed in the strain M18 (pMERS). When strain M18S (pMERS) was introduced with another plasmid pBBS containing the wild-type rpoS gene, its β-galactosidase expression level was enhanced and almost restored to that in strain M18 (pMERS). Similarly, expression of β-galactosidase from a chromosomal fusion of the promoter of the wild-type rpoS gene with lacZ (rpoSlacZ) was enhanced fivefold in the presence of a plasmid with the wild-type rpoS gene. With these findings, it is suggested that RpoS sigma factor may be involved in autoinducing its own gene expression in Pseudomonas sp. M18.  相似文献

本研究旨在观察胍丁胺 (agmatine ,Agm)对分离大鼠心室肌细胞内游离钙浓度 ( [Ca2 +]i)的影响。用酶解方法分离大鼠心室肌细胞 ,用Fluo 3 AM负载 ,然后用激光共聚焦法测定单个心室肌细胞 [Ca2 +]i 的荧光强度 (fluorescenceintensity ,FI) ,结果以FI或相对荧光强度 (F/F0 % )表示。实验结果表明 ,在正常台氏液 (含钙 1 0mmol/L)和无钙台氏液中 ,单个大鼠心室肌细胞的荧光密度分别为 12 8 8± 13 8和 119 6± 13 6,两者无差异。Agm 0 1、1和 10mmol/L浓度依赖性地显著降低细胞的钙浓度 ;在正常台氏液中加入EGTA 3mmol/L ,Agm同样降低细胞的钙浓度。KCl 60mmol/L ,PE 3 0 μmol/L ,和Bay K 864 410 μmol/L均升高心室肌细胞的[Ca2 +]i。Agm同样降低高浓度KCl、Bay K 864 4和PE诱发的心室肌细胞 [Ca2 +]i 升高。当细胞外液钙浓度由 1mmol/L增加到 10mmol/L时 ,诱发心室肌细胞钙超载 ,同时部分心室肌细胞产生可传播的钙波 (Ca2 +wave) ,Agm 1mmol/L降低钙波的传播速度和持续时间 ,最终阻断钙波。以上结果提示 ,Agm对心室肌细胞的胞浆[Ca2 +]i具有抑制作用 ,此作用通过阻断电压依赖性钙通道而实现 ;并可能与抑制大鼠心室肌细胞内钙释放有关  相似文献

Many studies have been conducted on ecosystem services and double counting is a frequent problem that causes uncertainty and poor reliability of estimating the value of ecosystem services. By referring to previous studies of ecosystem service valuation, our research has identified the basic causes of double counting. These include ambiguous definitions and inconsistent classifications of ecosystem services, poor understanding of ecosystem complexity, inadequate recognition of exclusiveness and complementarities of individual ecosystem services, spatio-temporal scale dependence of ecosystem services, and overlap and lack of cross-referencing between ecosystem service valuation methods. Measures for reducing double counting in ecosystem service valuation are proposed as follow: (1) identifying the spatio-temporal scales of ecosystem services; (2) valuing the final benefits obtained from ecosystem services; (3) establishing consistent classification systems for ecosystem services; and (4) selecting valuation methods appropriate for the study context.  相似文献

Rate-dependent repolarization (RDR) of action potential (AP) in cardiomyocyte plays a critical role in the genesis of arrhythmias and RDR in atrium has been linked with atrial fibrillation. However, detailed studies focusing on the role of RDR in rabbit atrium are scant. In this study, atrial cells were isolated from rabbit heart and rate-dependent property was explored in single atrial cell to elucidate the underlying mechanism. Our results indicated that rate-dependent prolongation was evident at the action potential duration at 20% (APD20) and 50% (APD50) repolarization but not at 90% repolarization (APD90) under control condition. Using transient outward potassium current (Ito) inhibitor 4-Aminopyridine (4-AP, 2 mM) effectively eliminated the changes in APD20 and APD50, and unmasked the rate-dependent reduction of APD90 which could be diminished by further adding L-type calcium current (ICaL) inhibitor nifedipine (30 μM). However, using the selective late sodium current (INaL) inhibitor GS-458967 (GS967, 1 μM) caused minimal effect on APD90 of atrial cells both in the absence and presence of 4-AP. In consistence with results from APs, Ito and ICaL displayed significant rate-dependent reduction because of their slow reactivation kinetics. In addition, the magnitude of INaL in rabbit atrium was so small that its rate-dependent changes were negligible. In conclusion, our study demonstrated that Ito and ICaL mediate RDR of AP in rabbit atrium, while minimal effect of INaL was seen.  相似文献
